Comparison of the exponential and stationary stages of Staphylococcus aureus (MRSA)

ABSTRACT: Proteomic analysis of Staphylococcus aureus strain NCTC8325 (MRSA) grown in rich medium. This strain produces 97% of persister in stationary phase. Exponential and stationary phase MRSA were compared to elucidate pathways that are modulated in the persister state compared to dividing cells.

Panton-Valentine Leukocidin Does Not Impact Virulence of Community-associated MRSA

Project description:Panton-valentine leukocidin (PVL) has been linked to worldwide emergence of community-associated methicillin resistant Staphylococcus aureus (CA-MRSA) -- its role in virulence in unclear. Here we show that PVL had no effect on global gene expression of prominent CA-MRSA strains nor did it affect bacterial clearance from lungs, spleen and kidneys in a highly discriminatory rabbit bacteremia model. These findings negate a large body of epidemiological research that implicated PVL in CA-MRSA virulence. Succinylome-controlled antibiotic resistance in MRSA - Succinylome

Project description:A sucC and a sucD mutant from the Nebraska Transposon Mutant Library (NTML, Strain NE569 and NE1770) were found to be susceptible to cefoxitin and oxacillin when other mutations in TCA cycle genes did not affect their resistance to these antibiotics. The SucC strain (Strain NE569) accumulated Succinyl CoA, which is known to result in higher succinylation rate. We compared the proteome of the sucC mutant in exponential and stationary stages to the one of the WT strain JE2. The global proteome of the following strains were analyzed by TMT-labelled proteomics: Early exponential JE2 (3Hrs), Exponential JE2 (4Hrs), Exponential SucC (4Hrs), Stationary JE2 (10Hrs), Stationary SucC (12Hrs). For each condition, the bacterial pellet was collected and washed twice with PBS. The proteins were extracted by bead beating, 1000 ug of proteins were reduced with DTT, alkylated with IAA and digested with 1:50 (trypsin:protein, w:w) for 3 Hr at 37 C. 100ug of peptides were labelled with TMT11Plex reagents. The samples were pooled and enriched for Succinyl-lysine containing peptides using the PTMScan Succinyl-Lysine Motif Enrichment Kit following the supplier's instructions. The retained fraction was cleaned-up by C18 solid phase extraction. The peptides were concentrated in 15 ul and 5 ul were analyzed by reverse-phase liquid chromatography-tandem mass spectrometry (LC-MS/MS) using a Waters nanoEquity UPLC system coupled with a QExactive HF-X mass spectrometer.

Proteome Differences between Daptomycin-Resistant and WT MRSA Biofilms after Elasnin Treatment

Project description:A recent study reported that daptomycin-resistant MRSA (DAPR) strain biofilm is more resistant to daptomycin and vancomycin, as compared to the WT strain biofilm. This pose a great danger since DAPR MRSA is prevalent in clinics and they often form biofilms in medical devices. In this study, we investigate the anti-biofilm activity of elasnin against DAPR MRSA biofilms, and we observed that elasnin is not only effective to eradicate the biofilm of the DAPR strain, but it shows a superior activity compared to the susceptible WT strain. Using proteomics, we compared the proteome profile of the DAPR and the WT strain biofilm cells under elasnin treatment to reveal why elasin is superior against the DAPR strain. Besides, we also employed adaptive laboratory evolution (ALE) experiment by repetitively treating MRSA culture with high dose of elasnin, and generated evolved MRSA strain with increased elasnin tolerance. Using quantitative proteomics, we compared the proteome differences in the elasnin-susceptible WT MRSA and elasnin-tolerant evolved strain.

Defining the impact of exoribonucleases in the shift between exponential and stationary phases

Project description:The transition between exponential and stationary phase is a natural phenomenon for all bacteria and requires a massive readjustment of the bacterial transcriptome. Exoribonucleases are key enzymes in the transition between the two growth phases. PNPase, RNase R and RNase II are the major degradative exoribonucleases in Escherichia coli. We analysed the whole transcriptome of exponential and stationary phases from the WT and mutants lacking these exoribonucleases (Δpnp, Δrnr, Δrnb, and ΔrnbΔrnr). When comparing the cells from exponential phase with the cells from stationary phase more than 1000 transcripts were differentially expressed, but only 491 core transcripts were common to all strains. There were some differences in the number and transcripts affected depending on the strain, suggesting that exoribonucleases influence the transition between these two growth phases differently. Interestingly, we found that the double mutant RNase II/RNase R is similar to the RNase R single mutant in exponential phase while in stationary phase it seems to be closer to the RNase II single mutant. This is the first global transcriptomic work comparing the roles of exoribonucleases in the transition between exponential and stationary phase.
