Project description:Abstract S. aureus is a major opportunistic pathogen infecting patients with diabetes. Increased mortality was observed following IV S. aureus infection in diabetic mice compared to non-diabetic controls, correlating with increased numbers of low density neutrophils (LDNs) and neutrophil extracellular traps (NETs). LDN development is dependent on TGFβ, which was more activated in the diabetic host. Neutralization of TGFβ, or the integrin responsible for its activation α5β8, reduced numbers of LDNs and improved survival. Comparison of high and low density neutrophils identified PTEN signaling as a central regulator of LDN NET release. Inhibition of PTEN improved survival and decreased NET production in infected diabetic mice. MEDI4893*, a monoclonal antibody that neutralizes alpha toxin (AT) in development for prevention of S. aureus infection, was able to block TGFβ activation, reduce LDNs and NETs, and significantly improve survival. Our data identify a population of neutrophils in infected diabetic mice which correlated with decreased survival and increased NET production. Targeting a single virulence factor of S. aureus prevented emergence of the LDN population and improved survival, supporting potential use of pathogen specific antibodies for treating diabetic infections.

Project description:The recruitment and/or retention kinetics of neutrophils in diabetic (db) cutaneous wounds is aberrant, and inflammation is prolonged. Intrinsic factors have been shown to play an important role in aberrant myeloid cell behavior. The pathogenesis of chronic inflammation may be due to dysfunction of db-derived neutrophils. microRNAs (miRNAs) have been shown to globally regulate a large range of biological processes within a cell. Therefore, in order to clarify the molecular mechanism of regulation of inflammation in db-derived neutrophils, we screened for changes in miRNAs in db-derived neutrophils. miRNAs are differentially expressed in db-derived neutrophils, particularly members of the miR-129 family. Our results suggest that deregulation of the miR-129 family contributes to the dysfunction of db-derived neutrophils. The retention kinetics of neutrophils and chronic inflammation may be initiated via miR-129 family-regulated genes such as Casp6, Dedd2 and Ccr2.

Project description:Neutrophil gene transcription following lipopolysaccharide exposure. Microarray analysis of lipopolysaccharide-treated human neutrophils. Neutrophils respond to infection by degranulation, release of reactive oxygen intermediates, and secretion of chemokines and cytokines; however, activation of neutrophil transcriptional machinery has been little appreciated. Recent findings suggest that gene expression may represent an additional neutrophil function after exposure to lipopolysaccharide (LPS). We performed microarray gene expression analysis of 4,608 mostly nonredundant genes on LPS-stimulated human neutrophils. Analysis of three donors indicated some variability but also a high degree of reproducibility in gene expression. Twenty-eight verifiable, distinct genes were induced by 4 h of LPS treatment, and 13 genes were repressed. Genes other than cytokines and chemokines are regulated; interestingly, genes involved in cell growth regulation and survival, transcriptional regulation, and interferon response are among those induced, whereas genes involved in cytoskeletal regulation are predominantly repressed. In addition, we identified monocyte chemoattractant protein-1 as a novel LPS-regulated chemokine in neutrophils. Included in these lists are five clones with no defined function. These data suggest molecular mechanisms by which neutrophils respond to infection and indicate that the transcriptional potential of neutrophils is greater than previously thought.

It was therefore proposed that synthesis and secretion of 24p3 by stimulated macrophages or release of 24p3 upon neutrophil degranulation sequesters iron-laden siderophores to attenuate bacterial growth. Accordingly, 24p3-deficient mice are susceptible to bacterial pathogens whose siderophores would normally be chelated by 24p3. Specific granule deficiency (SGD) is a rare congenital disorder characterized by complete absence of proteins in secondary granules. Neutrophils from SGD patients, who are prone to bacterial infections, lack normal functions but the potential role of 24p3 in neutrophil dysfunction in SGD is not known. Here we show that neutrophils from 24p3-deficient mice are defective in many neutrophil functions. Specifically, neutrophils in 24p3-deficient mice do not extravasate to sites of infection and are defective for chemotaxis. A transcriptome analysis revealed that genes that control cytoskeletal reorganization are selectively suppressed in 24p3-deficient neutrophils. Additionally, small regulatory RNAs (miRNAs) that control upstream regulators of cytoskeletal proteins are also increased in 24p3-deficient neutrophils. Further, 24p3-deficient neutrophils failed to phagocytose bacteria, which may account for the enhanced sensitivity of 24p3-deficient mice to both intracellular (Listeria monocytogenes) and extracellular (Candida albicans, Staphylococcus aureus) pathogens. Interestingly, Listeria does not secrete siderophores and additionally, the siderophore secreted by Candida is not sequestered by 24p3. Project description:Neutrophils are one of the first responders to infection and are a key component of the innate immune system through their ability to phagocytose and kill invading pathogens, secrete antimicrobial molecules and produce extracellular traps. Neutrophils are produced in the bone marrow, circulate within the blood and upon immune challenge migrate to the site of infection. We wanted to understand whether this transition shapes the mouse neutrophil protein landscape, how the mouse neutrophil proteome is impacted by systemic infection and perform a comparative analysis of human and mouse neutrophils. Using quantitative mass spectrometry we reveal tissue-specific, infection-induced and species-specific neutrophil protein signatures. We show a high degree of proteomic conservation between mouse bone marrow, blood and peritoneal neutrophils, but also identify key differences in the molecules that these cells express for sensing and responding to their environment. Systemic infection triggers a change in the bone marrow neutrophil population with considerable impact on the core machinery for protein synthesis and DNA replication along with environmental sensors. We also reveal profound differences in mouse and human blood neutrophils, particularly their granule contents and their receptor repertoires. Our proteomics data provides a valuable resource for understanding neutrophil function and phenotypes across species and model systems.

Project description:To identify transcriptional diversity of neutrophils in steady state we performed single cell sequencing analysis on 13033 individual neutrophils across 6 different body compartments (bone marrow, blood, spleen, lung, liver and intestine). Dimensional reduction analysis identified unprecedented heterogeneity of the mature neutrophil compartment in healthy mice. Maturation state and tissue origin were both highly associated with neutrophil diversity. Moreover, we found that transcriptional profiles of discrete neutrophil clusters were enriched in genes associated with specific functions or disease states, These results identified preexisting heterogeneity in neutrophils under steady state and suggest that a wide array of ready-to-use functional programs regulate neutrophil function both in health and disease.

Project description:Inducing ferroptosis in tumor cells serves as a promising strategy for treating malignancies that are refractory to traditional treatment modalities. However, the consequences of ferroptosis in immune cells in the tumor microenvironment (TME) are not completely understood. In this study, we found that neutrophils in chemoresistant breast cancer exhibited notable vulnerability to ferroptosis. In chemoresistant neutrophils, the cellular phospholipid profile was reprogrammed owing to a deficiency in membrane-bound O-acyltransferase domain-containing 1 (MBOAT1), shifting the preference of the phospholipid fatty acid pattern from monounsaturated fatty acids (MUFAs) to polyunsaturated fatty acids (PUFAs), thereby increasing susceptibility to ferroptosis. Ferroptotic neutrophils significantly suppress the proliferation and cytotoxicity of antitumor CD8 T cells via PGE2, IDO, and oxidized lipids. Furthermore, we found that neutrophil ferroptosis was closely associated with a distinctive subset of IL-1+ CXCL3+ CD4 (Fer-CD4) T lymphocytes that were enriched in chemoresistant tumors. Fer-CD4 T cells regulated neutrophil ferroptosis through MBOAT1 modulation via IL-1/IL-1R1/NF-B signaling. Moreover, we revealed that there is crosstalk between neutrophils and Fer-CD4 T cells, during which Fer-CD4 T cells replenish the neutrophil pool of the TME via CXCL3, IL-8, and S100A9. The ferroptotic neutrophils in turn promote Fer-CD4 T cell differentiation, resulting in extensive neutrophil ferroptosis. In spontaneously tumorigenic mice, targeting IL-1+ CD4 T cells or IL-1R1+ neutrophils disrupted this crosstalk, suppressed neutrophil ferroptosis, enhanced antitumor immunity, and overcame chemoresistance. Our findings revealed the immune-reshaping capacity of neutrophil ferroptosis and its reciprocal feedback regulation with Fer-CD4 T cells, identifying promising targets for restoring anticancer immunosurveillance and reversing chemoresistance
