Quantitative proteomics reveals differences in response of neutrophils isolated from healthy or diabetic subjects to infection with capsule-variant Burkholderia thailandensis

ABSTRACT: In this study, neutrophils isolated from healthy or diabetic subjects were infected with Burkholderia thailandensis E555, a variant strain with a B.pseudomallei-like capsular polysaccharide used here as a surrogate micro-organism for B. pseudomallei. At two hours post-infection, neutrophil proteins were subjected to 4-plex iTRAQ-based comparative proteomics analysis iTRAQ-based comparative proteomic analysis.

Project description:Abstract S. aureus is a major opportunistic pathogen infecting patients with diabetes. Increased mortality was observed following IV S. aureus infection in diabetic mice compared to non-diabetic controls, correlating with increased numbers of low density neutrophils (LDNs) and neutrophil extracellular traps (NETs). LDN development is dependent on TGFβ, which was more activated in the diabetic host. Neutralization of TGFβ, or the integrin responsible for its activation α5β8, reduced numbers of LDNs and improved survival. Comparison of high and low density neutrophils identified PTEN signaling as a central regulator of LDN NET release. Inhibition of PTEN improved survival and decreased NET production in infected diabetic mice. MEDI4893*, a monoclonal antibody that neutralizes alpha toxin (AT) in development for prevention of S. aureus infection, was able to block TGFβ activation, reduce LDNs and NETs, and significantly improve survival. Our data identify a population of neutrophils in infected diabetic mice which correlated with decreased survival and increased NET production. Targeting a single virulence factor of S. aureus prevented emergence of the LDN population and improved survival, supporting potential use of pathogen specific antibodies for treating diabetic infections.

Identification of specific microRNAs in diabetic-derived neutrophils

Project description:The recruitment and/or retention kinetics of neutrophils in diabetic (db) cutaneous wounds is aberrant, and inflammation is prolonged. Intrinsic factors have been shown to play an important role in aberrant myeloid cell behavior. The pathogenesis of chronic inflammation may be due to dysfunction of db-derived neutrophils. microRNAs (miRNAs) have been shown to globally regulate a large range of biological processes within a cell. Therefore, in order to clarify the molecular mechanism of regulation of inflammation in db-derived neutrophils, we screened for changes in miRNAs in db-derived neutrophils. miRNAs are differentially expressed in db-derived neutrophils, particularly members of the miR-129 family. Our results suggest that deregulation of the miR-129 family contributes to the dysfunction of db-derived neutrophils. The retention kinetics of neutrophils and chronic inflammation may be initiated via miR-129 family-regulated genes such as Casp6, Dedd2 and Ccr2.

Project description:Changes in gene expression in pancreatic Î² cells from type 2 diabetes should provide insights into their abnormal insulin secretion and turnover. Gene expression profile of Î² cells obtained from type 2 diabetic subjects and non-diabetic controls was evaluated by microarray analysis. Frozen sections were obtained from cadaver pancreases of 10 control and 10 type 2 diabetic human subjects. Î²-cell enriched samples were obtained by laser capture microdissection. RNA was extracted, amplified and subjected to microarray analysis.

Multi-modal proteomic target discovery and orthogonol confirmation of preclinical diabetic retinopathy drug development biomarkers

Project description:The aim of this study was to identify proteomic alterations associated with functional dysregulation of the retina with diabetes that could eventually be used as surrogate endpoints in preclinical drug testing studies. A multi-modal approach of antibody (Luminex)-, electrophoresis (2-DIGE)-, and LC-MS (iTRAQ)-based quantitation methods was used to provide broad coverage of the retinal proteome. Transcriptional profiling through microarray analysis was also included to increase coverage and provide insight into potential regulation of protein expression changes at the mRNA level. The different technologies proved complementary, with limited coverage overlap between methods. Diabetes was induced in Sprague-Dawley male rats (Charles River Laboratories, Wilmington, MA) by intraperitoneal injection of 65 mg/kg streptozotocin (STZ) (Sigma-Aldrich, St. Louis, MO) in 10mM sodium citrate pH 4.5 vehicle. Control rats were injected with an equal dose of vehicle only. Rats had free access to food and water, and were maintained on a 12 hour light/dark cycle. Blood glucose level and body weight were measured 6 days post-STZ or vehicle injection, and biweekly throughout the experiment. Only rats with blood glucose levels >250 mg/dL at the time of the original test and throughout the experiment were included in the diabetic groups. At the time of retina harvest, rats were given a lethal dose of pentobarbital, 100 mg/kg, (Ovation Pharmaceuticals Inc., Deerfield, IL) by intraperitoneal injection and sacrificed by decapitation. Retinas were rapidly excised snap- frozen in liquid nitrogen for subsequent experimentation.

Neutrophils expressing lysyl oxidase-like 4 protein are present in colorectal cancer liver metastases resistant to anti-angiogenic therapy

Project description:Colorectal cancer liver metastases (CRCLM) that present with a replacement histopathological growth pattern (HGP) are resistant to neoadjuvant anti-angiogenic therapy. Surrogate biomarkers are not available to preoperatively identifypatients with these tumors. Here we identify differentially expressed genes between CRCLM with a replacement HGP and those with a desmoplastic HGP using RNA sequencing.We demonstrate that LOXL4 is transcriptionally upregulated in replacement HGP CRCLM compared with desmoplastic HGP CRCLM and the adjacent normal liver. Interestingly, lysyl oxidase-like 4 (LOXL4) protein was expressed by neutrophils present in the tumor microenvironment in replacement HGP CRCLM. We further demonstrate that LOXL4 expression is higher in circulating neutrophils of cancer patients compared with healthy control patients and its expression can be induced by stimulation with lipopolysaccharide and TNF-alpha. Our study is the first to show the expression of LOXL4 in neutrophils and reveals the potential for LOXL4-expressing neutrophils to support the replacement HGP phenotype and to serve as a surrogate biomarker for this subtype of CRCLM.

Project description:miR-193b-3p and miR-365-3p expression values are increased in pancreas infiltrating neutrophils and basophils of new-onset diabetic NOD mice. We performed CITE-sequencing technology to investigate their phenotype and frequency. Using differentially expressed gene (DEGs) analysis, we further compared the different transcriptome profiles of pancreas infiltrating versus blood circulating neutrophils and basophils in new-onset diabetic NOD mice.
