Cryo-EM structure of photosystem II supercomplex from Chlorella ohadii, a green microalga with extreme phototolerance

Project description:Photosystem II (PSII) is essential for energy conversion during oxygenic photosynthesis in plants and algae. Chlorella ohadii, one of the fastest multiplying green alga, thrives under the harsh desert sun but lacks the standard PSII photoprotective mechanisms involving LhcSR/PsbS proteins or protein phosphorylation. Here, we present analysis of cryo-EM structure of the PSII supercomplex from C. ohadii at 2.9Å resolution to determine whether the exceptional resistance to desert conditions has a structural basis in PSII. The structure reveals a novel PsbO isoform and additional subunits, PsbR and PsbY, which enhance core complex stability through extensive interactions. Furthermore, the trimeric light-harvesting complexes (LHCII) are bound to the PSII core by specific light-harvesting proteins whose down-regulation in response to high light conditions implies a reduction in the number of bound LHCII trimers. These modifications play a crucial role in maintaining PSII stability and photoprotection, allowing C. ohadii to survive in extreme conditions.

High-resolution cryo-EM structure of the respiratory alternative complex III from Rhodothermus marinus

Project description:Electron transfer in respiratory chains generates the electrochemical potential that serves as the energy source for ATP synthesis, solute transport and motility. In eukaryotes and many bacteria, the respiratory chain consists of four electron transport complexes, known as complex I to IV. Respiratory chains of some prokaryotes differ in composition and organization. They can use a wide range of electron donors and acceptors and may have complexes performing the same catalytic reaction. The diversity and apparent redundancy of prokaryotic respiratory chains reflects the versatility and robustness of the organisms. Many of these alternative respiratory chain complexes are either unknown or their structures and mechanisms remain elusive. In this work we describe a 3.9 A cryo-EM structure of the alternative complex III (ACIII) from Rhodothermus marinus, which we demonstrated takes over the role of canonical respiratory complex III (bc1 complex), even though it is structurally unrelated. Our structure reveals that ACIII is an integral membrane protein complex of at least 7 subunits (ActABCDEFH). The periplasmic domain, with ActA, B, E and H, harbours four iron-sulphur clusters and six C-type hemes. The cofactors form two electron wires that converge on the putative quinol-binding site in subunit ActC. The two homologous subunits, ActC and ActF, each have two four-helix bundles in the membrane, with several conserved polar residues that delineate putative proton channels. ACIII meets all requirements for an energy-transducing machine that couples electron transfer from quinol to the oxygen reductase, to translocation of protons across the membrane.

Structural basis of respiratory complexes adaptation to cold temperatures

Project description:In response to cold, mammals activate brown fat for respiratory-dependent thermogenesis reliant on the electron transport chain. Yet, the structural basis of respiratory complex adaptation to cold remains elusive. Herein we combined thermoregulatory physiology and cryo-EM to study endogenous respiratory supercomplexes exposed to different temperatures. A cold-induced conformation of CI:III2 (termed type 2) was identified with a ~25 rotation of CIII2 around its inter-dimer axis, shortening inter-complex Q exchange space, and exhibiting different catalytic states which favor electron transfer. Large-scale supercomplex simulations in lipid membrane reveal how unique lipid-protein arrangements stabilize type 2 complexes to enhance catalytic activity. Together, our cryo-EM studies, multiscale simulations and biochemical analyses unveil the mechanisms and dynamics of respiratory adaptation at the structural and energetic level.

Cryo-EM structure and function of S. pombe complex IV with bound respiratory supercomplex factor.

Project description:Fission yeast Schizosaccharomyces pombe serves as model organism for studying higher eukaryotes. We combined the use of cryo-EM and spectroscopy to investigate the structure and function of affinity purified respiratory complex IV (CIV) from S. pombe. The reaction sequence of the reduced enzyme with O2 proceeds over a time scale of µs-ms, similar to that of the mammalian CIV. The cryo-EM structure of CIV revealed eleven subunits as well as a bound hypoxia-induced gene 1 (Hig1) domain of respiratory supercomplex factor 2 (Rcf2). These results suggest that binding of Rcf2 does not require the presence of a CIII-CIV supercomplex, i.e. Rcf2 is a component of CIV. An AlphaFold-Multimer model suggests that the Hig1 domains of both Rcf1 and Rcf2 bind at the same site of CIV suggesting that their binding is mutually exclusive. Furthermore, the differential functional effect of Rcf1 or Rcf2 is presumably caused by interactions of CIV with their different non-Hig1 domain parts.

Development, Cryo-EM structure and function of potent monospecific and bispecific monoclonal antibodies that neutralize SARS-CoV-2 and variant B.1.351

Project description:The COVID-19 pandemic prompted an unprecedented effort to develop effective countermeasures against SARS-CoV-2. While efficacious vaccines and certain therapeutic monoclonal antibodies are available, here, we report the development, cryo-EM structures and functional analyses of distinct potent monoclonal antibodies (mAbs) that neutralize SARS-CoV-2 and its variant B.1.351. We established a platform for rapid identification of highly potent and specific SARS-CoV-2-neutralizing antibodies by high-throughput B cell receptor single cell sequencing of spike receptor binding domain immunized animals. We identified two highly potent and specific SARS-CoV-2 neutralizing mAb clones that have single-digit nanomolar affinity and low-picomolar avidity. We also generated a bispecific antibody of these two lead clones. The lead monospecific and bispecific antibodies showed strong neutralization ability against prototypical SARS-CoV-2 and the highly contagious South African variant B.1.351 that post a further risk of reducing the efficacy of currently available therapeutic antibodies and vaccines. The lead mAbs showed potent in vivo efficacy against authentic SARS-CoV-2 in both prophylactic and therapeutic settings. We solved five cryo-EM structures at ~3 resolution of these neutralizing antibodies in complex with the ectodomain of the prefusion spike trimer, and revealed the molecular epitopes, binding patterns and conformations between the antibodies and spike RBD, which are distinct from existing antibodies. Our recently developed antibodies expand the repertoire of the toolbox of COVID-19 countermeasures against the SARS-CoV-2 pathogen and its emerging variants.

Reorganization of Mitochondrial Respiratory Supercomplexes Induces Myoblasts Differentiation

Project description:The existence of respiratory supercomplexes (SCs) has been well acknowledged especially with the development of cryo-EM in the past few years, but their physiological relevance remains elusive. Dynamics of OXPHOS complexes is tightly correlated to cell fate switch, but whether SCs involve in this process is unknown. During C2C12 myogenesis, CIII2 and CIV as well as the subunits of CIII and CIV were slightly increasing, while SC III2+IV was decreasing, which indicates that SC reorganization may regulate myogenesis. Knockdown Cox7a2l decreased SCs III2+IV, I+III2+IV2 and I+III2+IV3, but had little effect on I+III2+IV. Transcriptome and quantitative-proteome analysis indicated that Cox7a2l silence activated myogenic gene expression and repressed cell cycle-associated gene expression. Besides, knockdown of Cox7a2l decreased CII abundance and accumulated succinate, which in turn suppressed certain key transcription factors, such as HMGA1 which is important for stemness maintenance. Our results indicated that respiratory supercomplex reorganization changed succinate homeostasis and myogenesis related gene expression profiles, thus switching cell fate.
