In an effort to map the human proteome, the Chromosome-centric Human Proteome Project (C-HPP) was recently initiated. Project description:Core histone proteins collected from normal human dermal fibroblasts were separated using a 2-dimensional reverse phase hydrophilic interaction liquid chromatography (RP-HILIC) system. Histone H4 isolated in the first dimension of the separation was analyzed using an LTQ Orbitrap Velos with a resolution of 60k for MS and MS/MS spectra. In total, 1,626 CID and 1,626 ETD spectra were acquired.

We did this comparison in the context of different sample input amounts; we analyzed differences in quantitative precision; we compared the effectiveness in identifying protein abundance changes induced by treatment with covalent inhibitors of PIN1.

Direct evidence of milk consumption from ancient human dental calculus, UK

Project description:This study investigated the consumption of milk products in the archaeological record, utilizing human dental calculus as a reservoir of dietary proteins from archaeological samples from across Eurasia. Protein extraction and generation of tryptic peptides from dental calculus was performed using a filter-aided sample preparation (FASP) protocol, modified for ancient samples, on 92 samples of archaeological dental calculus. Samples were extracted at three laboratories; the Functional Genomics Centre Zürich (FGCZ), the Centre for GeoGenetics at the National History Museum of Denmark, and BioArCh at the University of York. Sample extracts were sequenced (LC-MS/MS) using an LTQ-Orbitrap Velos (FGCZ), a Q-Exactive Hybrid Quadrupole Orbitrap and an LTQ-Orbitrap Elite (Central Proteomics Facility, Target Discovery Institute, Oxford).

Project description:These assays represent an antigen discovery screening, and epitope mapping characterization. In this screening two complete proteomes from Trypanosoma cruzi, from two different strains (CL-Brener, Sylvio X10), were displayed in the form of short peptides (tiling array, overlapped) and assayed with pooled serum samples (antibodies) from Chagas Disease patients and matched negative (healthy) subjects selected from 6 geographic regions across the Americas. Peptide arrays (slides) were incubated with pooled serum samples (primary antibodies), washed, and then incubated with a fluorescently-labeled anti-human IgG commercial antibody (secondary antibodies). Raw readouts of fluoresence (signal), as well as normalized signal values are provided in this submission for all samples analyzed. All samples were analyzed in duplicate.
