Evaluation of the Safety and Efficacy of Esperanza Extract (PA001)

Project description:This is a phase Ib/II clinical study that has two phases. In phase Ib, the safety evaluation of the extract of Petiveria alliacea (Esperanza) will be carried out in patients with metastatic gastrointestinal tumors (colon, pancreas, stomach, and biliary tract) and patients with newly diagnosed and relapsed acute leukemia. In phase IIb, the safety will continue to be evaluated, and the efficacy of the Esperanza extract will be explored in combination with chemotherapy in patients with metastatic gastrointestinal tumors (colon, pancreas, stomach, and biliary tract) with newly diagnosed acute leukemias and relapses.

Project description:This study reports the ability of WEB-2170, an antagonist of platelet-activating-factor receptor, to induce apoptosis in human acute myelogenous leukemia (AML) cells. Action mechanisms of WEB-2170 were first investigated in promyelocytic NB4 cells by DNA microarray profiling followed by morphologic, cytofluorimetric and biological analyses which were then extended to other AML cell lines including KG1, NB4-MR4, THP1 and U937, and, eventually, to blasts from patients with different AML subtypes (M0-M5).

Project description:Validation of a custom low density microarray. Gene expression differences were analysed in the following groups of blood samples and cell lines: Two haematological neoplasia cell lines: U937 (promonocytic lymphoma) versus Jurkat (T leukemia). Different types of haematological neoplasias: healthy donors, B-CLL patients, myeloid leukemia patients and U937 and Jurkat cell cultures samples. Two hematological neoplasia with different origin (lymphoid for B-CLL and myeloid for the myeloid group). Two types of myeloid leukemias (Acute myeloid leukemia and myelodisplastic syndrome). B-CLL patients with different clinical behaviour. Healthy donors and B-CLL patients. Normalization was performed using two different methods, robust quantile normalization (QRN) and variance stabilization normalization (VSN), so that a set of statistcially significant probes found using both methods could be obtained.

Transcription profiling of human U937 cells expressing AML1/ETO compared to vector transfected U937 cells to identify genes regulated by AML1/ETO

Project description:Approximately 20% of Acute Myelogenous Leukemia (AML) cases carry the t(8;21) translocation, which involves the AML1 and ETO genes, and express the resulting AML1/ETO fusion protein that functions as a transcriptional repressor by recruiting NCoR/SMRT/HDAC complexes to DNA. We used microarrays to identify genes differentially expressed in U937 cells expressing AML1/ETO compared to vector transfected U937 cells. Experiment Overall Design: A U937 cell line that conditionally expresses HA-tagged AML1/ETO under the control of the mouse metallothionine promoter (U937-A1E) (Alcalay et al., J.Clin.Invest, 2003,112, 1751-1761) was used. A cell line caryying the empty vector was used as control Cells were treated for 8h with 100uM ZnSO4 to induce transgene expression. For each of the U937 cell lines (AML1/ETO and Mt), three independent RNA extractions were performed, and an equal quantity of each of the three RNA preparations was then mixed to generate an RNA pool for each sample.

To explore molecular function of USP2 in macrophages especially during lipopolysaccharide(LPS) response, we assess expression profiles of HL60-derivatives continuously expressing shRNA for USP2 and control shRNA.

Effects of PMA on exon usage of HL60, THP-1, and U937 cells

Project description:Myeloid leukemia cell lines HL60, THP-1, and U937 undergo macrophage-like differentiation after treatment with phorbol ester. To explore genes whose exon usage was altered during macrophage differentiation, we compared exome of PMA-treated (differentated) and vehicle-treated (undifferentiated) myeloid cell lines. HL60, THP-1, and U937 cells were treated with either phorbol 12-myristate 13-acetate (PMA,30nM) or its vehicle (DMSO) for 3 days, and subjected to exome analysis using Affymetrix human exon 1.0ST arrays.
