Project description:Bothrops leucurus is identified as a snake of medical interest in the State of Bahia, Brazil. However, so far, there are no studies that provide a refined mapping of the composition of this venom. The aim of this work was to deepen the knowledge of the toxins from the B. leucurus venom and to isolate and biologically characterize the most abundant toxin, a new basic PLA2 like. Shotgun proteomics approach identified 137 protein hits in B. leucurus venom subdivided into 19 protein families. These findings may enable the improvement of the clinical management of accidents caused by this snakebite.

Comparative analysis of high molecular mass subproteomes of Bothrops snake venoms

Project description:Snake venoms are extremely active biological secretions containing primarily various classes of enzymes. The genus Bothrops comprises various pit viper species that represent the most medically significant taxa in Central and South America, accounting for more human envenomations and fatalities than any other snake taxa. Venom proteomes of many Bothrops species have been characterized but, although proteins of molecular mass higher than 100 kDa have been documented in Bothrops venoms, these large proteins have not yet been closely investigated. This study sought to achieve detailed identification of major components in the high molecular mass subproteome of venoms from eight Bothrops species (B. brazili, B. cotiara, B. insularis, B. jararaca, B. jararacussu, B. leucurus, B. moojeni and B. neuwiedi). The identification of proteins eluting in the first fractions of a size-exclusion chromatography of these venoms revealed that they are comprised mainly of enzymes, including minor components such as 5'-nucleotidase, aminopeptidase, phosphodiesterase, and phospholipases A2 and B, but with metalloproteinases and L-amino acid oxidases representing the most abundant components. Most of these components disappeared in electrophoretic profiles under reducing conditions suggesting that they may be composed of more than one polypeptide chain. A significant shift in the molecular masses of these protein bands was observed after enzymatic N-deglycosylation, indicating that they may contain N-glycans. Furthermore, the finding that none of the high molecular mass proteins is shared by all eight species reveals a high level of interspecies venom variability among these components.

Characterization of extracellular vesicles from Bothrops jararaca snake venom

Project description:Venoms are important evolutionary keys and innovation for several animal taxons., including snakes. Their use is specially related to feed, predation and defense. Snake venoms are composed mainly by free secreted proteins, about 98%, and stored in the lumen of the venom gland, where they are processed through poorly understood mechanisms. In the perspective of protein-diversity, venoms undergo evolutionary pressure which generate a rapid evolutionary response, causing a great diversity in their toxin-components. We know now that extracellular vesicles exist in snake venom, although their biological role are still unknown. We believe that understanding EV-mediated effects could change our way of seeing envenoming, especially long-term sequelae. From what we know about EVs and snake venoms, there is a lot of potential of cross-organism communication occurrence between snakes and human victims. To advance in the comprehension of venom EVs function, we used fresh B. jararaca venom as our model. Fresh venom was fractionated by sequential centrifugation, resulting in two populations of vesicles (Bj-EVs). Purified Bj-EVs were analyzed by electron microscopy, NTA and proteomics. The interaction of Bj-EVs with mammalian cells was accessed by fluorescence and electron microscopy.
