Iron starvation in Bifidobacterium breve UCC2003 increases bile resistance through enhanced expression of a bile salt hydrolase

Project description:Bifidobacterium are considered to be beneficial for human health and are classified as probiotic bacterium. They must resist many environmental stress factors in order to survive in the gastrointestinal environment including; pH, oxygen availability, bile and nutrient starvation (eg: iron or carbon). This study investigates Bifidobacterium breve UCC2003 global genome response to growth under ferrous and/or ferric iron limiting conditions. Revealing that growth under iron limitation effects many processes in the cell including carbon and nitrogen metabolism and induces/reduces the expression of numerous genes; including multiple iron uptake systems, DPS proteins (which are predicted to be involved in iron storage/DNA protection), Fe-S cluster associated proteins and a bile salt hydrolase (bshB). Insertional mutagenesis and survival assays were employed and demonstrated that iron starvation imposed on B. breve UCC2003 results in an increased resistance to bile stress due to in part the iron-inducible transcription of the bshB gene. Furthermore, this study links BSH activity in B. breve UCC2003 to its ability to survive the deleterious effects of bile salt and suggest that B. breve UCC2003 may be use iron as a signal to adapt to the constantly changing environment within the small intestine.

Obeticholic acid accelerates portal vein embolization-induced liver hypertrophy in rabbits by improving bile salt homeostasis

Project description:Purpose: Obeticholic acid (OCA), an agonist of the bile salt-activated transcription factor farnesoid X receptor (FXR), accelerates liver growth following portal vein embolization (PVE) in rabbits, with largest effect seen on day 3 after PVE (volume gain of 56% vs. 26% in the control group). The aim of this research was to study underlying mechanisms. Methods: Specimens from rabbits treated by oral gavage with OCA or vehicle (n=6-11/group) and subjected to PVE were analyzed, focusing on aspects of bile salt homeostasis, FXR-regulated gene expression in the ileum and liver, and their correlation with liver growth parameters. Results: Following PVE, OCA treatment led to earlier normalization of serum total bile salts and reduced bile salt content in the non-embolized liver lobe. Serum levels of the hepatotoxic bile salt lithocholic acid were lower in OCA-treated rabbits on day 1 (p=0.002) and day 3 (p=0.03) post-PVE. Furthermore, serum lithocholic acid showed a strong negative correlation with liver volume gain (ρ=‑0.726, p<0.001) and with the number of proliferating hepatocytes (ρ=-0.718, p=0.009). OCA treatment induced expression of mitotic entry regulator CDC25B in the non-embolized lobe. Untargeted transcript analysis identified candidate signaling factors (C1Qa, BMP3, IHH) as potential mediators of the growth-enhancing effect of OCA in this model. Conclusion: OCA improved bile salt homeostasis, which was associated with augmented hypertrophy of the non-embolized liver lobe in rabbits subject to PVE. OCA treatment may represent an approach to increase the efficacy of PVE in patients with a small future liver remnant planned for extended partial liver resection.

Hepatic Farnesoid X-Receptor Isoforms α2 and α4 Differentially Modulate Bile Salt and Lipoprotein Metabolism in Mice

Project description:The nuclear receptor FXR acts as an intracellular bile salt sensor that regulates synthesis and transport of bile salts within their enterohepatic circulation. In addition, FXR is involved in control of a variety of crucial metabolic pathways. Four FXR splice variants are known, i.e. FXRα1-4. Although these isoforms show differences in spatial and temporal expression patterns as well as in transcriptional activity, the physiological relevance hereof has remained elusive. We have evaluated specific roles of hepatic FXRα2 and FXRα4 by stably expressing these isoforms using liver-specific self-complementary adeno-associated viral vectors in total body FXR knock-out mice. The hepatic gene expression profile of the FXR knock-out mice was largely normalized by both isoforms. Yet, differential effects were also apparent; FXRα2 was more effective in reducing elevated HDL levels and transrepressed hepatic expression of Cyp8B1, the regulator of cholate synthesis. The latter coincided with a switch in hydrophobicity of the bile salt pool. Furthermore, FXRα2-transduction caused an increased neutral sterol excretion compared to FXRα4 without affecting intestinal cholesterol absorption. Our data show, for the first time, that hepatic FXRα2 and FXRα4 differentially modulate bile salt and lipoprotein metabolism in mice.

Development of a covalent inhibitor of gut bacterial bile salt hydrolases.

Project description:Bile salt hydrolase (BSH) enzymes are widely expressed by human gut bacteria and catalyze the gateway reaction leading to secondary bile acid formation. Bile acids regulate key metabolic and immune processes by binding to host receptors. There is an unmet need for a potent tool to inhibit BSHs across all gut bacteria to study the effects of bile acids on host physiology. Here, we report the development of a covalent pan-inhibitor of gut bacterial BSHs. From a rationally designed candidate library, we identified a lead compound bearing an alpha-fluoromethyl ketone warhead that modifies BSH at the catalytic cysteine residue. This inhibitor abolished BSH activity in conventional mouse feces. Mice gavaged with a single dose of this compound displayed decreased BSH activity and decreased deconjugated bile acid levels in feces. Our studies demonstrate the potential of a covalent BSH inhibitor to modulate bile acid composition in vivo.

Candida tropicalis biofilm formation under secondary bile salt sodium deoxycholate

Project description:Candida species are components of the normal intestinal microbiota and are under constant exposure to bacterial metabolites, including secondary bile salts. Here, we examined the effect of a secondary bile salt, sodium deoxycholate (NaDCA), on the formation of biofilms by Candida tropicalis. In contrast to C. albicans, C. tropicalis tended to maintain its absolute biofilm biomass and surface hydrophobicity in the presence of NaDCA. Fluorescent 3D microscopic imaging of the biofilm revealed that NaDCA treatment reduced filamentous projection to the top of the biofilm. RNA-seq analysis revealed that some genes, especially those associated with iron metabolism, were differentially expressed in NaDCA-treated C. tropicalis. Although NaDCA altered the appearance of C. tropicalis biofilms, analysis of the expression of key virulence factor genes encoding agglutinin-like sequences and candidalysin revealed that these genes were less affected by NaDCA in C. tropicalis than in C. albicans. High-iron exposure had a negative effect on C. tropicalis biofilm biomass. These results suggest a difference in the intestinal niche occupied by C. albicans and C. tropicalis according to the local availability of secondary bile salts.

Chemical Proteomics Reveals Regulation of Bile Salt Hydrolases via Oxidative Post-translational Modifications

Project description:Bile salt hydrolase is a cysteine hydrolase that catalyzes the deconjugation of amino acids from bile acid amidates. The catalytic residue, cysteine 2, must be in a thiolate form for hydrolysis to occur. Using a chemical probe, CA-Dim (file name suffix 'D'), we identified cysteine 2 reversibly forms a sulfinic acid, whose profiling was enabled by this probe. We profiled this posttranslation modification in the murine fecal microbiome, comparing to a DMSO vehicle control (file name suffix 'C') and a thiolate-selective probe, CA-FMK (file name suffix 'F').

A selective gut bacterial bile salt hydrolase alters host metabolism

Project description:The human gut microbiota impacts host metabolism and has been implicated in the pathophysiology of obesity and metabolic syndromes. However, defining the roles of specific microbial activities and metabolites on host phenotypes has proven challenging due to the complexity of the microbiome-host ecosystem. Here, we identify strains from the abundant gut bacterial phylum Bacteroidetes that display selective bile salt hydrolase (BSH) activity. Using isogenic strains of wild-type and BSH-deleted Bacteroides thetaiotaomicron, we selectively modulated the levels of the bile acid tauro-b-muricholic acid in monocolonized gnotobiotic mice. B. thetaiotaomicron BSH mutant-colonized mice displayed altered metabolism, including reduced weight gain and respiratory exchange ratios, as well as transcriptional changes in metabolic, circadian rhythm, and immune pathways in the gut and liver. Our results demonstrate that metabolites generated by a single microbial gene and enzymatic activity can profoundly alter host metabolism and gene expression at local and organism-level scales.
