Proteomics - LCWE Kawasaki Model

ABSTRACT: Murine vasculitis was induced via Lactobacillus casei cell wall extract (LCWE) injection in 5-week-old male mice. Groups of mice were injected with LCWE alone, LCWE and the IL-1 receptor antagonist anakinra, or saline for controls. Upper heart tissue was assessed by quantitative mass spectrometry analysis.

Project description:To explore the roles of IL-17 in asthma and the relationship between IL-17 and miRNAs, we used a model of severe asthma driven by chronic respiratory exposure to house dust mite (HDM) exposure in wild type and IL-17KO mice, followed with miRNA profiling assays and analysis. Male and female C57BL/6 mice (6-8 weeks old) and IL-17KO mice (C57BL/6 background) were exposed to purified HDM extract intranasally for 5 days/week for 5 consecutive weeks. Sterile saline was used as the control.

Suppression of molecular inflammatory pathways by Toll-like Receptor 7, 8, and 9 antagonists in a model of IL-23-induced skin inflammation

Project description:Psoriasis is a complex inflammatory disease resulting from the activation of T helper (Th) 1 and Th17 cells. Recent evidence suggests that abnormal activation of Toll-like receptors (TLRs) 7, 8 and 9 contributes to the initiation and maintenance of psoriasis. We have evaluated the effects of TLR antagonists on the gene expression profile in an IL-23-induced skin inflammation model in mice. Psoriasis-like skin lesions were induced in C57BL/6 mice by intradermal injection of IL-23 in the dorsum. Two TLR antagonists were compared: IMO-3100, an antagonist of TLRs 7 and 9, and IMO-8400, an antagonist of TLRs 7, 8 and 9, both of which previously have been shown to reduce epidermal hyperplasia in this model. Skin gene expression profiles of IL-23-induced inflammation were compared with or without TLR antagonist treatment. IL-23 injection resulted in alteration of 5100 gene probes (fold change ≥ 2, FDR < 0.05) including IL-17 pathways that are up-regulated in psoriasis vulgaris. Targeting TLRs 7, 8 and 9 with IMO-8400 resulted in modulation of more than 2300 mRNAs while targeting TLRs 7 and 9 with IMO-3100 resulted in modulation of more than 1900 mRNAs. Both agents strongly decreased IL-17A expression (>12-fold reduction), normalized IL-17 induced genes such as beta-defensin and CXCL1, and normalized aberrant expression of keratin 16 (indicating epidermal hyperplasia). These results suggest that IL-23-driven inflammation in mouse skin may be dependent on signaling mediated by TLRs 7, 8, and 9 and that these receptors represent novel therapeutic targets in psoriasis vulgaris and other diseases with similar pathophysiology. Identifying overtraining biomarkers through proteomic analysis of extracellular vesicles derived from the central nervous system of male mice

Project description:Overtraining, which results in central nervous system (CNS)-fatigue-related symptoms, lead to a long-term decline in performance. This study investigated CNS-derived EV protein content under overtraining conditions to identify potential biomarkers. Eight-week-old C57BL/6J mice were randomly divided into three groups: sedentary (SED, n=7), exercise control (EX, n=8), and overtraining (OT, n=9) groups. The OT underwent an 8-week downhill treadmill-based overtraining induction protocol. Exercise capacity was assessed using the incremental load, exhaustion, grip strength, and rotarod tests, while motor deficits, depression and anxiety were assessed using the nest building test. Proinflammatory cytokines were measured in blood plasm, skeletal muscle, and brain tissue. CNS-derived EVs were isolated using a two-step EV isolation protocol. Isolated EVs underwent proteomic analyses. Mice exhibited a significant decrease in aerobic exercise capacity, high-intensity exercise tolerance, and muscular strength. OT increased quadriceps IL-6 and hypothalamic IL-1β/TNFα compared to EX. Plasma IL-2 levels tended to be higher in OT than in EX. Proteomic analysis of CNS-derived EVs revealed a decrease in lipid metabolism-related proteins. Valosin-containing proteins and catalase, which are upregulated in organs under oxidative stress, were increased. Therefore, CNS-derived EV protein contents indicated CNS fatigue under overtraining conditions.

Comparison of mouse genes from the lungs of chronic, acute, tolerant, and non-asthmatic mouse models

Project description:Microarray analysis of whole lung tissue from three mouse models of asthma Acute asthma was produced by immunization twice, one week apart, of Balb/C mice 8-12 weeks of age with Aspergillus species (5 μg) in adjuvant (1:1 vol/vol). Adjuvant was aluminum and magnesium hydroxide (Pierce). Asthma was initiated by three consecutive intranasal exposures to Aspergillus species (5μg in 15μL saline) and asthma was evaluated 72 hours after the final exposure. Tolerant asthma was produced by intranasal delivery of Aspergillus species (5μg in 15μL saline) twice a week for six consecutive weeks to Balb/C mice 8-12 weeks of age. Asthma was evaluated after mice were rested for an additional three weeks. Chronic asthma was produced by intranasal delivery of the Dustmite/ Ragweed/ Aspergillus (5,50,5ug in 15ul saline) mixture twice a week for six consecutive weeks in female mice Balb/C mice 8-12 weeks of age. Asthma was evaluated 3 weeks after cessation of allergen exposure. Saline control mice were treated intransally with 15ul of saline twice a week for 6 weeks. Asthma was assesed 3 weeks after the final exposure.

Aged Mouse Muscle: Sedentary or Exercised Animals with or without Nicotinamide N-methyltransferase Inhibitor Treatment

Project description:Aged (22-mo) female mice (n=35), obtained from the US NIH National Institute on Aging (NIA) Aged Rodent Colony, were randomly assigned to one of four groups: saline-treated sedentary ( n=9), nicotinamide N-methyltransferase inhibitor (NNMTi)-treated sedentary (10 mg/kg body weight; n=9), saline-treated progressive weighted wheel running (PoWeR; saline; n=10), and NNMTi-treated PoWeR (10 mg/kg body weight; n=7) . Group-housed Sed cohorts were compared to singly-housed PoWeR cohorts that underwent a 1-week introduction to an unweighted wheel, followed by eight weeks of weighted wheel running. Forelimb grip strength was assessed by a single NNMTi treatment-blinded investigator during week six and averaged across 2-4 trials/mouse. At the end of week 8, when mice were ~24.5-months-old, the strength of the right limb plantarflexor muscle complex was measured using an in vivo isometric peak tetanic torque technique and a fatigue test. After the fatigue test, mice were euthanized, and tissues were weighed and collected. Of note, hindlimb muscles from the right limb (the limb that underwent in vivo isometric peak tetanic torque and fatigue testing) were processed for immunohistochemistry to avoid acute effects of muscle functional testing on the proteome and metabolome. Hindlimb muscles from the left limb that did not undergo torque and fatigue testing were flash-frozen for proteome and metabolome analyses.
