Proteomics and ultrastructural analysis of Hermetia illucens (Diptera: Stratiomyidae) larval peritrophic matrix

ABSTRACT: This is a set of three raw data from the larval of Hermetia illucens. The total protein was extracted with protein lysis buffer (8M urea with appropriate protease inhibitor). The trypsin-digested peptides were analyzed by online nano-flow liquid chromatography tandem mass spectrometry performed on an EASY-nLC system (Thermo, USA) connected to a Q Exactive HF-X Quadrupole Orbitrap mass spectrometer (Thermo, USA) through a nanoelectrospray ion source.

Whole genome expression analysis in the third-instar larval midgut of Drosophila melanogaster

Project description:Survival of insects on a substrate containing toxic substances such as plant secondary metabolites or insecticides is dependent on the metabolism or excretion of those xenobiotics. The primary sites of xenobiotic metabolism are the midgut, Malpighian tubules and fat body. In general, these organs are treated as single tissues by online databases, but several studies have shown that gene expression within subsections of the midgut is compartmentalized. In this article, RNA sequencing analysis was used to investigate whole-genome expression in subsections of the third-instar larval midgut. The results support functional diversification in subsections of the midgut. Analysis of the expression of gene families that are implicated in the metabolism of xenobiotics suggests that metabolism may not be uniform along the midgut. These data provide a starting point for investigating gene expression and xenobiotic metabolism in the larval midgut.

Then the powder was suspended in lysis buffer [1% sodium deoxycholate (SDS), 8M urea]. The mixture was allowed to settle at 4 鎺矯 for 30 min during which the sample were vortexed at every 5 min, and treated by ultrasound at 40 kHz and 40 W for 2 min. After centrifugation at 16000 rpm at 4鎺矯 for 30 min, the concentration of protein supernatant was determined by Bicinchoninic acid (BCA) method by BCA Protein Assay Kit (Pierce, Thermo, USA). Scallop byssal protein identification

Project description:To explore the protein components for scallop byssus, the soluble fractions of scallop byssus was extract. For mass spectrometric analysis, proteins were extracted from byssal adhesive plaques, and the major SDS-PAGE fractions was treated with trypsin and analyzed using an Easy-nLC nanoflow HPLC system connected to an Orbitrap Elite mass spectrometer (Thermo Fisher Scientific, USA). The mass spectrometry raw data were searched against the full set of predicted proteins from the C. farreri genome using Mascot v2.3.0 (Matrix Science, London, UK).
