Project description:Background: There is some evidence demonstrating the effect of psychological interventions in improvements in health biological parameters. To best of our knowledge, no study had addressed the impact of any psychological intervention on extracellular vesicles. In addition, Mindfulness-Based Cognitive Therapy (MBCT) and Emotion Focused Therapy for Cancer Recovery (EFT-CR) in the group have never been explored regarding extracellular vesicles and the effectiveness of these was not compared yet.
Objectives:
1. To explore and compare the effect of MBCT and EFT-CR on biological parameters and psychological variables in distressed people who have had breast, prostate and colorectal cancer;
2. In addition, we will explore the acceptability through recruitment and retention rates of MBCT and EFT-CR in group and evaluate whether these interventions are appropriate for a larger clinical trial.
Methods: The design of this study is a parallel randomized controlled trial. Participants will be randomized into MBCT, EFT-CR or usual care. Outcome measures will be assessed before, at the end of the intervention (8 weeks) and follow-ups (24 and 52 weeks from the baseline moment).
Hypotheses: The researchers expected that both interventions will have an effect on extracellular vesicles and other study biomarkers as well as improvements in psychological outcomes, compared to treatment as usual (TAU) group. Regarding the comparative effectiveness, we did not have evidence to hypothesize which one of the interventions will be superior in both biological (extracellular vesicles) and psychological outcomes.
Contribution for practice: The results of this preliminary study would permit to know if there are benefits of these psychological interventions on changes in extracellular vesicles and on psychological outcomes related to health. In addition, this study will permit to determine the acceptability of conducting a larger randomized controlled trial.

Project description:Extracellularly released particles, including membrane vesicles, have increasingly been recognized as important for bacterial community functions and host-interaction processes, but their compositions and functional roles differ between species and also between strains of the same species. In this study, we have determined the composition of membrane vesicles and protein particles identified in the cell-free pellets of two strains of Apilactobacillus kunkeei, a defensive symbiont of honeybees. The membrane vesicles were separated from the extracellular particles using density gradient ultracentrifugation. The peaks of the RNA and protein distributions were separated from each other and the highest concentration of RNA was observed in the fractions that contained the membrane vesicles while the highest protein concentration coincided with the fractions that contained extracellular particles. A comparative proteomics analysis by LC-MS/MS showed that 37 proteins with type-I signal peptides were consistently identified across the fractionated samples obtained from the cell-free pellets, of which 29 were orthologs detected in both strains. Functional predictions of the extracellular proteins revealed the presence of glycoside hydrolases, glycosyltransferases, giant proteins and peptidases. The extracellular transcriptomes mapped to a broad set of genes with a similar functional profile as the whole cell transcriptome. This study provides insights into the composition of membrane vesicles and extracellular proteins of a bee-associated symbiont.

Project description:Extracellular vesicles are small (50-200 nm diameter) membrane-bound structures released by cells from all domains of life. Vesicles have been implicated in numerous microbial processes, including gene transfer, signaling, pathogenesis, and defense. While vesicles are abundant in the oceans, little is known about their contents or whether they interact with cells in this dilute environment. Here we show that extracellular vesicles contain a remarkable diversity of organic carbon compounds and likely mediate a complex network of export, transport and exchange within marine microbial communities. We report that vesicles from selected strains of Prochlorococcus, the most abundant cyanobacterium in the sea, and other bacteria can interact with cells of various marine microbes, including the numerically abundant heterotroph Pelagibacter. Through a combination of lipidomic, metabolomic and proteomic analyses, we show that two strains of Prochlorococcus export an enormous array of organic and inorganic compounds – including active, biogeochemically relevant enzymes – within vesicles. Vesicles from both strains contained some materials in common as well as numerous strain-specific differences, reflecting an even greater degree of functional complexity within natural vesicle populations. These data suggest additional roles for vesicles in carrying out extracellular biogeochemical reactions, mitigating toxicity from reactive oxygen species, and mediating energy and nutrient transfers in the oceans.

While extremely abundant in the oceans, our understanding of their functions, both for cells and the emergent ecosystem, is in its infancy. To advance this understanding, we analyzed the lipid, metabolite, and protein content of vesicles produced by two strains of the most abundant phytoplankton cell in the ocean, the cyanobacterium Prochlorococcus. We show that Prochlorococcus exports an enormous array of cellular compounds into their surroundings via extracellular vesicles. The vesicles produced by the two different strains contained some materials in common, but also displayed numerous strain-specific differences, reflecting functional complexity within natural vesicle populations. Prochlorococcus vesicles contain active enzymes, indicating that they can mediate biogeochemically relevant extracellular reactions in the wild. Interaction assays demonstrate that vesicles from Prochlorococcus and multiple genera of heterotrophic bacteria can associate with other marine microbes, including Pelagibacter, the most abundant heterotrophic group in the oceans. Our observations suggest that vesicles may play diverse functional roles in the oceans, including but not limited to mediating energy and nutrient transfers, catalyzing extracellular biochemical reactions, and mitigating toxicity of reactive oxygen species. These findings further indicate that a portion of the ‘dissolved’ compounds in the oceans are not truly dissolved, but are instead packaged within locally structured, colloidal vesicles.

Project description:We have investigated the genomic and epigenetic consequences of co-culturing colorectal carcinoma cells with membrane vesicles from pathogenic bacteria Vibrio cholerae and non-pathogenic commensal bacteria Escherichia coli. Our study has revealed that membrane vesicles from pathogenic and commensal bacteria have a global impact on the gene expression of coloncarcinoma cells. The changes in gene expression correlated positively with both epigenetic changes and chromatin accessibility of promoters at transcription start sites of genes induced by both types of membrane vesicles. Moreover, we have demonstrated that membrane vesicles obtained only from V. cholerae induced the expression of genes associated with tumour differentiation. Altogether, our study suggests that the observed genomic changes in host cells might be due to specific components of membrane vesicles and does not require communication by direct contact with the bacteria.
