Project description:This dataset consists of 37 raw MS files and associated peak lists and result files, acquired on TripleTOF6600 mass spectrometer operated in Data Dependent Acquisition mode. The files are associated with a manuscript submitted for publication by Segal et al. that determines the protein-protein and proximity interaction landscape of 14-3-3 proteins.
This particular dataset consists in files associated with three baits (14-3-3 interactors) profiled in both a wild type and a 14-3-3 binding site mutant. Each bait was profiled in biological duplicates by BioID (*using the BioID2 enzyme), alongside a total of 25 negative controls (used for analysis with SAINTexpress). See details within the README file.
Samples were generated by Dmitri Segal and affinity purification and mass spectrometry were performed by Zhen-Yuan Lin with oversight from Brett Larsen and Anne-Claude Gingras. Analysis was performed by Anne-Claude Gingras and Mikko Taipale.

Project description:This dataset consists of 36 raw MS files acquired on Lumos Fusion Orbitrap mass spectrometer operated in Data Dependent Acquisition mode for TMT-10plex. The files are associated with a manuscript submitted for publication by Segal et al. that determines the protein-protein and proximity interaction landscape of 14-3-3 proteins. All samples comprise either BioID or FLAG AP-MS purifications of each of the human 14-3-3 proteins (with negative controls) expressed to the same level in HEK293 Flp-In T-REx cells. Purified samples were labeled with TMT reagents, and analyzed by mass spectrometry. 32 files correspond to the BioID dataset (fractionated; MS3), and 4 files to the AP-MS dataset (non fractionated, HCD MS2).
Samples were generated by Dmitri Segal and Stefan Maier and affinity purification, labeling, mass spectrometry, and data analysis were performed by Stefan Maier.
