Discovery of Dehydroamino Acid Residues in the Capsid and Matrix Structural Proteins of HIV-1

ABSTRACT: Bottom-up mass spectrometry-based proteomic analysis (trypsin) was performed on four biological replicates of HIV-1 virions. These virions were isolated from HEK293T cells transfected with a HIV-1 proviral plasmid derived from the pNL4-3 molecular clone, rendered biosafe due to inactivating point mutations in both the env and vpr reading frames. There are 8 total spectra, 4 are from unlabeled aliquots of sample, and 4 are from aliquots of sample treated with glutathione to label dehydroamino acids.
