Prior heat stress can have a profound impact on later thermal tolerance, but the mechanism for this plasticity is not clear. The understanding of gene expression changes behind physiological acclimatization is critical in forecasts of coral health in impending climate change scenarios. Acropora millepora fragments were preconditioned to sublethal bleaching threshold stress for a period of 10 days; this prestress conferred bleaching resistance in subsequent thermal challenge, in which non-preconditioned coral bleached. Using microarrays, we analyze the transcriptomes of the coral host, comparing the bleaching-resistant preconditioned treatment to non-preconditioned and control treatments.

Project description:Coral bleaching and coral reef degradation become severe as the surface seawater temperature rises. Much research to date has focused on the bacterial community composition properties within the coral holobiont, but less attention has been paid to the interactions of bacteria and corals under thermal stress. We investigated the changes of coral symbiotic bacteria and metabolites under thermal stress, and analyzed the internal relationship between bacteria and metabolites as well as their relationship with coral health. We found obvious signs of coral bleaching after heating treatment, and the interaction within symbiotic bacterial community became closer. The coral symbiotic bacterial community and metabolites changed significantly under thermal stress, and bacteria such as Flavobacterium, Shewanella and Psychrobacter increased significantly. Bacteria associated with stress tolerance, biofilm formation and mobile elements decreased, and bacterial DMSP metabolism increased slightly after heating treatment. Differential metabolites in corals after heating treatment were associated with cell cycle regulation and antioxidant. This study revealed the correlation between differential metabolites and bacterial community composition changes in corals under thermal stress, and providing valuable insight on metabolomics research of corals.

The analysis implements 45 arrays, representing 5 sampling points of three treatments (n=3).

Project description:Publication Abstract: As climate changes, sea surface temperature anomalies that negatively impact coral reef organisms continue to increase in frequency and intensity. Yet, despite widespread coral mortality, genetic diversity remains high even in those coral species listed as threatened. While this is good news in many ways it presents a challenge for the development of biomarkers that can identify resilient or vulnerable genotypes. Taking advantage of three coral restoration nurseries in Florida that serve as long-term common garden experiments, we exposed over thirty genetically distinct Acropora cervicornis colonies to hot and cold temperature shocks seasonally and measured pooled gene expression responses using RNAseq. Targeting a subset of twenty genes, we designed a high-throughput qPCR array to quantify expression in all individuals separately under each treatment with the goal of identifying predictive and/or diagnostic thermal stress biomarkers. We observed extensive transcriptional variation in the population, suggesting abundant raw material is available for adaptation via natural selection. However, this high variation made it difficult to correlate gene expression changes with colony performance metrics such as growth, mortality, and bleaching susceptibility. Nevertheless, we identified several promising diagnostic biomarkers for acute thermal stress that may improve coral restoration and climate change mitigation efforts in the future.

Project description:Given the overwhelming evidence that symbiont genotypes differentially affect host processes such as growth, bleaching susceptibility, and nutrient acquisition, we set out to measure gene expression differences in fragments of Montastraea faveolata harboring two different clades of Symbiodinium. On the reefs near Puerto Morelos, México, colonies of M. faveolata are known to shift algal symbiont clade with depth, often associating with clade A at the top, clade B in the middle, and clade C near the bottom of the colony. By measuring photosynthetic efficiency and gene expression in control and heat-stressed fragments containing either clade B, clade C, or a mix of both, we found that: 1) the algal response to thermal stress is due to both host and algal factors; 2) fragments of M. faveolata express different genes in response to sub-bleaching thermal stress depending on algal genotype; 3) the overall effect of heat stress on coral gene expression is less significant than the effect of housing different zooxanthellae types. Overall, we present convincing evidence that different Symbiodinium clades may be functionally distinct, which in turn, greatly influences host gene expression.

Project description:Abstract The coral–dinoflagellate symbiosis is increasingly disrupted by global and local anthropogenic stressors. Coral bleaching is primarily a result of high sea surface temperatures, while eutrophication is associated with reef ecosystem degradation. Excess inorganic nitrogen relative to phosphate has been proposed to directly sensitise corals to thermal bleaching and accelerate reef decline. We assessed the proteomic response of the dinoflagellate coral symbiont Symbiodinium microadriaticum to elevated temperatures under multiple nutrient conditions by mass spectrometry. Elevated temperatures resulted in reductions of many chloroplast proteins, particularly light-harvesting complexes, with simultaneous increases in chaperone proteins. N:P imbalance had a larger effect on the proteome than temperature, but the biological processes and proteins responding to each stressor largely overlapped. The proteomes were highly similar at low N:P ratios but were strongly affected by phosphate starvation. High N:P ratios inhibited cell division, reflected by changes in proteins involved in protein translation. Imbalanced N:P did not increase sensitivity to high temperatures as measured by physiological means; however, imbalanced N:P strongly upregulated cell redox homeostasis proteins at high temperatures. As redox balance is critical during thermal bleaching, these data provide insight into the mechanisms of cellular responses to thermal and multiple stresses in the coral–dinoflagellate symbiosis.

Project description:Twelve coral colonies of Montipora capitata were collected from patch reefs located in Kāne’ohe Bay, O’ahu, Hawai’i. Colonies were brought to shore where they were immediately split into two equally sized pieces, which were acclimated before bleaching. Half of each colony went to an increased water temperature treatment (30°C, bleaching treatment) and the other stayed at ambient temperature (25°C, nonbleached treatment) for three weeks. For the bleaching treatment, experimental tank temperatures were increased 2°C per day ( 1°C every 12 hours) for four days to a final temperature of 30°C. Samples were collected before bleaching (T1) and 24 hours after bleached colonies were returned to ambient temperature (T2), through 3 months post-bleaching (T6).

Bleaching events can span large geographic areas and are currently a potent threat to coral reefs worldwide. Much effort has been focused on understanding the molecular and cellular events underlying bleaching, and these studies have mainly utilized heat and light stress regimes. In an effort to determine whether different stressors share common bleaching mechanisms, we used cDNA microarrays for the corals Acropora palmata and Montastraea faveolata (containing > 10,000 features) to measure differential gene expression during darkness stress. This is the first coral microarray experiment aimed at darkness stress, and the first for these species to interrogate gene expression at such a large scale. Our results reveal a striking transcriptomic response to darkness in A. palmata involving chaperone and antioxidant up-regulation, growth arrest, and metabolic modifications. As these responses were also measured during thermal stress, our results suggest that different stressors may share common bleaching mechanisms. Furthermore, our results point to ER stress as a critical cellular event involved in darkness-specific (and possibly more general) molecular bleaching mechanisms. On the other hand, we identified a meager transcriptomic response to darkness in M. faveolata where gene expression differences between host colonies and/or sampling locations were greater than differences between control and stressed fragments. To this end, we discuss the importance of factors related to host genotype, Symbiodinium genotype, and the abiotic environment that influence host gene expression and thereby can hinder an investigator’s ability to measure gene expression during a condition of interest.
