Project description:M. hominis cells were grown in liquid medium supplied with arginine or thymidine as a carbon source. LC-MS analysis was performed on Ultimate 3000 Nano LC System (Thermo Fisher Scientific) coupled with Q Exactive HF benchtop Orbitrap mass spectrometer (Thermo Fisher Scientific) via a nanoelectrospray source (Thermo Fisher Scientific), an untargeted label-free bottom-up proteomic strategy was used, DDA (Data Dependent Acquisition) approach. Protein identification and label-free quantification were performed with PEAKS software.
Project description:We report a mass spectrometry-based proteomics approach to advance the study of the feline proteome. We analyzed immortalized feline cells, various tissues and plasma using data-dependent acquisition (DDA) on a Q Exactive HF mass spectrometer to develop a Felis Catus PeptideAtlas from 157 acquired runs. Data were analyzed using the Trans-Proteomic-Pipeline. We report in the first PeptideAtlas of the domestic cat the identification of 242,844 unique peptides representing 8,972 feline proteins or 45.6% of the predicted UniProtKB Felis catus proteome based on 19,655 unique genes and their representative protein sequences.

Project description:Human iPSCs and NSCs were engineered by AAVS1 and/or C13 safe-harbor TALENs which mediated targeted integration of various reporter genes at single or dual safe-harbor loci. Multiple clones of targeted human iPSCs were used to compare with parental untargeted NCRM5 iPSCs. Polyclonal targeted human NSCs were used to compare with their parental untargeted NCRM1NSCs or H9NSCs. Total RNA obtained from targeted human iPSCs or NSCs compared to untargeted control iPSCs or NSCs.
