Untargeted_Urine_Carnitine_Efficacy_Study_Males_Females

ABSTRACT: 5-week old male and female Swiss Webster mice were infected with 500,000 Trypanosoma cruzi strain Sylvio X10/4 parasites. Urine was collected from uninfected mice pre-infection, from infected and uninfected mice every week for 5 weeks post infection, from infected and uninfected mice 67 (females)/68 (males) days post infection, and from infected and uninfected mice 117 (females)/124 (males) days post infection for acute, mid-chronic, and chronic time-points. Metabolites were extracted with methanol and an untargeted LCMS analysis was ran on a Thermo Q Exactive Plus instrument with a Phenomenex Luna Omega Polar C18 column.

Host transcriptional responses during chronic Toxoplasma gondii infection reveals sex specific differences that lead to altered protection against secondary infection

Project description:We sequenced the cerebral cortex transcriptome of both male and female mice either infected with T. gondii or age-matched uninfected controls. These transcriptomes were determined along the infection timeline with early (21 and 28 days) and late (3 and 6 months) chronic represented. We obtained 5.7 billion paired-end sequences and aligned them to the Mus musculus genome. We found that a majority of the host genes with greater abundance at day 21 post-infection are still more abundant 6 months post-infection in male and female mice, although, females have far fewer genes that are significantly less abundant. Genes that were significantly less abundant in males are involved with the immune response, and may lead to the slight increase in cyst numbers in males during chronic infection.

Sex differences during acute myocarditis against coxsackievirus B3 (CVB3)-induced myocarditis (10 dpi) and chronic myocarditis/dilated cardiomyopathy (DCM) against CVB3-induced myocarditis (90 dpi)

Project description:Men are at an increased risk of dying from heart failure caused by inflammatory heart diseases such as atherosclerosis, myocarditis and dilated cardiomyopathy (DCM). We previously showed that immune responses in the heart are phenotypically distinct in male compared to female mice 10 days after infection resulting in severe DCM in males. Groups consisted of Infected Males, Infected Females, Uninfected Males and Uninfected females. There are 3 mice per group. A total of 12 samples were analyzed in this experiment (12 for 10 dpi and 12 for 90 dpi).

Project description:4-week-old male mice were infected with 50,000 CL+luc parasites or remained uninfected. Urine was collected pre-infection and 4 days after infection. 7 days post infection, mice were treated with 0.06% carnitine in drinking water ad libitum or with benznidazole 100 mg/kg ip for 10 days. 17 days post infection, urine was collected and euthanized. Metabolites were extracted from urine in methanol and untargeted LCMS analysis ran on a Q Exactive plus instrument using a Phenomenex Luna Omega Polar C18 column.

Testis proteome of Trypanosoma cruzi infected mice

Project description:Male CD-1 mice (N=8) were intraperitoneally infected by culture-derived Trypanosoma cruzi trypomastigotes of the Brazil strain. One group (N=4) was treated with an antitrypanosomal drug (nifurtimox) on day 74 post-infection (chronic phase) for 20 days. A group of uninfected mice (N=3) was also included as control. All groups (N=11) were enthanized on day 110 post-infection, testes were collected and subjected to 2D-LC-MS/MS analysis. Label-free quantitation revealed that the testes of T. cruzi-infected mice had an altered proteome pattern relative to treated mice and uninfected controls. The analysis also demonstrated that the left and right testes had different proteomic profiles in infected animals.

There are 5 mice per group. A total of 20 samples were analyzed in this experiment.

Gene expression changes in liver upon LCMV Cl13 infection.

Project description:2-3 month old female C57Bl/6J mice were intravenously infected with 2x10^6 focus forming units of lymphocytic choriomeningitis virus strain Clone-13. Whole liver tissue was harvested in duplicated from uninfected control mice, uninfected 123d old mice, and from infected at 2, 8, 30, 60, and 123 days post infection. Three TMT 6-plex runs were carried out to monitor the changes in liver protein abundance during the entire course of infection. The first run included samples from day 2 and day 8 after infection along with the uninfected controls. Similarly, the second run included uninfected controls and samples from day 30 and day 60 after infection. The third run included samples day 123 after infection, the corresponding age matched control that was left uninfected for 123 days and the uninfected controls used in the other two runs. The uninfected controls were included in all the three runs as an internal control to monitor reproducibility between the runs.
