Retrospective Timing of Tissue storage by Kinetics of Proteome Alterations - The "Proteomic Clock" concept.


ABSTRACT: Tissue samples were obtained from different organs (lung, liver and kidney) during human autopsies. Samples were left out of storage during different time intervals (6, 12, 18, 24, 48, 72, 96 hours) before sample processing. Proteins were extracted after tissue homogeneization, reduced (5 mM TCEP) and alkylated (10 mM iodoacetamide) and digested with trypsin and Lysyl endopeptidase. Sample were cleaned up with HyperSep and measured in QE Plus using a label-free DDA approach.
