Terpenes a common language between fungi and bacteria

Project description:Previously, we investigated the effect of fungal VOCs on the behavior of phylogenetically different soil bacteria (Schmidt et al 2015). In these experiments we showed that VOCs emitted by several fungi can lead to phenotypical responses in bacteria, for example, by inducing a change in motility (Schmidt et al 2015). We observed that the plant pathogenic fungus Fusarium culmorum produced a unique cluster of VOCs consisting primarily of terpenes. When exposed to the VOCs emitted by this fungus, the rhizobacterium Serratia plymuthica PRI-2C responded with an induction of motility. It is plausible that in soil, microorganisms sense changes in their environments via shifts in VOCs blend and adapt their behavior accordingly (Garbeva et al 2014). Although several studies indicated that VOCs can be used as signaling molecules in microbial inter-species interactions, the following questions remain unanswered as how are VOCs perceived as signals by the microorganisms and which regulatory pathways and genes are involved in the response? To answer these questions, the rhizosphere isolate S. plymuthica PRI-2C was grown alone or exposed to VOCs emitted by F. culmorum. The bacterial transcriptome and proteome were analyzed under each situation to identify the molecular basis of the bacterial response to fungal VOCs.

Transcriptional responses of Fusarium graminearum when exposed to bacterial MAMPs

Project description:The plant fungal pathogen Fusarium graminearum which has part of its life cycle in the soil generally comes in contact with bacteria. Therefore the fungus was used as fungal model to test fungal responses to bacterial presence and ascertain the presence of an innate immune system in fungi through a transcriptomics study. Bacterial presence was established by exposing young fungal cultures to the well characterised MAMPs (Microbe Associated Molecular Pattern) such as Flagellin (FLG), Lipooligosacharides (LOS) and Peptidoglycans (PGN) and water was used as the control. The results showed that the fungus reacts to the presence of MAMPs within hours and that the 3 MAMPs up-regulated a set of common genes. These significantly up-regulated common genes can be classified into categories such as energy generation, amino acid metabolism, iron uptake, secondary metabolism and transporters related to secondary metabolism. Analysis of the transcriptional reponse from FLG showed that the MAMP which was obtained comercially was blended with sucrose and this was later confirmed by the commercial provider.

Effect of 3,OC12-HSL on the transcriptome of the ectomycorrhizal fungus Laccaria bicolor S238N

Project description:Bacteria use small diffusible molecules to communicate and to coordinate population response by a cell density-depend mechanism named quorum sensing (QS). If such a cross-talk was first described between bacterial species, it was recently evidenced that bacterial QS signal molecules can also be sensed by eukaryotic organisms thus playing as inter-kingdom signalling molecules. Although ectomycorrhizal fungi interact physically and metabolically with myriads of bacterial species in soils, nothing is known about the ability of ectomycorrhiza-associated bacteria to produce QS molecules and the potential impact of these molecules in the interaction. While investigating the potential effect of these molecules on the physiology of the ectomycorrhizal fungus, we found that 3O,C12-HSL also induced an alteration of the transcriptome of L. bicolor S238N. This study used the microarray design as follows: cDNA libraries from pure culture of Laccaria bicolor S238N mycelium and from three stages of L. bicolor S238N sporocarp development (Lb2 library: stipes and caps of 5–10 mm growing sporocarps; Lb3 library: caps of 30– 40 mm mature sporocarps), collected under Douglas ﬁr seedlings grown in a glasshouse, were constructed in the λTriplEx2 vector. The cDNA inserts from bacterial clones were PCR-ampliﬁed and 4992 cDNAs were arrayed from 384-well microtitre plates on Nylon membranes. Three independent biological replicates were performed for the treatment (3,OC12-HSL) and for the control (ethyl acetate)
