Waste water treatment plant effluent from Muenster, Germany

Project description:Samples were collected daily (19.06.2023-10.07.2023) the in the Waste water treatment plant "Rieselfelder" in Muenster, Germany, as full-day average sample. During the collection period, a festival took place in Muenster (24.06.2023). Field blanks of miliq were processed like the samples. 250 mL of sample were filtered 0.2 um cellulose acetate filters and extracted using Agilent Bond Elut PPL cartridges. Elution was performed with MeOH. Samples were reduced to dryness under an N2 stream and dissolved in 1 mL (MeOH:H2O (1:1, v/v)). UHPLC was performed using a Waters Acquity BEH C18 column (150 mm length, 1.7 um pore size). Eluents were miliq water (0.1% FA) and Acetonitrile (0.1% FA). Data was acquired on a timsTOF fleX in PASEF mode in positive and negative polarity and an Agilent 6530A qTOF in positive and negative polarity.

Metaproteome of the waste water treatment plant Gerwisch/Germany

Project description:In this study the impact of protein fractionation techniques prior to LC/MS analysis was investigated on activated sludge samples derived at winter and summer condition from a full-scale wastewater treatment plant (WWTP). For reduction of the sample complexity, different fractionation techniques including RP-LC (1D-approach), SDS-PAGE and RP-LC (2D-approach) as well as RP-LC, SDS-PAGE and liquid IEF (3D-approach) were carried out before subsequent ITMS analysis. The derived spectra were identified by MASCOT search using a combination of the public UniProtKB/Swiss-Prot protein database and metagenome data from a WWTP (data are available via ProteomeXchange with identifier PXD001547). The results showed a significant increase of identified spectra, enabled by applying IEF and SDS-PAGE to the proteomic workflow. Based on meta-proteins, a core metaproteome and the corresponding taxonomic profile of the wastewater activated sludge was revealed and functional aspects using the KEGG (Kyoto Encyclopedia of Genes and Genomes) pathway library were analyzed. Hereby, the KEGG Orthology identifiers (KO numbers) of protein hits were plotted into pathway maps of the central carbon (map01200) and nitrogen metabolism (map00910). Using the 3D-approach, most proteins involved in glycolysis and citrate cycle and nearly all proteins of the nitrogen removal were identified, qualifying this approach as the most promising for future studies.

Project description:We reported the gene expression profile of T47D cells treated with the organic extract of Particulate matter 2.5 (PM2.5) sampled next to the municipal solid waste incineration plant of Bologna city. Based on a air pollution distribution model that takes the incinaration plant as point source of emission, two sites were chosen to sample particulate matter near incineration plant: "FrulloEst" representing the maximum effect of the incineration plant, "Calamosco" representing the negative control of "FrulloEst" (minimun effect of incineration plant, same effect of other air pollution fonts). Another site, "Giardini Margherita", is chosen to sample the urban background air pollution. for each site sample collection was performed in winter and in summer season.
