Proteomic characterization of SAS-derived extracellular vesicles following treatment with BPA and doses of neutron irradiation

ABSTRACT: Proteomic analysis of SAS cells culture medium derived extracellular vesicles, treated on not with BPA (10B-p-boronophenylalanine) and different times and doses of neutron irradiation (0 min - 0 Gy, 10 min - 1.9 Gy and 60 min - 11.3 Gy) to study the effect of Boron administration and BNCT treatment.

Immediately after irradiation we put seeds for germination to Petri dishes containing water filter paper; embryos were taken 2h after irradiation, embryos with embrionic roots were taken 24 and 48h after irradiation. The following comparisons have been made: non-irradiated (0 Gy), 2h vs. 20 Gy, 2h; 0 Gy, 24h vs. 20 Gy, 24h; 0 Gy, 48h vs. 20 Gy, 48h; 0 Gy, 24h vs. 100 Gy, 24h. Three independent biological replicates were used.

Project description:In this study, we measured mRNA and lncRNA profiles of gastric tissues following 0, 6 and 12 Gy irradiation. The stomach of C57 mice were exposed to 0, 6 and 12 Gy X-ray irradiation at a dose rate of 2 Gy/min. The mice were sacrificed 7 days after irradiation and gastric tissues were collected. RNA was extracted and quantified. mRNA and lncRNA profilings of each groups were analyzed by microarray.

Next Generation Sequencing Facilitates Quantitative Analysis of 0 and 5 Gy HuR binding RNAs

Project description:Purpose: Next-generation sequencing (NGS) has revolutionized systems-based analysis of RNA. The goals of this study are to compare HuR binding RNAs between 0 or 5 Gy irradiation, and analyze its effect on RNA expression. Methods: Human skin fibroblast cell line WS1 cells were treated with 0 or 5 Gy irradiation. After 24 h, RNA expression was measured using using Illumina HiSeq4000. Meanwhile, RIP was performed using HuR antibody. HuR bound mRNAs after 0 or 5 Gy irradiation was measured using using Illumina HiSeq4000. Results: RIP-sequencing reveals HuR binding RNAs between 0 or 5 Gy irradiation in WS1 cells. RNA sequencing reveals dyeregulated RNAs between 0 and 5 Gy irradiation. Conclusions: Our study represents the first detailed analysis of HuR binding RNAs between 0 and 5 Gy irradiation generated by RNA-seq technology. This study contributes to our understanding of the role of HuR inresponse to radiation.

Transcriptional response of mouse thyroids following in vivo 211At exposure reveals distinct gene expression profiles

Project description:Transcriptomic profiling of normal mouse thyroid tissue following 211At irradiation Astatine-211 (211At) is an alpha particle emitting halogen with almost optimal linear energy transfer (LET) for creating DNA double strand breaks, and is thus proposed for radionuclide therapy when bound to tumor-seeking agents. Un-bound 211At accumulates in the thyroid gland. The concept of basal radiation-induced biological effects in thyroid tissue is to a high degree unknown and is most valuable. Female BALB/c nude mice were i.v. injected with 0.064-42 kBq of 211At resulting in absorbed doses to the thyroid gland of 0.05-32 Gy. Thyroids were removed at 24 h after injection and total RNA was extracted from pooled thyroids and processed in triplicate using Illumina MouseRef-8 Whole-Genome Expression Beadchips. Nexus Expression 2.0 was used for data analysis. Thyroids exposed to 211At revealed distinctive gene expression profiles compared to non-irradiated controls. More genes were affected at low absorbed doses (0.05 and 0.5 Gy) compared to intermediate (1.4 Gy) and high absorbed doses (11 and 32 Gy) and the proportion of dose-specific genes increased with decreased absorbed dose. This result might be the manifestation of increased heterogeneous irradiation with decreasing absorbed dose, indicating a bystander effect. Also, 1.4 Gy often had an opposite regulation compared to the other absorbed doses. No inflammatory effects were seen while 0.05 and 11 Gy affected the immune system. Effects on the cellular response to outer stress and cell cycle regulation and proliferation were seen at 1.4 and 11 Gy. These results indicate that the cellular response to ionizing radiation is complex and differs with absorbed doses.

Gene expression profiles from heart, lung and liver samples of total body irradiated minipigs

Project description:The present work identified gene expression changes in response to total body irradiation (TBI) in heart, lungs, and liver tissue in Gottingen minipigs. Animals were exposed to 1.7 Gy, 1.9 Gy, 2.1 Gy and 2.3 Gy of TBI and were followed for 45 days. Organ samples were collected at the end of 45 days or sooner if the animal displayed morbidity necessitating euthanasia. RNA was extracted and gene expression profiling studied using Agilent arrays.

Gene expression in mouse blood following low dose-rate or acute x-ray exposure

Project description:Radiation biodosimetry can play a critical role in the response to a large-scale radiologic emergency, and gene expression profiles have shown promise for providing biodosimetric information. This study was designed to test if gene expression could be used to distinguish between doses received from acute exposures and more protracted exposures, such as those that would result from fallout. Mice were exposed to whole body X-rays at low dose rate (LDR, 3.09 mGy/min) for 6, 12, or 24 hours (1.1, 2.2, or 4.4 Gy), or to equivalent doses delivered at high dose rate (HDR, 1.03 Gy/min). Global gene expression was measured in their blood 24 h after the start of exposure, and genes with the potential to classify samples by radiation dose and dose rate were identified.

Project description:Gottingen minipigs mirror the physiological radiation response observed in humans and hence make an ideal candidate model for studying radiation biodosimetry for both limited-sized and mass casualty incidents. We examined the whole blood gene expression profiles at day 1, 3 and 7 after total-body irradiation with 1.7, 1.9, 2.1 and 2.3 Gy doses of gamma-rays. Blood taken one day prior to radiation served as control. Six animals per dose group were used The minipigs were monitored for up to 45 days or time to euthanasia necessitated by radiation effects. Survival correlative signatures were identified from the data.
