Post-transcriptional characterization of the vertebrate aging brain sheds light on the origin of protein-transcript decoupling

Project description:Aging is associated with impaired proteostasis and reduced correlation between protein and mRNA levels. The mechanisms underlying such age-dependent alterations remain unclear. Here, we used the short-lived killifish Nothobranchius furzeri to define how aging affects the transcriptome, translatome and proteome of the vertebrate brain. Integrating these analyses with protein subcellular localization, solubility, and posttranslational modifications data we uncover that basic proteins, including ribosomal and RNA-binding proteins, progressively decrease with age. In contrast, abundant and long-lived proteins, including components of the mitochondrial respiratory chain, increase or remain stable despite a decrease in their mRNA levels. Chronic proteasome inhibition can induce some aging signatures in vivo, but it does not recapitulate the age-related protein-transcript decoupling. Instead, we find increased ribosome pausing in the aging brain reprograms the translation landscape accounting for age-dependent proteome alterations independent of transcription. These changes in protein biogenesis likely reduce availability of key protein complexes in older brains.

A stem-deuterostome origin of the vertebrate pharyngeal transcriptional network.

Project description:Hemichordate worms possess ciliated gills on their trunk, and the homology of these structures with the pharyngeal gill slits of chordates has long been a topic of debate in the fields of evolutionary biology and comparative anatomy. Here, we show conservation of transcription factor expression between the developing pharyngeal gill pores of the hemichordate Saccoglossus kowalevskii and the pharyngeal gill slit precursors (i.e. pharyngeal endodermal outpockets) of vertebrates. Transcription factors that are expressed in the pharyngeal endoderm, ectoderm and mesenchyme of vertebrates are expressed exclusively in the pharyngeal endoderm of S. kowalevskii. The pharyngeal arches and tongue bars of S. kowalevskii lack Tbx1-expressing mesoderm, and are supported solely by an acellular collagenous endoskeleton and by compartments of the trunk coelom. Our findings suggest that hemichordate and vertebrate gills are homologous as simple endodermal outpockets from the foregut, and that much vertebrate pharyngeal complexity arose coincident with the incorporation of cranial paraxial mesoderm and neural crest-derived mesenchyme within pharyngeal arches along the chordate and vertebrate stems, respectively.

Origin of Positive Aging in Quantum-Dot Light-Emitting Diodes.

Project description:The phenomenon of positive aging, i.e., efficiency increased with time, is observed in quantum-dot light-emitting diodes (QLEDs). For example, the external quantum efficiency (EQE) of blue QLEDs is significantly improved from 4.93% to 12.97% after storage for 8 d. The origin of such positive aging is thoroughly investigated. The finding indicates that the interfacial reaction between Al cathode and ZnMgO electron transport layer accounts for such improvement. During shelf-aging, the Al slowly reacts with the oxygen from ZnMgO, and consequently, leads to the formation of AlO x and the production of oxygen vacancies in ZnMgO. The AlO x interlayer reduces the electron injection barrier while the oxygen vacancies increase the conductivity of ZnMgO and, as a result, the electron injection is effectively enhanced. Moreover, the AlO x can effectively suppress the quenching of excitons by metal electrode. Due to the enhancement of electron injection and suppression of exciton quenching, the aged blue, green, and red QLEDs exhibit a 2.6-, 1.3-, and 1.25-fold efficiency improvement, respectively. The studies disclose the origin of positive aging and provide a new insight into the exciton quenching mechanisms, which would be useful for further constructing efficient QLED devices.

The Origin of Vertebrate Gills.

Project description:Pharyngeal gills are a fundamental feature of the vertebrate body plan [1]. However, the evolutionary history of vertebrate gills has been the subject of a long-standing controversy [2-8]. It is thought that gills evolved independently in cyclostomes (jawless vertebrates-lampreys and hagfish) and gnathostomes (jawed vertebrates-cartilaginous and bony fishes), based on their distinct embryonic origins: the gills of cyclostomes derive from endoderm [9-12], while gnathostome gills were classically thought to derive from ectoderm [10, 13]. Here, we demonstrate by cell lineage tracing that the gills of a cartilaginous fish, the little skate (Leucoraja erinacea), are in fact endodermally derived. This finding supports the homology of gills in cyclostomes and gnathostomes, and a single origin of pharyngeal gills prior to the divergence of these two ancient vertebrate lineages.

Transcriptome analyses reveal key features of mouse seminal vesicle during aging

Project description:Despite the dramatic changes in morphology occurring in the seminal vesicles during aging, the transcriptomic characteristics are poorly understood. Thus, we performed a bulk RNA sequencing on seminal vesicle samples from 3-month-old, 13-month-old, and 21-month-old mice to elucidate transcriptomic changes in this study. Comparing 3-month-old mice with the 21-month-old, inflammatory and immune response were identified as the most significantly biological process in upregulated differentially expressed genes (DEGs). Meanwhile, “chemokine signaling pathway” was identified as a key pathway. Indeed, a series of DEGs were enriched in “chemokine signaling pathway”, including Ccl6, Ccl8, Ccl9, Ccr2, Ccr5 and Ccr7. Hub genes such as Ptprc, Adgre1, H2-Ab1 and Ctss were obtained through Protein-Protein interaction network analysis among the continuously up-regulated differentially expressed genes in mouse seminal vesicles at three different ages. These analyses revealed an increased status of immune and inflammatory response in the aged seminal vesicles. In addition, multiple aging-associated DEGs were identified, including Vcam1, Apoe and Cdkn1a. To our knowledge, this study is the first to investigate the general transcriptome landscape of the seminal vesicles in a murine model of natural aging, which may provide novel insights involved in aging-related seminal vesicle dysfunction.
