Autophagy collaborates with apoptosis pathways to control oligodendrocyte number

ABSTRACT: The samples are in vitro cultured oligodendrocyte precursor cells in differentiation medium for 3 days. There are 2 groups: D3HET (ATG5F/+ CNP-Cre, TMT channels 126, 127, 128) and D3KO (ATG5F/F; CNP-Cre, TMT channels 129, 130, 131), with each group comprising 3 biological replicates. Subsequently, all the samples were lysed and subjected to TMT (Tandem Mass Tag) labeling, high pH fractionation into 8 fractions (F1-F8).

Phospho- and total- proteomic analysis of FAK knockout in primary mouse lung endothelial cells

Project description:We assessed the impact of Endothelial Focal Adhesion kinase (ECFAK) knockout, using primary lung endothelial cells isolated (at P10-P14) from a FAK flox/flox mice crossed with mice expressing a tamoxifen-inducible Cre(iCreERT2) that is driven by endothelial cell (EC)-selective platelet-derived growth factor subunit B (Pdgfb) promoter (Pdgfb-iCreERT2) (Tavora et al., 2010). To induce EC restricted FAK deletion, we injected 4-hydroxytamoxifen (4-OHT) from postnatal day 1 (P1) to P2. Molecular signaling events modulated by ECFAK loss, with or without ex vivo VEGF stimulation, were then examined by phospho and total quantitative proteomics analysis using 6plex isobaric tandem mass tagging multiplexing (TMT). In total, three experiments were performed (a an initial pilot experiment not included in this study, followed by two main biological replicates included - i.e. experiments 2, and 3). Low abundance of the matrix arm of complex I in the mitochondria predicts longevity

Project description:The mouse liver mitochondrial proteome was analysed in four different mouse groups (allocation of the samples to Exp1 and Exp2 in brackets): AL (ad libitum fed) C57Bl6 (Exp1 129, Exp1 130, Exp2 130), DR (dietary restriction) C57Bl6 (Exp1 126, Exp1 127, Exp1 128), AL ICRFa (Exp2 128, Exp2 129) and old ICRFa (Exp2 126, Exp2 127). To achieve quantitative standardisation between Exp1 and Exp2, all 10 samples were pooled and one portion of the pool was anlaysed in each Exp (Exp1 131 and Exp2 131). Following tryptic digest and TMT 6-plex labelling, samples were separated by OffGel electrophoresis and all fractions were subsequently analysed by LCMSMS on an Orbitrap LTQ XL. One survey scan (res. 30,000) was followed by a high energy HCD scan (res. 6000) and a low energy CID scan in the LTQ. HCD data were used for the quantitation and CID data for the identification of peptides. Data were searched using Mascot (v. 2.2) through the Proteome Discoverer interface. Peptide raw quantitations were extracted as text files and further processed using dpeaqms (http://r-forge.r-project.org/projects/dpeaqms/) to obtain probability values for the differences in protein amounts for specific proteins between the different animal groups.

Impact of oligodendrocyte secreted factors on hippocampal GABAergic neurons: an electrophysiological and transcriptomic study

Project description:In the present work, we gain insight into the impact of oligodendrocyte secreted factors on hippocampal GABAergic neuron physiology, and performed patch-clamp recordings followed by single-cell RNA sequencing of neurons. Our results demonstrate that the GABAergic neuron excitability is influenced by glial cells and oligodendrocyte-secreted factors. Moreover, we show specific ion channels alterations and find a correlation with electrophysiological parameters suggesting possible mechanisms for the OCM-induced regulation of neuronal function.

Investigation of Oligodendrocyte Precursor Cell Differentiation by Quantitative Proteomics

Project description:Oligodendrocytes, the myelinating cells of the central nervous system, are essential for correct brain function. They originate from oligodendrocyte precursor cells (OPCs) through a differentiation process which is only incompletely understood and impaired in a variety of demyelinating diseases. Better knowledge of this differentiation holds the promise to develop novel therapies for these disorders. We investigated the differentiation of rat oligodendrocyte precursor cells to oligodendrocytes in vitro. After confirmation of differentiation by immunohistochemical analysis using cell type-specific marker proteins, we conducted a quantitative proteomics study using tandem mass tags (TMT). We investigated four time points of differentiation covering early, intermediate, and late stages. Data analysis was perfomed using Mascot and Maxquant.
