Alkyne-probe pull-down in LAPC4-CR cells

ABSTRACT: LAPC4-CR cells were treated with alkyne-probe or DMSO for 2 h in triplicate, lysed and conjugated with biotin-picolyl-azide. Biotinylated proteins were pulled down with neutravidin agarose. After the pulldown, the neutravidin agarose beads were treated with trypsin, and the tryptic peptides were labelled with TMT6 and analyzed by tandem mass spectrometry. DMSO = Reporter 126-128, alkyne-probe = Reporter 129-131. Two replicate injections.
