Project description:Social caste determination in the honey bee is assumed to be determined by the dietary status of the young larvae and translated into physiological and epigenetic changes through nutrient-sensing pathways. We have employed Illumina/Solexa sequencing to examine the small RNA content in the bee larval food source, and show that worker jelly is enriched in miRNA complexity and abundance relative to royal jelly. The miRNA levels in worker jelly were 7-215 fold higher than in royal jelly, and both jellies showed dynamic changes in miRNA content during the 4th to 6th day of larval development. Adding specific miRNAs to royal jelly elicited significant changes in queen larval mRNA expression and in morphological characters of the emerging adult queen bee. We propose that miRNAs in the nurse bee secretions constitute an additional element in the regulatory control of caste determination in the honey bee.

After total RNA was extractedM-BM- and quantified , relative equal amounts of total RNAs from each of the three sampling days were pooled into respectively worker and royal jelly samples, and the fraction of small RNAs less than 30nt long was retained and sequenced on the Illumina/Solexa high-throughput platform (HTP).

Project description:Using artificial Parafilm™ dummies, we explored the nutrition of V. destructor females and assessed their survival when fed on hemolymph from bee larvae, pupae or adults. We compared the results to fully synthetic solutions and to filtered larval plasma. The results showed that the parasites could survive for several days or weeks on our different diets. Hemolymph from larva yielded the highest survival rates and filtered cell-free hemolymph was sufficient to maintain the mites for 14 days or more. This cell free solution thus theoretically contains all necessary nutrients for the mite survival. A proteomic analysis on the larval plasma outlined a list of 54 proteins among which several energy metabolism proteins such as vitellogenin, hexamerin or transferrin. These molecules represent key candidate nutrients that could be crucial for V. destructor survival.

Using digital gene expression, we determined the changes in gene expression induced by pollen intake in worker bees parasitized or not by the mites Varroa destructor, known for suppressing immunity and decreasing lifespan of bees.

Project description:Background: Honey bee is a major insect used for pollination of a number of commercial crops worldwide. However, the number of managed honey bee colonies has recently declined in several countries, and a number of possible causes are proposed. Although the use of honey bees for pollination can be considered as disruption of the habitat, its effects on honey bees' physiology have never been addressed. In Japan, more than 100 thousands colonies are annually used for pollination, and intriguingly 80% of them are used in greenhouses. Recently, honey bee colonies have often collapsed when they are introduced into greenhouses. Thus, to suppress colony collapses and maintain the number of worker bees in the colonies are essential for successful long-term pollination in greenhouses and recycling honey bee colonies.

The purpose is to test whether behavioral phenotype can be predicted by expression profiles in individual brains in a naturalistic context (i.e., colonies in the field). The two behavioral phenotypes examined are 'nurse' and 'forager'. Project description:Adult honey bee queens and workers drastically differ in ovary size. This adult ovary phenotype difference becomes established during the last two larval instars, when massive programmed cell death in the ovaries of worker larvae leads to the degeneration and removal of 95-99% of the ovariole anlagen. The higher juvenile hormone (JH) levels in queen larvae protect their ovaries against such degeneration. To gain insights into the molecular architecture underlying this divergence critical for adult caste fate we performed a microarray analysis contrasting RNA extracts from fourth and early fifth instar queen and worker ovaries. While for the fourth instar we found differential expression (log2FC > 1.0) for only nine genes, the number of differentially represented transcripts (DRTs) increased to 56 in early fifth instar ovaries. From these, 18 had their expression levels further analyzed by real-time PCR (RT-qPCR). For 13 of these the expression differed significantly between queen and worker ovaries at least one time point in development, and interestingly, genes with enzyme functions were overexpressed in workers, while genes related to transcription and signaling were so in queens. For the RT-qPCR confirmed genes we further analyzed their response to JH, revealing a significant up-regulation for two genes, one encoding a short chain dehydrogenase (SDR) and the other heat shock protein 90 (Hsp90). Five other genes, including Hsp60 and hexamerin 70b, were significantly down-regulated by JH. As SDR genes have previously come up as differentially expressed in different transcriptome assays in honey bee larvae, and heat shock proteins are involved in hormone responses, these are interesting candidates for further functional assays.

Project description:Complex organisms are composed of organs and tissues which evolved to their present state as they function together to improve an organism's overall reproductive fitness. Studies of individual organs help us understand their basic functions but this reductionist approach misses the larger context of the whole organism. This problem can be circumvented if all the organs in an organism were comprehensively studied by the same methodology and analyzed together. Using honey bees (Apis mellifera L.) as a model system,we report here the first whole proteome of a complex organism,measuring 29 different tissue types among the three honey bee castes: queen,drone,and worker. The data reveal that,e.g.,workers have a heightened capacity to deal with environmental toxins and queens have a far more robust immune system than their nestmates. Most intriguingly,our analysis reveals the path by which organs of complex organisms probably evolved.Raw data were processed by MaxQuant (v1.2.0.13) using default parameters, plus lysine and N-terminal dimethylation for three isotopologues for relative quantitation. Data were searched against the Apis Official Gene Set version 2 with common contaminants, containing a total of 22037 sequences. Normalized intensity ratios were used for relative quantitation.
