Dietary lipid is largely deposited in skin and rapidly affects insulating properties

ABSTRACT: Dietary fat is rapidly deposited in skin resulting in change in skin thermal properties. Skin was found to be the largest target of deposition of dietary fat. Lipidomic analysis was performed on skin, white adipose tissue, serum, and hair.

Project description:Adipose tissue plays an important role in storing excess nutrients and preventing ectopic lipid accumulation in other organs. Obesity leads to excess lipid storage in adipocytes, resulting in the generation of stress signals and the derangement of metabolic functions. SIRT1 is an important regulatory sensor of nutrient availability in many metabolic tissues. Here we report that SIRT1 functions in adipose tissue to protect from the development of inflammation and obesity under normal feeding conditions, and the progression to metabolic dysfunction under dietary stress. Genetic ablation of SIRT1 from adipose tissue leads to gene expression changes that highly overlap with changes induced by high fat diet in wild type mice, suggesting that dietary stress signals inhibit the activity of SIRT1. Indeed, we show that high fat diet induces the cleavage of SIRT1 in adipose tissue by the inflammation-activated caspase-1, providing a link between dietary stress and predisposition to metabolic dysfunction. FGF18 affects hair cycle.

Project description:Fgf18 gene is strongly expressed in hair follicles of mouse dorsal skin during regressing (catagen) and resting (telogen) phases of hair cycle, but not in growth (anagen) phase. This study aims at identifying the effects of FGF18 local delivery on the anagen phase of hair cycle. To define genes affected by local delivery of FGF18 during anagen phase of hair cycle, we injected FGF18 protein subcutaneously into back skin of C3H/HeN mice on day 4 of depilation-induced anagen. As control PBS was injected in place of FGF18. After 24 h (61-d-old), total RNA was isolated from the back skin and purified to poly A RNA. The RNA samples were pooled for each group. Gene expression was analyzed by one-color analysis using single array for each group.

The impact of myriocin and dietary serine restriction on paw skin sphingolipid diversity

Project description:We analyzed paw skin polar and lipid metabolites in mice (C57BL/6J) in response to two dietary interventions. The first was: 1) low fat (LFD), 2) serine/glycine-free LFD (-SG LFD), high fat (HFD), and serine/glycine-free HFD (-SG HFD). And the second was: 1) low fat (LFD), high fat (HFD), serine/glycine-free HFD (-SG HFD), and serine/glycine-free HFD (-SG HFD) with myriocin (0.3 mg/kg every other day) treatment. The goal of the study was to determine the impact of dietary serine/glycine restriction and myriocin treatment on the hepatic lipidome.

Project description:The effect of dietary calcium and dairy proteins on adipose tissue gene expression profile in diet induced obesity Experiment Overall Design: 9-week-old C57Bl/6J-mice were divided into two groups (n=10/group). The control diet was a standard high-fat diet (60% of energy) low in calcium (0.4%). The whey protein diet was a high-calcium (1.8%) high-fat diet with whey protein isolate. After the 21-week treatment, the adipose tissue transcript profiling (2 mice/group) was carried out using Affymetrix Mouse Genome 430 2.0 array.

Interactions between host and gut microbiota following dietary interventions

Project description:Increasing the consumption of dietary fibre has been proposed to alleviate the progression of non-communicable diseases such as obesity, type 2 diabetes and cardiovascular disease, yet the effect of dietary fibre on host physiology remains unclear. In this study, we performed a multiple diet feeding study in C57BL/6J mice to compare high fat and high fat modified with dietary fibre diets on host physiology and gut homeostasis by combining proteomic, metagenomic, metabolomic and glycomic techniques with correlation network analysis. We observed significant changes in physiology, liver proteome, gut microbiota and SCFA production in response to high fat diet. Dietary fibre modification did not reverse these changes but was associated with specific changes in the gut microbiota, liver proteome, SCFA production and colonic mucin glycosylation. Furthermore, correlation network analysis identified gut bacterial-glycan associations.

Acetyl-CoA metabolism determines the partitioning of lipids between adipose tissue and skin [RNA-seq 1]

Project description:ATP-citrate lyase (ACLY) generates cytosolic acetyl-CoA for lipid synthesis and is a promising therapeutic target in disease contexts with altered lipid metabolism. Here, we developed inducible whole-body Acly knockout mice to determine the requirement for ACLY in normal tissue functions, uncovering its crucial role in skin homeostasis and barrier function. ACLY-deficient skin upregulates the acetyl-CoA synthetase ACSS2; and skin-specific double knockout (DKO) of Acly and Acss2 exacerbates skin abnormalities, with differential effects on two major lipid-producing skin compartments. While the epidermis is depleted of key barrier lipids, the sebaceous glands unexpectedly increase their lipid-rich sebum secretion, supplied in part by fatty acids released from adipose tissues. Accordingly, DKO mice rapidly lose fat mass, and dietary lipid supplementation partially rescues both the lipoatrophy and aberrant skin phenotypes. Thymic stromal lymphopoietin (TSLP), a cytokine with a role in regulating sebaceous glands, is overexpressed in DKO epidermis and thus may stimulate sebum secretion in response to deficient epidermal acetyl-CoA synthesis. Together, the data demonstrate that cytosolic acetyl-CoA synthesis in the skin both maintains local tissue homeostasis and influences systemic lipid partitioning and storage.
