Project description:Extracellular vesicles (EVs) can mediate intercellular communication by transferring cargo proteins and nucleic acids between cells. The pathophysiological roles and clinical value of EVs are under intense investigation, yet most studies are limited by technical challenges in the isolation of nanoscale EVs (nEVs). Here, we report a lipid nanoprobe that enables spontaneous labelling and magnetic enrichment of nEVs in 15 minutes, with isolation efficiency and cargo composition similar to what can be achieved by the much slower and bulkier method of ultracentrifugation. We also show that the lipid nanoprobes, which allow for downstream analyses of nucleic acids and proteins, enabled the identification of EGFR and KRAS mutations following nEV isolation from blood plasma from non-small-cell lung-cancer patients. The efficiency and versatility of the lipid nanoprobe opens up opportunities in point-of-care cancer diagnostics.

Project description:Extracellular vesicles (EVs) have been identified as promising biomarkers for the noninvasive diagnosis of various diseases. However, challenges in separating EVs from soluble proteins have resulted in variable EV recovery rates and low purities. Here, we report a high-yield ( > 90%) and rapid ( < 10 min) EV isolation method called FLocculation via Orbital Acoustic Trapping (FLOAT). The FLOAT approach utilizes an acoustofluidic droplet centrifuge to rotate and controllably heat liquid droplets. By adding a thermoresponsive polymer flocculant, nanoparticles as small as 20 nm can be rapidly and selectively concentrated at the center of the droplet. We demonstrate the ability of FLOAT to separate urinary EVs from the highly abundant Tamm-Horsfall protein, addressing a significant obstacle in the development of EV-based liquid biopsies. Due to its high-yield nature, FLOAT reduces biofluid starting volume requirements by a factor of 100 (from 20 mL to 200 µL), demonstrating its promising potential in point-of-care diagnostics.

Project description:Extracellular vesicles (EVs) have found diverse applications in clinical theranostics. However, the current techniques to isolate plasma EVs suffer from burdensome procedures and limited yield. Herein, we report a rapid and efficient EV isolation platform, namely, EV-FISHER, constructed from the metal-organic framework featuring cleavable lipid probes (PO4 3- -spacer-DNA-cholesterol, PSDC). The EV-FISHER baits EVs from plasma by cholesterol and separates them with an ordinary centrifuge. The captured EVs could be released and collected upon subsequent cleavage of PSDC by deoxyribonuclease I. We conclude that EV-FISHER dramatically outperforms the ultracentrifugation (UC) in terms of time (∼40 min vs. 240 min), isolation efficiency (74.2% vs. 18.1%), and isolation requirement (12,800 g vs. 135,000 g). In addition to the stable performance in plasma, EV-FISHER also exhibited excellent compatibility with downstream single-EV flow cytometry, enabling the identification of glypican-1 (GPC-1) EVs for early diagnosis, clinical stages differentiation, and therapeutic efficacy evaluation in breast cancer cohorts. This work portrays an efficient strategy to isolate EVs from complicated biological fluids with promising potential to facilitate EVs-based theranostics.

Project description:Background and study aims
Colorectal cancer is the third most common cancer in men and the second in women, accounting for 10% of all cancers worldwide. It ranks second in terms of cancer-related deaths, just behind lung cancer. In certain European member states, a home-based screening test searching for occult blood in a stool sample has been in operation in recent years for people aged 50 years and over. Although this method is simple to perform, it only detects the already symptomatic stage of the disease. Therefore, although more invasive, colonoscopy remains the most reliable method of screening for colorectal cancer, as it enables polyps and other lesions to be visualized and removed using an endoscope with a camera. The risk of colorectal cancer following colonoscopy has been shown to be reduced by 70-90%. Early detection and removal of a pre-cancerous polyp prevents its progression to a tumor. In this way, colonoscopy saves many lives. Nevertheless, although colorectal cancer is now considered an easily preventable disease thanks to screening, long waiting and preparation times for colonoscopy prevent the implementation of large-scale screening for systematic surveillance and follow-up. Since the 1990s, there has been a gradual increase in the rate of colorectal cancer in adults under the age of 50. Although the reasons for this are still unknown, it has been suggested that environmental and behavioral changes influencing the microbiome (gut bacteria) are at the root of colorectal cancer in people under 50. The need to develop a large-scale, inexpensive, and non-invasive method of early detection of colorectal cancer is therefore urgent.
This study aims to develop a routine blood test accessible to all ages in order to identify people who would not otherwise be screened according to current European or national guidelines. The previous part of the DIOPTRA project would have identified in around 200 participants a protein group whose quantity varies during a precancerous stage of colon cancer. By quantifying this group of proteins, this blood test will be able to identify those citizens who absolutely should undergo further screening by colonoscopy. To validate this method, participants are invited to give a blood sample during their colonoscopy visit to the gastroenterology department. Once validated, this blood test has many advantages: it is almost non-invasive, inexpensive and could be well accepted by most of the population. As a result, DIOPTRA is positioning itself in the increasingly personalized medicine of the future, capable of adapting to the particularities of each individual.
Other aims:
1. In addition to an early detection method for colon cancer, numerous scientific studies have identified parameters called risk factors which could be associated with the development of colorectal cancer, and their importance is not negligible. Suggestions for daily habits can be generated from these risk factors and may be very useful in the prevention of colorectal cancer.
2. Another aim of the study is to validate some of the risk factors identified in the previous part of the project. The DIOPTRA application would be created to help collect information, offer up-to-date personalized suggestions and raise awareness of early detection of colorectal cancer.
3. The findings and the final DIOPTRA solution will be the subject of a study of healthcare performance indicators in view of widening screening eligibility thanks to an effective, minimally invasive and financially affordable method.
Who can participate?
Individuals between 18 and 80 years old who visit the clinical sites for a colonoscopy
What does the study involve?
According to each clinical site’s standards and pre-existing practice, enrolled individuals will undergo a screening colonoscopy, while blood will be collected via a minimally invasive method (before the colonoscopy) for the purposes of the study. All biological data will be used for protein-based analysis, allowing the construction of preliminary decision algorithms and AI analysis models.

Project description:Cancer cell-derived extracellular vesicles (EVs) are promising biomarkers for cancer diagnosis and prognosis. However, the lack of rapid and sensitive isolation techniques to obtain EVs from clinical samples at a sufficiently high yield limits their practicability. Chimeric nanocomposites of lactoferrin conjugated 2,2-bis(methylol)propionic acid dendrimer-modified magnetic nanoparticles (LF-bis-MPA-MNPs) are fabricated and used for simple and sensitive EV isolation from various biological samples via a combination of electrostatic interaction, physically absorption, and biorecognition between the surfaces of the EVs and the LF-bis-MPA-MNPs. The speed, efficiency, recovery rate, and purity of EV isolation by the LF-bis-MPA-MNPs are superior to those obtained by using established methods. The relative expressions of exosomal microRNAs (miRNAs) from isolated EVs in cancerous cell-derived exosomes are verified as significantly higher than those from noncancerous ones. Finally, the chimeric nanocomposites are used to assess urinary exosomal miRNAs from urine specimens from 20 prostate cancer (PCa), 10 benign prostatic hyperplasia (BPH), patients and 10 healthy controls. Significant up-regulation of miR-21 and miR-346 and down-regulation of miR-23a and miR-122-5p occurs in both groups compared to healthy controls. LF-bis-MPA-MNPs provide a rapid, simple, and high yield method for human excreta analysis in clinical applications.

Project description:A cell's ability to secrete extracellular vesicles (EVs) for communication is present in all three domains of life. Notably, Gram-negative bacteria produce a specific type of EVs called outer membrane vesicles (OMVs). We previously observed the presence of OMVs in human blood, which could represent a means of communication from the microbiota to the host. Here, in order to investigate the possible translocation of OMVs from the intestine to other organs, the mouse was used as an animal model after OMVs administration. To achieve this, we first optimized the signal of OMVs containing the fluorescent protein miRFP713 associated with the outer membrane anchoring peptide OmpA by adding biliverdin, a fluorescence cofactor, to the cultures. The miRFP713-expressing OMVs produced in E. coli REL606 strain were then characterized according to their diameter and protein composition. Native- and miRFP713-expressing OMVs were found to produce homogenous populations of vesicles. Finally, in vivo and ex vivo fluorescence imaging was used to monitor the distribution of miRFP713-OMVs in mice in various organs whether by intravenous injection or oral gavage. The relative stability of the fluorescence signals up to 3 days post-injection/gavage paves the way to future studies investigating the OMV-based communication established between the different microbiotas and their host.

Project description:Extracellular vesicles (EVs) play a pivotal role in various biological pathways, such as immune responses and the progression of diseases, including cancer. However, it is challenging to isolate EVs at high purity from blood plasma and other biofluids due to their low abundance compared to more predominant biomolecular species such as lipoprotein particles and free protein complexes. Ultracentrifugation-based EV isolation, the current gold standard technique, cannot overcome this challenge due to the similar densities of such species. We developed several novel approaches to enrich EVs from plasma while depleting contaminating molecular species using multimode chromatography-based strategies. On average, we identified 716±68 proteins and 1054±35 proteins in EV isolates from 100 µL of plasma using multimode chromatography- and ultracentrifugation-based techniques, respectively. The developed methods resulted in similar EV isolates purity, providing significant advantages in simplicity, throughput, scalability, and applicability for various downstream analytical and potential clinical applications

Project description:Small extracellular vesicles (sEVs; <200 nm) that contain lipids, nucleic acids, and proteins are considered promising biomarkers for a wide variety of diseases. Conventional methods for sEV isolation from blood are incompatible with routine clinical workflows, significantly hampering the utilization of blood-derived sEVs in clinical settings. Here, we present a simple, viscoelastic-based microfluidic platform for label-free isolation of sEVs from human blood. The separation performance of the device is assessed by isolating fluorescent sEVs from whole blood, demonstrating purities and recovery rates of over 97 and 87%, respectively. Significantly, our viscoelastic-based microfluidic method also provides for a remarkable increase in sEV yield compared to gold-standard ultracentrifugation, with proteomic profiles of blood-derived sEVs purified by both methods showing similar protein compositions. To demonstrate the clinical utility of the approach, we isolate sEVs from blood samples of 20 patients with cancer and 20 healthy donors, demonstrating that elevated sEV concentrations can be observed in blood derived from patients with cancer.
