High-resolution lipidomics for decoding the soil biome: Improved lipid annotation, quantitation, and response to climate stress


ABSTRACT: total lipid extracts in solvent mixture containing acetonitrile, water, and isopropanol (ACN:H2O:IPA, 5:4:1, v/v/v). Seperation using UHPLC Ultimate 3000 system with 5 uL of sample injection onto an Accucore C30 column (2.1 mm * 150 mm, 2.6-um particle size) maintained at 40C. flow rate was set to 300 ul min-1. ACN:H2O (60:40, v/v) and ACN:IPA (10:90, v/v) were used as mobile phase A and mobile phase B, respectively. Both eluents were supplemented with 0.1% formic acid and 10 mM ammonium formate.
