Junctophilin 2 (JPH2) and ryanodine receptor 2 (RYR2) protein complex in WT and FKBP12 deficient mice heart

ABSTRACT: To determine if the absence of FKBP12 in cardiac tissue alters JPH2 and RYR2 interactions with other dyadic proteins, we immunoprecipitated JPH2 from hearts of FL, MCK-FKD, MHC-Cre+ only, and MHC-FKD mice.

Specific proteins were identified in the IPs from the FL mice and included JPH2, CASQ2 , SPEG, RYR2, CACNA1c, CACNB2, ESD, CMYA5, and LRRC10.

Effect of cardiac specific depletion of one allele of Mrps5 on gene expression in heart at postnatal day 4

Project description:To investigate the function of reduction in Mrps5 expression in the regulation of cardiomyocyte proliferation and postnatal heart development, we established the mouse with cardiac specific depletion of one allele of Mrps5 through Cre-Loxp system by crossing Mrps5 fl/fl mouse with Myh6-Cre mouse. We cross Mrps5 fl/fl mouse with Myh6-Cre mouse to obtain the Mrps5 fl/+;Myh6-Cre mouse and its control littermates Mrps5 fl/+ mouse. So that one allele of Mrps5 gene was depleted upon Myh6 gene promoter drivened Cre expression at the later stage of embyronic stage. We then performed gene expression profiling analysis using data obtained from RNA-seq of Mrps5 fl/+; Myh6-Cre and Mrps5 fl/+ mouse hearts at postnatal day 4.

Transcription profiling of mouse heart from cardiac-specific conditional knockout deletion of menage-a-trios-1 (MAT1)

Project description:The Cdk7/cyclin H/menage-a-trois 1 (MAT1) heterotrimer has proposed functions in transcription as the kinase component of basal transcription factor TFIIH and is activated in adult hearts by hypertrophic pathways. Using cardiac-specific Cre, we ablated MAT1 in myocardium. Despite reduced Cdk7 activity, MAT1-deficient hearts grew normally. However, fatal heart failure ensued at 6-8 weeks. By microarray profiling, quantitative RT-PCR, and Western blotting at 4 weeks, genes for energy metabolism were found to be suppressed selectively, including targets of peroxisome proliferator-activated receptor-gamma coactivator-1 (PGC-1). Cardiac metabolic defects were substantiated in isolated perfused hearts and isolated mitochondria. In culture, deleting MAT1 with Cre disrupted PGC-1 function: PGC-1Î± failed to activate PGC-1-responsive promoters and nuclear receptors, GAL4-PGC-1Î± was functionally defective, and PGC-1Î² likewise was deficient. PGC-1 was shown to interact with MAT1 and Cdk7, in co-precipitation assays. Thus, we demonstrate an unforeseen essential role for MAT1 in operation of the PGC-1 family of co-activators. Experiment Overall Design: MAT1F/F mice (Korsisaari et al., 2002) were bred with mice expressing Cre recombinase under the control of the cardiomyocyte-specific Î±-myosin heavy chain (Î±MHC) promoter (Gaussin et al., 2002a) and back-bred to MAT1F/F mice to generate the cardiac-specific knockout (Î±MHC-Cre+/0; MAT1F/F; CKO; Fig. 1A, B). Control mice were Î±MHC-Cre+/0; MAT1F/+ littermates, differing by the presence of one wild-type MAT1 allele, and excluding Cre-mediated toxicity as a basis for phenotypic disparity. Effects of increased YY2 expression on heart development

Project description:Yin Yang 2 (YY2) is a member of the Yin Yang family of transcription factors. Although the bioactivity of YY2 has been previously studied, its role in cardiovascular diseases is not known. We observed the increased expression of YY2 in failing human hearts compared with control hearts, raising the question of whether YY2 is involved in the pathogenesis of cardiomyopathy. To investigate the potential contribution of YY2 to the development of cardiomyopathy, we crossed two transgenic (Tg) mouse lines, pCAG-YY2-Tg+ and -MHC-Cre, to generate two independent double transgenic (dTg) mouse lines in which the conditional cardiomyocyte-specific expression of YY2 driven by the -MHC promoter was mediated by Cre recombinase, starting at embryonic day 9.0. In dTg mice, we observed partial embryonic lethality and hearts with defective cardiomyocyte proliferation. Surviving dTg mice from both lines developed cardiomyopathy and heart failure that occurred with aging, showing different degrees of severity that were associated with the level of transgene expression. The development of cardiomyopathy was accompanied by increased levels of cardiac disease markers, apoptosis, and cardiac fibrosis. Our studies further revealed that the Cre-mediated cardiomyocyte-specific increase in YY2 expression led to increased levels of Beclin 1 and LC3II, indicating that YY2 is involved in mediating autophagic activity in mouse hearts in vivo. Also, compared with control hearts, dTG mouse hearts showed increased JNK activity. Because autophagy and JNK activity are important for maintaining cardiac homeostasis, the dysregulation of these signaling pathways may contribute to YY2-induced cardiomyopathy and heart failure in vivo.
