ABSTRACT: To define the mitochondrial complexes interacting with MICU proteins we evaluated the mtCU-independent interactome of MICU1 in a co-expression system with MICU2 and/or MICU3. MICU1-containing MICU permutation plasmids (MICU1-HA, MICU1-HA-T2a-MICU2-Myc, MICU1-HA-T2a-MICU3-Flag, MICU1-HA-T2a-MICU2-Myc-T2a-MICU3-Flag) were expressed in Mcu-/- HEK 293T cells. IPs of MICU1-HA were performed and prepped for liquid chromatography mass spec (LC/MS) to identify interacting proteins

Project description:To investigate and identify that the host proteins interacting with PEDV NSP12, we transfected the p3×Flag-NSP12 and p3×Flag-CMV-10 plasmids into 293T cells, respectively. Then the proteins were immunoprecipitated with anti-Flag antibody. PEDV NSP12 interacting proteins and identified the precipitated proteins by Liquid Chromatography-Mass Spectrometer/Mass Spectrometry (LC-MS/MS).

Project description:To investigate proteins interacting with STING mutants, HEK 293T cells were co-transfected with three STING mutant expression plasmids (human STING WT, STING delCTT (aa343-379 deletion), STING del6 (aa325-379 deletion) in a pcDNA3.1+ backbone) and either pcDNA-cGAS expression plasmid or control empty pcDNA plasmid. Project description:To identify candidate mTOR-interacting molecules, we performed an anti-Flag immunoprecipitation (IP) followed by mass spectrometry (MS)-based proteomic analysis in 293T cells overexpressing Flag-tagged mTOR. We identified P4HA2 binding proteins with anti-HA IP followed by (MS)-based proteomic analysis in 293T cells overexpressing HA-tagged P4HA2. To speculate that mTOR can be hydroxylated by P4HA2. We used 293T cells overexpressing Flag-mTOR, Flag-mTOR/HA-P4HA2, and Flag-mTOR/HA-P4HA2-overexpressing cells treated with EDHB, we conducted an anti-Flag IP to capture and enrich mTOR protein, and the immunoprecipitates were subjected to MS analysis.

Project description:293T were transfected with plasmids expressing DNA-dependent activator of IFN-regulatory factors (DAI) or retinoic acid-inducible gene 1 (RIG-I). Transfected cells were subsequently infected with influenza A/Puerto Rico/8/34 virus for 12 hours. Cell extract was subjected to FLAG Ab beads, washed, and retained. RNA was sequenced using the Illumina TruSeq Stranded Total RNA Library kit. The input sample represents total RNA from vector-infected cells.
