Quorum sensing by Streptococcus pyogenes induces global inflammatory suppression in host macrophages

Project description:Streptococcus pyogenes, or Group A Strep (GAS), a significant human pathogen, employs quorum sensing (QS) systems to coordinate its behavior and genetic regulation in order to enhance survival. Our previous research established that one such QS system, the Rgg2/3 system, has the ability to suppress macrophage NFkB activity and production of pro-inflammatory cytokines. Yet, the scope of suppression was undetermined, and the mechanism remained elusive. In this study, we used transcriptomic and phosphoproteomic approaches to elucidate the extent and mechanisms of QS-mediated immune suppression. We found QS-ON GAS broadly suppressed most inflammatory transcriptional pathways including those of NFkB, type I and type II interferon responses, and intracellular stress responses. Yet, we found no alternative transcriptional programs being activated instead. Additionally, phosphoproteomics showed no disruption upstream in typical inflammatory pathways such as those related to NFkB and MAPK activation, which we successfully confirmed. Instead, the proteomic data highlighted a potential role for epigenetic mechanisms of inflammatory regulation. Despite this, measuring DNA methylation and inhibiting important histone and chromatin modifiers did not reveal which if any epigenetic regulation protein might be involved in the mechanisms of suppression. Still, these findings expand our understanding of QS-mediated suppression and of GAS virulence strategies that appear to target irregular means of restricting inflammation. Uncovering this mechanism will offer invaluable insight into GAS, itself, as well as understudied immunological pathways.

Peptide-based quorum sensing systems in Paenibacillus polymyxa

Project description:Paenibacillus polymyxa is an agriculturally important plant growth promoting rhizobacterium (PGPR). Many Paenibacillus species are known to be engaged in complex bacteria-bacteria and bacteria-host interactions, which in other bacteria were shown to necessitate quorum sensing communication, but to date no quorum sensing systems have been described in Paenibacillus. Here we show that the type strain P. polymyxa ATCC 842 encodes at least 16 peptide-based communication systems. Each of these systems comprises a pro-peptide that is secreted to the growth medium and further processed to generate a mature short peptide. Each peptide has a cognate intracellular receptor of the RRNPP family, and we show that external addition of P. polymyxa communication peptides to the medium leads to reprogramming of the transcriptional response. We found that these quorum sensing systems are conserved across hundreds of species belonging to the Paenibacillaceae family, with some species encoding more than 25 different peptide-receptor pairs, representing a record number of quorum sensing systems encoded in a single genome.

Project description:In the human pathogen Streptococcus mutans, the canonical peptide-based quorum sensing system is an inducible DNA repair system that is pivotal for bacterial survival. Previous work showed that the CSP signaling peptide is a stress-signaling alarmone that controls different stress-induced phenotypes. In this study, we exposed S. mutans to the CSP pheromone to mimic DNA damage conditions. Transcriptome analysis was then performed to evaluate the differential gene expression between the normal stationary phase cells and the CSP-induced stationary phase cells. The data obtained contribute to the understanding of the CSP-induced phenotypes in S. mutans.

A New Class of Homoserine Lactone Quorum Sensing Signals

Project description:Quorum sensing is a term used to describe cell-to-cell communication that allows cell density-dependent gene expression. Many Gram-negative bacteria use acyl-homoserine lactone (acyl-HSL) synthases to generate fatty acyl-HSL quorum sensing signals, which function with signal receptors to control expression of specific genes. The fatty acyl group is derived from fatty acid biosynthesis and provides signal specificity, but the variety of signals is limited. We have discovered that the photosynthetic bacterium Rhodopseudomonas palustris uses an acyl-HSL synthase to produce p-coumaroyl-HSL by using environmental p-coumaric acid rather than fatty acids from cellular pools. The bacterium has a signal receptor with homology to fatty acyl-HSL receptors that responds to p-coumaroyl-HSL to regulate global gene expression. We also found that p-coumaroyl-HSL is made by other bacteria including Bradyrhizobium BTAi1 and Silicibacter pomeroyi DSS-3. This discovery extends the range of possibilities for acyl-HSL quorum sensing and raises fundamental questions about quorum sensing within the context of environmental signaling.
