Dissection of Vitronectin-Regulated Secretome in Hepatocellular Carcinoma

ABSTRACT: Vitronectin is a glycoprotein found in the extracellular matrix. It has potential applications in the diagnosis and treatment of HCC. Secretome analysis identified proteins regulated by vitronectin in HCC, and an evaluation was conducted to determine its suitability for use.

Cellular, molecular and proteomic characteristics of early Hepatocellular carcinoma

Project description:Ηepatocellular carcinoma (HCC) accounts for the majority of primary liver cancers. Early de-tection/diagnosis is vital for the prognosis of HCC whereas diagnosis at late stages is associated with very low survival rate. Early diagnosis is based on patient’s 6 month surveillance and use of at least two imaging modalities. The aim of this study was to investigate diagnostic markers for the detection of early HCC based on proteome analysis, microRNAs (miRNAs) and circulat-ing tumor cells (CTCs) in the blood of patients with cirrhosis, early and advanced HCC. We studied 89 patients with HCC of whom 33 had early HCC and 28 cirrhotic patients. CTCs were detected by Real-Time Quantitative Reverse Transcription PCR and immunofluorescence using the markers Epithelial cell adhesion molecule (EPCAM), Vimentin, A Fetoprotein (AFP) and surface major Vault protein (sMVP). Expression of the five most commonly HCC-involved miRNAs (miR-122, miR-200a, miR-200b, miR-221, miR-222) was examined in the serum by qRT-PCR. Finally patient serum was analyzed by whole proteome analysis (LC/MS). Twenty seven out of 53 (51%) patients with advanced HCC had detectable CTCs. Among them, 10/27 (37%) presented evidence of mesenchymal or intermediate stage cells (vimentin and/or sMVP positive). Moreover, 5/17 (29%) patients with early HCC and 6/28 (21%) cirrhotic patients had detectable CTCs. Patients with early and advanced HCC exhibited a significant increase of miR-200b when compared to cirrhotic patients. Our proteome analysis indicated that early HCC patients present a significant upregulation of APOA2, APOC3 proteins when compared to cir-rhotic patients, that when used in combination, cover the 100% of the patients with early HCC. In conclusion, miR-200b, APOA2 and APOC3 proteins are sensitive markers and can be poten-tially useful in combination for the early diagnosis of HCC.

3-D culture cell lines mimic tumor in hepatocellular carcinoma

Project description:Hepatocellular carcinoma (HCC) is one of the main subtypes of primary liver cancer, which accounts for 75-85% of all cases [1]. Despite increasing availability of treatments for liver cancer, HCC clinical trials are not promising since most HCC patients are in an advanced stage of HCC at the time of diagnosis. Additionally, highly metastatic and recurrent phenotype aggravate poor prognosis of liver cancer [2]. Therefore, there is an urgent need to explore the underlying molecular mechanism in HCC progression.

Quantitative proteome analysis of hepatocellular carcinoma tissue

Project description:Hepatocellular carcinoma (HCC) is one of the most aggressive tumors and the treatment outcome of this disease is improved when the cancer is diagnosed at early stages. This requires biomarkers allowing for an accurate and early tumor diagnosis. To identify potential markers for such applications we performed a label-free proteome analysis using samples collected from 19 patients. We analyzed the data considering events known to take place in early events of HCC development such as abnormal regulation of Wnt/b-catenin and activation of receptor tyrosine kinases (RTKs). We expected that such analysis can lead to the identification of potential biomarkers for early-stage HCC diagnosis. 31 proteins functionally linked to downstream interactors of Wnt/b-catenin and parts of RTKs´-activated pathways Ras and JAK/STAT were selected for verification in a larger and independent cohort (n= 31) using selected (multiple) reaction monitoring (SRM/MRM).

Proteomic analysis of genes regulated by miR-4510 in hepatocellular carcinoma

Project description:Patients with advanced hepatocellular carcinoma (HCC) are currently treated by Sorafenib and Regorafenib but efficacy of these multikinase inhibitors is disappointing. Thus, new drugs and therapies are needed to improve HCC management in clinic. We recently reported the remarkable capacity of miR-4510 to impede the growth of HCC and hepatoblastoma cells in vitro and in vivo through GPC3 targeting and Wnt pathway inactivation (Cartier F et al, Oncotarget 2017). Here, we used a label-free proteomic approach to identify new targets of miR-4510 in HCC-deriving Huh7 cells. We transfected HCC-derived Huh7 cells with either a synthetic miR-4510 mimic or a small RNA control, extracted total proteins 24hr later and comparatively analyzed proteomes of control and miR-4510-transfected cells using a quantitative label-free approach.

Transcriptome analysis in tumor budding positive hepatocellular carcinoma tissue samples relative to tumor budding negative hepatocellular carcinoma tissue samples

Project description:Tumor budding (TB) has become a crucial factor for predicting the malignancy grade and prognostic outcome for multiple solid cancers.Studies have investigated the prognostic value of TB within hepatocellular carcinoma (HCC). However, its molecular mechanism within HCC remains unclear. This is the first study to discover and compare the expression of differentially expressed genes (DEGs) between TB-positive and TB-negative HCC tissues. Total RNA was extracted from 40 fresh HCC tissue samples with different TB statuses and then sequenced. This study provides insights into the possible mechanisms of TB in HCC and revealed potential predictive diagnosis markers and treatment targets for HCC.

Hepatocellular carcinoma cell lines LC-MS/MS

Project description:Hepatocellular carcinoma (HCC) ranks sixth among the most common malignancies and fourth in mortality among all kinds of cancers in the world. Recent advances in early diagnosis and therapeutics have led to improved survival of HCC patients, but the recurrence and metastasis are still main reasons for the poor prognosis and high mortality of HCC. Many researches on the mechanism of HCC deterioration have uncovered the epithelial-mesenchymal transition (EMT) as a major trigger for HCC metastasis and invasion.In this study, in order to systematically investigate the dynamic site-specific glycosylation alterations associated with the EMT process of HCC, two hepatoma cell lines SMMC-7721 and HepG2 were treated using HGF and the cell proteins were harvested at six treatment time points. Using TMT-labeling, solid phase extraction and mass spectrometry, we not only detailly profiled N-glycoproteome of both cell lines at site-specific glycosylation level, but also dynamically quantified those intact glycopeptides among six increasing HGF-treated time points. By dynamically quantifying enriched glycopeptides and proteins among six HGF-treated time points in two cell lines, we identified 20 up-regulated intact glycopeptides during the process of EMT, with the majority changed at the glycosylation level instead of protein expression level. The site-specific glycosylation change of those glycoproteins was related to the process of EMT, which was further verified by a series of molecular biology methods, mass spectrometry, and migration and invasion assay in vitro.

ADAMTSL5 is an epigenetically activated oncogene conferring tumorigenic properties in hepatocellular carcinoma

Project description:The remarkable molecular heterogeneity in hepatocellular carcinoma (HCC) continues to challenge development of effective treatments and biomarkers. Here we report that the gene encoding a secreted glycoprotein, ADAMTSL5, is overexpressed and displays hypermethylated CpG islands in its gene body region in a clinically relevant mouse genetic model of HCC as well as in a significant proportion of human HCC patients. Immunohistological analysis of human HCCs revealed strong ADAMTSL5 immunostaining in tumour cells and histiocytes, in contrast to normal liver. Functional targeting of ADAMTSL5 in oncogenesis using shRNA interfered with tumorigenic properties of HCC cells both in vitro and in vivo. Furthermore, ADAMTSL5 overexpression conferred tumorigenic capability to genetically sensitized, non-transformed hepatocytes in nude mice. Mechanistically, ADAMTSL5 abrogation led to reduction of several oncogenic inputs, including the receptor tyrosine kinases MET, EGFR, PDGFRβ, IGF1Rβ, and FGFR4, suggesting it as a master regulator of HCC tumorigenicity when overexpressed.
