Enterococcal gelatinase mediated cleaving of IL-1beta proform

ABSTRACT: Human IL1beta proform was incubated with Enterococcus faecalis OG1RF for 0, 6 and 18h in brain heart infusion media. IL1beta fragments generated by E. faecalis at 6 and 18h were sent for Mass spectrometry analysis, with the 0-h sample serving as control for the full length IL1beta proform.

Nucleosome mapping data from GR and cell-cycle regulated genes in human cell lines

Project description:Human UL3 cells (U20S derivatives containing GR and an MMTV-luc transgene) were treated with dexamethasone for 0, 1 or 4 hrs. Chromatin was harvested, digested with to ~70% mononucleosomes with MNase, and isolated mononucleosome fragments were used to probe custom Nimblegen genomic tiling microarrays. In addition, human HL60 cells were treated with or without DMSO to induce granulocyte differentiaton, and mononucleosome positions mapped. MNase digested bare DNA fragments of ~500 bp average length from UL3 cells was used as a control.

High salt intake exacerbates aortic dissection through IL-17 pathway

Project description:Rationale— Aortic dissection (AD) is a fatal disease that occurs suddenly without preceding clinical signs or symptoms. It is desirable to prevent AD before the onset. Although high salt intake is an established risk factor for cardiovascular diseases, the relationship between high salt intake and AD has not been clarified. Although recent studies have shown that high salt intake promotes interleukin (IL)-17-dependent inflammation, it is unknown whether this mechanism is involved in the pathogenesis of AD. Objective— The purpose of this study was to investigate the effect of high salt challenge on AD and involvement of IL-17. Methods and Results— We used a mouse AD model that was induced by continuous infusion of -aminopropionitrile and angiotensin II. High salt challenge exacerbated AD lesion length compared to the mice without high salt challenge, which was abolished in IL-17 knockout mice. Unexpectedly, deletion of IL-17 resulted in the activation of Smad pathway, induction of synthetic phenotype of smooth muscle cells, remodeling of extracellular matrix in aortic tunica media, reduced stiffness of aortic walls, and less stress-induced activation of NFkB. Conclusions— These findings establish the worsening effect of high salt intake on AD that involves IL-17 pathway through the extracellular matrix metabolism. Salt restriction may represent a low-cost and practical way to reduce AD risk.

Brief Hydrocortisone Infusion Differentially Influences Blood Leukocyte and Monocyte Gene Expression Before and After Endotoxin Administration in Humans (supported by NIH grant GM34695)

Project description:Fourteen human subjects (18-40 years of age) were studied. The control group (saline + LPS [n = 7]) received a 12 hour infusion of saline and the experimental group (cortisol + LPS [n = 7]) received a 12 hour infusion of cortisol (3 µg/kg/min). After 6 hours of saline or cortisol infusion, intravenous lipopolysaccharide (2 ng/kg) was administered to all subjects. Blood samples were collected at -24 hours, 0 hours, 6 hours and 24 hours relative to LPS administration at 0 hours. Leukocytes and monocytes were isolated from these blood samples, total RNA extracted, and microarray analyses performed using Affymetrix Focus GeneChips®.
