Gene Expression Time Courses of CH999 with High or Low Copy Number Plasmids

Project description:Time courses of Streptomyces CH999/pKOS011-26 and CH999/pKOS011-26* the low and high copy number strains, respectively, to examine gene expression differences between these two strains. Of particular interest were the transcript levels of the activator gene actII-ORF4 and the polyketide synthase gene cluster eryA. Mycelia were grown in 50 mL R5 liquid medium with thiostrepton drug selection at 50 micrograms per mL in a 30 degrees Celsius shaker incubator. mRNA were harvested at the time points indicated. These data show that actII-ORF4 and eryA transcripts were more highly expressed in the high copy number strain than in the low copy number strain. Streptomyces hygroscopicus 5008 Transcriptome or Gene expression

Project description:The RNA sequencing approach was utilized to carry out a comparative transcriptome analysis between Streptomyces hygroscopicus 5008 wild-type and a genetically engineered strain. The A-factor-like cascade play an important role in the regulation of validamycin biosynthesis by Streptomyces hygroscopicus 5008, and the pleiotropic regulator AdpA-H may positively regulate the transcription of gene cluster for the biosynthesis. shbR1 and shbR3 as the A-factor receptor homolog genes, could repress the transcription of AdpA-H. By tandem deletions of these genes, the production and productivity of validamcyin was significantly enhanced. To explore the effects of the shbR1/R3 double deletion of the overall cellular metabolism, the RNA sequencing approach was utilized to carry out a comparative transcriptome analysis between wild-type and shbR1/shbR3 double mutant (genetically engineered strain).

Production of chain-extended cinnamoyl compounds by overexpressing two adjacent cluster-situated LuxR regulators in Streptomyces globisporus C-1027

Project description:Natural products from microorganisms are important sources for drug discovery. With the development of high-throughput sequencing technology and bioinformatics, a large amount of uncharacterized biosynthetic gene clusters (BGCs) in microorganisms have been found, which show the potential for novel natural product production. 9 BGCs containing PKS and/or NRPS in Streptomyces globisporus C-1027 were transcriptionally low/silent under the experimental fermentation conditions, and the products of these clusters are unknown. Thus, we tried to activate these BGCs to explore cryptic products of this strain. We constructed the cluster-situated regulator overexpressing strains which contained regulator gene(s) under the control of the constitutive promoter ermE*p in S. globisporus C-1027. Overexpression of regulators in cluster 26 resulted in significant transcriptional upregulation of biosynthetic genes. With the separation and identification of products from the overexpressing strain OELuxR1R2, 3 ortho-methyl phenyl alkenoic acids (compounds 1-3) were obtained. Gene disruption showed that compounds 1 and 2 were completely abolished in the mutant GlaEKO, but were hardly affected by deletion of the genes orf3 or echA in cluster 26. The type II PKS biosynthetic pathway of chain-extended cinnamoyl compounds was deduced by bioinformatics analysis. This study showed that overexpression of the 2 adjacent cluster-situated LuxR regulator(s) is an effective strategy to connect the orphan BGC to its products.

S. fradiae WT and KOS155-3C R5/No Glucose 02/09/03

Project description:RNA timecourse data for Streptomyces fradiae wildtype (ATCC19609) and overproducing strain KOS155-3C(RUS). Strains were grown at 30 C in shake flask cultures in R5 medium with no glucose. RNA samples were harvested over 5 days as tylosin was produced. 12 h RNA samples of each strain were used as the reference sample(green) within their respective timecourses. The WT gDNA vs. 12 h RNA control displays relative gene expression at the beginning of the timecourse. The 12 h RNA control compares initial RNA levels between the WT and overproducer.
