Worker honey bee heat stress proteomics

ABSTRACT: Worker honey bees were held in cages and subject to chronic heat stress or control conditions (see publication for experimental details). At the end of the experiment, hemolymph was collected by severing one antenna and retrieving the exuding liquid. Final sample sizes are 10 per cage and 30 per group.

Transcription profiling of QMP pheromone-mediated gene expression in the honey bee brain

Project description:This experiment was designed to monitor the gene expression changes of young bees raised with and without queen mandibular pheromone (QMP). This experiment was a timecourse, comparing QMP- to QMP+ bees over 4 days of exposure. Bees that had eclosed over 16 hours were collected from a brood frame and placed in cages, 35 bees/cage. 8 hours later, queen mandibular pheromone (QMP, 0.1 queen equivalents) treatment was initiated, and fresh QMP was placed in the cage every day. One hour after QMP treatment had been refreshed, QMP- and QMP+ cages were collected into liquid nitrogen. 8 cages of each were collected on each day of the timecourse (1 day of treatment = Day 1, as well as Day 2, Day 3, and Day4).

Project description:We studied the molecular mechanisms underlying the impact of pollen nutrients on honey bee (Apis mellifera) health and how those nutrients improve resistance to parasites. Using digital gene expression, we determined the changes in gene expression induced by pollen intake in worker bees parasitized or not by the mites Varroa destructor, known for suppressing immunity and decreasing lifespan of bees.

Brain transcriptome of honeybees (Apis mellifera) infected by Nosema ceranae and/or Black Queen Cell Virus

Project description:Experimental infection of (2 days old) adult honey bee workers (30 bees per replicates, 3 replicates per treatments, from 3 different colonies (one colony per cage for each treatment)) with 10^9 genome equivalent of Black Queen Cell Virus (BQCV) in 10µl of sugar solution and/or 10^5 fresh Nosema ceranae spores (control bees were given a similar bee extract in PBS, without pathogen). Bees were kept in cages of 30 bees in incubator (30°C/50%RH). At day 13 p.i., bees were flash frozen, and stored at -80°C.

Transcription profiling of genes related to ovary activation in worker honey bee brains

Project description:This experiment examines differences in gene expression between wildtype and an experimental strain (anarchistic) of worker honey bees (Apis mellifera). Mature wildtype and anarchistic workers tend to have non-active and active ovaries, respectively. Thus young workers from these strains are expected to show differential expression at loci involved in the regulation of worker reproduction, which occurs via arrhenotokous parthenogenesis.

Transcription profiling of honey bee brain and ovary reveals pheromone response is correlated with reproductive traits and brain gene expression in individual honey bees

Project description:In honey bees (Apis mellifera), the reproductive queen produces a pheromonal signal that regulates many aspects of worker behavior and physiology and is critical for maintaining colony organization. Queen mandibular pheromone (QMP) inhibits worker reproduction, attracts workers from a short distance (retinue response), inhibits the rearing of new queens, modulates age-related division of labor and globally alters brain gene expression in worker bees. Interestingly, substantial variation in worker retinue responses to QMP has been found between colonies, but the molecular and physiological bases for variation in individual responses to the queen have not been characterized. Here, we demonstrate that individual retinue response is negatively correlated with traits associated with reproductive potential. Workers with low response to QMP have more ovarioles and higher levels of vitellogenin transcripts than workers with a high response to QMP, suggesting that workers with greater reproductive potential may be attempting to escape queen control. Retinue response appears to be associated with a suite of behavioral and physiological traits that may be pleiotropically linked. However, while these phenotypes are all correlated at the organismal level, the underlying brain expression patterns and gene networks associated with each trait are independent, suggesting that these phenotypes are uncoupled at the molecular level in adult bees. These studies provide insights into the ultimate and proximate causes of natural variation in pheromone response in honey bees.

Gene expression data for worker honey bees experimentally exposed to a dual infection with Deformed wing virus and Nosema ceranae.

Project description:Bees from 3 unrelated colonies were injected with 1ul PBS extract containing 10^9 genome equivalent of Deformed wIng virus (DWV) and/or fed 10Âµl sucrose solution containing 10^5 fresh Nosema ceranae spores. Control bees were injected and fed with an equivalent DWV- and Nosema-free extract respectively). Bees were kept in cages of 21 bees (7 from each colony), and each cage was replicated 5 times per each of the 4 treatments). Bess were kept in an incubator at 30Â°C/50%RH. At day 12 p.i., bees were flash frozen in liquid nitrogen, and stored at -80Â°C. Bee abdomen RNA was sent to Christina Grozinger lab (Penn State, USA). RNA was pooled for 3 abdomens per replicate for 5 replicates per treatment. Arrays were hybridized in a dye-swap loop design.

Honey Bee Proteome Atlas - Honey bee protein atlas at organ-level resolution

Project description:Complex organisms are composed of organs and tissues which evolved to their present state as they function together to improve an organism's overall reproductive fitness. Studies of individual organs help us understand their basic functions but this reductionist approach misses the larger context of the whole organism. This problem can be circumvented if all the organs in an organism were comprehensively studied by the same methodology and analyzed together. Using honey bees (Apis mellifera L.) as a model system,we report here the first whole proteome of a complex organism,measuring 29 different tissue types among the three honey bee castes: queen,drone,and worker. The data reveal that,e.g.,workers have a heightened capacity to deal with environmental toxins and queens have a far more robust immune system than their nestmates. Most intriguingly,our analysis reveals the path by which organs of complex organisms probably evolved.Raw data were processed by MaxQuant (v1.2.0.13) using default parameters, plus lysine and N-terminal dimethylation for three isotopologues for relative quantitation. Data were searched against the Apis Official Gene Set version 2 with common contaminants, containing a total of 22037 sequences. Normalized intensity ratios were used for relative quantitation.

Project description:The drug phenobarbital induces cytochrome P450 monooxygenase (P450) gene expression in many animals, but no changes in P450 expression, or expression of any detoxification genes, were observed in worker honey bees fed on phenobarbital-candy relative to bees fed plain candy.
