Structural evidence that RNA contributes to polymorphism of tau amyloid fibrils

Project description:RNA colocalizes with tau deposits in Alzheimer’s disease (AD) and other tauopathies, and drives tau aggregation in vitro. However, molecular details of RNA-tau interactions remain unclear, and in particular whether these interactions contribute to neurodegeneration. Previously, we determined a cryo-EM structure of fibrils of full-length tau bound to unfractionated mammalian RNA, revealing a small tau C-terminal core. Here, we present the cryo-EM structure of fibrils of full-length recombinant tau bound to unfractionated mammalian RNA and seeded by pathological tau fibrils extracted from the postmortem brain of an AD patient. This structure reveals an expanded tau C-terminal core with partial resemblance to AD tau fibrils. RNA sequencing identified 18S ribosomal RNA as the primary fibril-bound species, consistent with the well-documented link between pathological tau and ribosomal dysfunction in neurodegenerative diseases. Next, we determined the cryo-EM structure of fibrils of full-length recombinant tau bound to mammalian 18S ribosomal RNA, revealing a core that consists of the R2 to R4 repeat domains previously seen in several pathological polymorphic tau fibrils. All three of our recombinant RNA-tau fibrils dissolve upon RNase treatment, demonstrating that RNA is the molecular glue of these complexes. In the presence of different RNAs, tau fibrils adopt distinct folds, suggesting that RNA is a cofactor that shapes tau fibril polymorphism.

Mass spectrometry analysis of the blood plasma and serum samples from healthy donors before and after a meal

Project description:Serum samples were taken from four healthy donors before a meal and 3, 5, 7, 9 h after a meal (Supplementary Table S6, Sheet 1). The analysis of the serum peptidome against the HuMiProt90 database showed that the amount of non-human peptides (potentially bacterial peptides) in the bloodstream significantly increased 5 h after a meal (Figure 3D, Supplementary Table S6, Sheet 3). The comparison of these data with the transit time of food in different parts of the gastrointestinal tract suggests that bacterial peptides enter the bloodstream predominantly in the small intestine. At the same time, phyla Proteobacteria, Firmicutes and Actinobacteria are the main contributors to the increase in (potentially) bacterial peptides 5 h after a meal (Supplementary Table S6, Sheets 5 and 6).

Ribonucleotide synthesis by NME6 fuels mitochondrial gene expression

Project description:Replication and expression of the mitochondrial genome depend on the sufficient supply ofnucleotide building blocks to mitochondria. Dysregulated nucleotide metabolism is detrimental tomitochondrial genomes and can destabilise mitochondrial DNA and trigger inflammation. Here, wereport that a mitochondrial nucleoside diphosphate kinase, NME6, supplies mitochondria withpyrimidine ribonucleotides to drive the transcription of mitochondrial genes. Perturbation of NME6leads to the depletion of mitochondrial transcripts, destabilisation of the electron transport chainand impaired oxidative phosphorylation. These deficiencies are rescued upon supplementation withpyrimidine ribonucleosides. Moreover, NME6 is required for the maintenance of mitochondrial DNAwhen the access to cytosolic pyrimidine deoxyribonucleotides is limited. Our results, therefore, shedlight on the importance of mitochondrial ribonucleotide salvage for the synthesis of RNA andmitochondrial gene expression.The repository contains two internal projects which are identified by the internal IDs: 0187 and 0196.
