Project description:This study was conducted to determine the impact of extreme temperatures, aiming at controlling microorganisms by cooking, on the physiology and the transcriptome of Escherichia coli. ABSTRACT: Because the genome of Escherichia coli K12 is well understood, this organism was used as a model to investigate physiological and molecular changes during cell adaptation and survival to cooking temperatures used in the food industry. Bacteria grown at 37°C to stationary phase in Brain Heart Infusion (BHI) broth, were heated at 58°C or 60°C to reach a process lethality value (F) of 2 and 3, or until an internal core temperature of 71°C was reached (control cooking temperature). Both growth recovery and cell membrane integrity were evaluated immediately after heating and a global transcription analysis was performed by using gene expression microarrays. Only cells heated at 58°C F = 2 were still able to grow on liquid or solid BHI after heat treatment. However, their transcriptome did not differ significantly compared to those of bacteria heated at 58°C F = 3 (P-FDR > 0.01) where no growth recovery was observed post treatment. Genome-wide transcriptomic data obtained at 71°C were distinct from the other treatments. Quantification of heat-shock genes expression by real-time PCR revealed that dnaK and groEL mRNA levels were significantly lower at 71°C than at 58°C and 60°C (P < 0.0001). Furthermore, despite of similar cell inactivation but growth on BHI media after heating, 132 and 8 genes were differentially expressed at 71°C when compared to 58°C and 60°C at F = 3 respectively (P-FDR < 0.01). Among them, genes like aroA, citE, glyS, oppB and asd, whose expression was up-regulated at 71°C, may be considered as good biomarkers to determine more accurately the efficiency of heat treatments.

Project description:Quinoa is a highly nutritious seudocereal crop that is not well adapted to heat. Improving understanding of quinoa responses to heat would help expand its cultivation to regions with warmer climates. This study profiled gene expression of quinoa plants from accession QQ74 (PI 614886) treated with high tempreatures during anthesis of the main panicle, which lasted 11 days. Heat was applied in four different treatments to profile gene expression changes from root or shoot heating. Treatments were: 1. control with both roots and shoots at 22°C, 2. heated roots with roots at 30°C and shoots at 22°C, 3. heated shoots with roots at 22 °C and shoots at 35°C, and 4. heated roots and shoots with roots at 30°C and shoots at 35°C. Healthy leaves from 3 plants per treatment were sampled from the mid section of the plants, during the first (day 1) and last day (11) of heat treatment, at Zeitgeber time 2. Gene expression of each of the three heat treatments was compared to control using the Kallisto-Sleuth pipeline. These gene expression profiles of quinoa under heat are made available as a community resource to improve understanding of quinoa responses to heat.

Project description:Extended platelet-rich fibrin (e-PRF) combines the prolonged resorption properties of heat-coagulated platelet-poor plasma (PPP), becoming an albumin gel (Alb-gel) that is mixed back with the respective native cell-rich buffy coat layer (BC), the concentrated PRF (C-PRF). E-PRF or Alb-PRF is utilized as a barrier membrane in various clinical applications, such as guided tissue regeneration. Heating of PPP might lower its biological activity, but testing this hypothesis is necessary. To this end, we exposed gingival fibroblasts to lysates of regular PPP, heated PPP (hPPP), and BC, followed by bulk RNA sequencing. Gingival fibroblasts respond to PPP lysates with a total of 153 up- and 71 down-regulated genes when considering a minimum 3.0-fold log2 expression change and a significance level 2.0 log-10. In sharp contrast, the response to hPPP was characterized by only five up-regulated and five down-regulated genes, clearly indicating that heating almost completely abolished the biological activity of PPP. As expected, BC was more potent than PPP and broadened the spectrum of regulated genes. RT-PCR and immunoassays confirmed the heat sensitivity of PPP as exemplified by IL11 and other genes. Moreover, PPP, but not hPPP, drives the phosphorylation of p65, representing NF-κB signaling. Taken together, these findings extend previous observations that PPP causes a robust response in gingival fibroblasts and also strengthen the hypothesis that this response is heat-sensitive. These operations support the clinical concept of e-PRF by mixing back the heated inactive PPP with the bioactive buffy coat C-PRF layer.

Project description:During the application of thermal treatment, cell survival depends on biological factors (such as growth stage, culture conditions, and the composition of the stress medium) as well as physical factors inherent to the thermal process (including amplitude, duration at the stress temperature, and the rate of temperature increase). In this study, the yeast Saccharomyces cerevisiae BY4742 was exposed to two different thermal treatment kinetics in a nutrient medium after being cultivated in YPD medium at 25 °C for 48 hours (to reach the stationary growth phase). The thermal treatments were applied between 25 °C and 50 °C, followed by a 30-minute hold at 50 °C before returning to 25 °C. The yeast cells were exposed either to a heat shock (heating kinetics: 50 °C/min) or to a gradual temperature increase (heating kinetics: 0.5 °C/min). Generally, it is known that heat shock induces lower survival (30%) compared to the gradual increase (80%). In this study, we demonstrate that despite the small difference in survival rate (less than one logarithmic reduction in both cases), the transcriptomes vary significantly. The use of bioinformatics tools such as WGCNA revealed a correlation between differentially expressed gene clusters and phenotypic traits (measured by spectroscopy, fluorescence microscopy, flow cytometry, among others). Here, we focused on both mRNA and ncRNA (non coding RNA).
