MesoMap: Mesoscale proximity labeling of chromatin for small molecule mechanism of action

ABSTRACT: Mesoscale proximity labeling of chromatin is used to understand the impact that inhibitors have on chromatin structure and dynamics. We use uMap proximity labeling, distributed throughout chromatin to study how HDAC inhibition alters interactions. We then apply this approach to de-orphan a high throughput screening hit SR1815. We show that SR1815 is a kinase inhibitor that hits CLK proteins and alters RNA splicing.
