Specifically, tetracycline-inducible HIF-PPN mice (Pro402, Pro564 and Asn803 of HIF1a were mutated to Ala residues to limit degradation, termed HIF-PPN) were crossed to a-MHC-tTA expressing mice. HIF-PPN expression was induced by removal of doxycycline for 3 days and compared to control hearts that remained on doxycycline (uninduced).

Project description:Induction of the transcription factor Sox2 from a doxycycline-inducible promoter in iSox2-DAOY medulloblastoma cells. Affymetrix microarrays were used to characterize the gene expression profile of iSox2-DAOY cells in the absence and presence of doxycycline. Human DAOY medulloblastoma cells were engineered to express epitope tagged Sox2 under the control of a doxycycline inducible promoter, to produce iSox2-DAOY cells. iSox2-DAOY cells were FACS sorted into CD133 low and high populations. The CD133 low population was cultured in the absence and presence of 0.5 Âµg/mL doxycycline for 24 hours. RNA was extracted from cells cultured without and with doxycycline, and was used for microarray analysis.

Project description:We studied the impact of hsa-miR-139-5p on the protein output by means of an iTRAQ-based approach. First, we established two CAL-62 isogenic cell lines expressing either the mature hsa-miR-139-5p or a non-targeting control upon a doxycycline inducible promoter (PTRE3G-tGFP, Dharmacon). Total proteins of P-tGFP-hsa-miR139-5p untreated or treated with doxycycline (1ug/ml) for 96 and 120 hours were isolated and labeled with iTRAQ® reagent 8-plex. Two independent experiments were performed.

SILAC phosphoproteomics of 293-F10 cells [induced (H) vs uninduced (L)]

Project description:The objective of this experiment was to identify cellular targets of F10-mediated phosphorylation through Stable Isotope Labeling of Amino acids in Cell culture (SILAC). 293-F10 cells were propagated in heavy (H) or light (L) media for 10 cellular doublings. Mass spectrometry was used to confirm that the majority of tryptic peptides showed >90% incorporation of the heavy isotope. H-labeled cells were treated with doxycycline to induce expression of F10 for 10 hours, and then these cells were mixed with uninduced, L-labeled cells in a 1:1 ratio. Three biological replicates were obtained from separately passaged cultures and doxycycline inductions. Cells were lysed and post-nuclear supernatant proteins were precipitated and subjected to tryptic digestion. Tryptic peptides were desalted, enriched for phosphopeptides, and analyzed in triplicate by LC-MS/MS. The resulting 9 data files were pooled and phosphoprotein identification and quantification was completed using MaxQuant software (version 1.2.2.5). Differential phosphorylation in response to F10 induction was determined using an outlier score for log protein ratios. The Benjamini-Hochberg test was then used to correct for the biased statistical spread of highly abundant proteins.
