Scenedesmus obliquus UTEX393 Proteomics

ABSTRACT: This study is part of a comprehensive multi-omics study to characterize the responses under brackish conditions for the biofuel relevant and historically significant strain, Scenedesmus obliquus UTEX393. The time-course study sampled over a 38-hour window comparing growth in the presence of 15 ppt versus 5 ppt NaCl media conditions.

Transcriptome analysis of the phytopathogen Xylella fastidiosa in response to salt and osmotic stress

Project description:The global transcriptional response of the phytopathogenic bacterium X. fastidiosa to a sudden increase in salinity and osmolarity of the medium was investigated using DNA microarrays. Time-course experiments were carried out by exposing bacterial cells to high salinity (250 mM NaCl) and high osmolarity (300 mM sucrose) conditions revealing 142 upregulated genes under both stresses, including pathogenicity related genes, genes encoding transcriptional regulators, as well as genes related to DNA metabolism and mobile genetic elements. In addition, 38 genes were downregulated under both conditions, most of them related to ribosomal and heat shock proteins. A total of 192 genes were upregulated exclusively in the presence of NaCl, including genes encoding transporters, genes involved in oxidative-stress response and genes related to cell structure. A smaller number of genes (44 genes) were induced only in the presence of sucrose, most of them encoding hypothetical and conserved hypothetical proteins. Interestingly, 57% of the genes differentially expressed under both stress conditions have no putative function assigned, emphasizing the importance of high-throughput experiments to start the characterization of these genes in X. fastidiosa. Project description:We study the transcriptional reactions of bacteria interesting for biodegradation under laboratory conditions that mimic water stress. We compared the transcriptomes of cultures growing exponential phase under optimal conditions versus their responses to an osmotic shock of 30 min in exponential phase. The osmotic shock consisted in a reduction of water potential induced by salt (NaCl, solute stress) or by polyethylene glycol (PEG8000, matric stress). The stress was such that cells are not more than 20% affected in their maximum growth rate.

Effect of p38 SAPK activation on HeLa cells

HeLa cells treated with 100 mM NaCl during 2h versus HeLa cells treated with 100 mM NaCl during 2h after pre-treatment with 10 μM SB203580 for 30 min. Alternative splicing is a crucial mechanism for gene regulation that is modulated in response to a wide range of extracellular stimuli. Stress-activated protein kinases (SAPKs) play a key role in controlling several steps of mRNA biogenesis. Here, we show that osmostress has a major impact on the regulation of alternative splicing (AS), which is partly mediated through the action of the p38 SAPK.

Project description:To identify mediators of the osmotic stress response in Mycobacterium tuberculosis (Mtb), we performed transcriptional profiling of WT CDC1551 following treatment with 140 mM NaCl for 1 hr relative to an untreated control. 140 mM NaCl was chosen to reflect the approximate osmolarity of human plasma (i.e., 280 mOsm/L), which may be relevant during the course of infection.
