ABSTRACT: The study focuses on phragmalin-type limonoids found in leaf extracts from S. macrophylla at three different maturation stages, utilizing ultra performance liquid chromatography coupled with mass spectrometry to build molecular networks on the GNPS platform.

Identification and analysis of microRNAs in the SAM and leaves of Populus tomentosa

Project description:High-throughput sequencing was performed to build four sRNA libraries (each sample with two replications) derived from shoot tips and mature leaf tissues of P. tomentosa. Millions of small RNA (sRNA) reads were obtained, and many known and novel miRNAs in SAM and leaf libraries were established. Several miRNAs that were differentially expressed between the SAM and its surrounding tissues, and their targets involved in important biological processes were identified and analyzed. Our study provide insights for better understanding the regulatory mechanisms of miRNAs involved in the SAM of woody plants.

Molecular detection of maturation stages in the developing kidney

Project description:To identify maturation-dependent genes, we here performed single cell RNA sequencing (scRNA-seq) analysis using developing kidneys at different stages in the mouse, followed by highly sensitive in situ hybridization. We identified multiple genes expressed abundantly in newborn kidneys, but minimally at embryonic day 15.5. We then applied these maturation markers to the transplanted embryonic kidneys and found that the maturation process did not occur equally throughout nephron segments upon transplantation: glomeruli and proximal renal tubules became more mature than the other nephron segments. Thus, our scRNA-seq data at multiple stages of kidney development and identified maturation-dependent genes will serve as useful bases for assessing maturation of the developing kidney, and eventually of kidney organoids.

Leaf maturation transcriptional dynamics are plastic in response to stress (hormone data)

Project description:Leaf development is dynamic in response to environmental stress. Although such developmental plasticity in response to stress is well described at the physiological level, the underlying transcriptional changes that adjust leaf development and produce adaptive phenotypes remain poorly understood. Here, using single-nucleus transcriptome sequencing, we demonstrate that that drought-induced changes in leaf development involve promoting transcriptional responses linked to leaf maturation across leaves of all ages. Notably, we find that these transcriptional changes scale with stress intensity, ensuring the resulting tissue size is dose-responsive with environmental change.

MicroRNA-15/16 Antagonizes c-Myb to Control Natural Killer Cell Maturation

Project description:NK cells develop in the bone marrow and complete their maturation in peripheral organs, but the molecular events controlling maturation are incompletely understood. Utilizing an NK cell-specific miR-15/16 deficient genetic model (15aKO), we identified a critical role for miR-15/16 family microRNAs in the normal maturation of NK cells in vivo, with a specific reduction in mature CD11b+CD27- NK cells in multiple tissues. The mechanism responsible was a block in differentiation, since accelerated NK cell death was not evident, and earlier intermediates of NK cell maturation were expanded. Further, we identified Myb as a direct target of miR-15/16 in NK cells, with Myb expression increased in immature 15aKO NK cells. Following adoptive transfer, immature 15aKO NK cells exhibited defective maturation, which was rescued by ectopic miR-15/16 expression or Myb knockdown. Moreover, Myb overexpression resulted in defective NK cell maturation. Thus, miR-15/16 regulation of Myb controls the normal NK cell maturation program. 3 technical replicates each of CD27+ 15a/16-1FKO NK cells, and CD27+ Ctrl NK cells
