The Protein Complex Proteome of Phaeodactylum tricornutum organelles
Ontology highlight
ABSTRACT: A native organelle-enriched fraction isolated from Phaeodactylum tricornutum was separated by two-dimensional Blue-native / SDS Polyacrylamid Gelelectrophoresis (2D BN / SDS PAGE). After vizualization of proteins on the gel with Coomassie, protein spots were cut, cysteine residues were carbamidomethylated and an in-gel digestion was carried out with trypsin over night. Peptides were extracted and used for HPLC-tandem MS using an online U3000 (Dionex) - Q Exactive Orbitrap MS system. Data were analyzed with MaxQuant (Version 2.6.8.0) and further processed in Excel (Microsoft Office). The final processed dataset was used to generate a reference map at www.gelmap.de (https://www.gelmap.de/5848).
