MS sequencing of GFAP fragments from TBI patients CSF

ABSTRACT: Protein Glial fibrillary acidic protein, GFAP fragment identification (breakdown products, BDPs) and their label free peptide/ion quantification of GFAP immunoprecipitated from TBI patient's cerebrospinal fluid using a polyclonal anti-GFAP antibody. GFAP fragments of distinct molecular weights were cut out and in gel-digested prior to LC-MS/MS using Proteome discoverer 1.4. Peptide results from PD output files were combined from four TBI patients, 6 CSF samples to generate a multiconsensus peptide coverage map for each molecular weight fragment. Mutant GFAP alters lineage commitment of neural organoids [AxDorganoids_bulk_experiment2]

Project description:Glial fibrillary acidic protein (GFAP) is a type-3 intermediate filament protein mainly expressed in astrocytes in the central nervous system. Mutations in GFAP cause Alexander Disease (AxD), a rare and fatal neurological disorder. How exactly mutant GFAP eventually leads to white and grey matter deterioration in AxD remains unknown. GFAP is also expressed in radial glial cells, which are neural stem cells in the developing brain. Here, we used AxD patient-derived induced pluripotent stem cells (iPSCs) to explore the impact of mutant GFAP during neurodifferentiation. Our results show that GFAP is already expressed in iPSCs. Moreover, we have found that mutations in GFAP can severely affect neural organoid development through altering lineage commitment in embryoid bodies. Together, these results support a key role for GFAP as an early modulator of neurodevelopment.

GFAP and MBP peptides in Traumatic Brain Injury

Project description:Glial fibrillary acidic protein (GFAP) is the major intermediate filament III protein of astroglia cells and is upregulated in astrogliosis following traumatic brain injury (TBI). Here we reported that GFAP is truncated at both the C- and N-terminals by cytosolic protease calpain to GFAP breakdown products (GBDP) of 46-40 kDa then 38 kDa following pro-necrotic (A23187) and pro-apoptotic (staurosporine) challenges to primary cultured astroglia or neuron-glia mixed cells. In addition, with another pro-apoptotic challenge (EDTA) where caspases are activated but not calpain, GFAP was fragmented internally, generating C-terminal GBDP of 20 kDa. Following controlled cortical impact in mice, GBDP of 46-40K and 38K were formed at day 3 to 28 post-injury. Purified GFAP protein treated with calpain-1 and -2 revealed that they produce (i) major N-terminal cleavage site between A-56 and A-75, and (ii) major C-terminal cleavage sites between T-383 and Q-388, producing a limit fragment of 38K and concurrently releasing several small GFAP peptides. Caspase-6 treated GFAP was cleaved at D-78, R-79, D-266 and A-267, where GFAP was relatively resistant to caspase-3. We also derived a GBDP-38K N-terminal specific antibody which only labels injured astroglia cell body in both cultured astroglia and in mouse cortex and hippocampus after TBI. Lastly as clinical translation, we observed that CSF samples collected from severe human TBI have elevated levels of calpain-generated GBDP-38K. Thus by examining the various forms of GFAP breakdown products as biomarkers, one can gain mechanistic insights of tracking astroglia injury under conditions where calpain and/or caspase-6 are activated.

Identification of genes associated with the astrocyte-specific gene Gfap during astrocyte differentiation

Project description:Chromosomes and genes are non-randomly arranged within the mammalian cell nucleus. Clustering of genes is of great significance in transcriptional regulation. However, the relevance of gene clustering in their expression during differentiation of neural precursor cells (NPCs) into astrocytes remains unclear. We performed a genome-wide enhanced circular chromosomal conformation capture (e4C) to screen genes associated with an astrocyte-specific gene, glial fibrillary acidic protein (Gfap), during astrocyte differentiation. We identified 13 genes that were specifically associated with Gfap and expressed in NPC-derived astrocytes. These results provide evidence for functional significance of gene clustering in transcriptional regulation during NPCs differentiation. comparison of NPCs vs LIF+ vs LIF- cells.

Hippocampal gene expression in the GFAP-R237H rat model of Alexander disease at early and late stages of disease

Project description:Alexander disease is a rare neurodegenerative disorder caused by mutations in the gene for glial fibrillary acidic protein (GFAP), the major intermediate filament of astrocytes in the central nervous system. GFAP mutation causes a toxic gain-of-function and protein aggregation, ultimately leading to gliosis, astrocyte dysfunction, and neurodegeneration. To better understand the disease process, a rat model has been generated to mimic the common R239H mutation observed in the human disease (R237H in the rat). This study focuses on hippocampus, a brain region with a heavy burden of pathology, to determine the impact of GFAP mutation at presymptomatic (3 weeks of age) and severe stages ( 8 weeks of age) of disease. Transcription profiling shows progressive neuroinflammation and neurodegeneration.

Differentially regulted genes in neural stem cells treated by oleanolic acid

Project description:Oleanolic acid significantly inhibited neurosphere formation in a dose-dependent manner, and achieved a maximum effect at 10 nM. OA also reduced the 5-ethynyl-2’-deoxyuridine (EdU) incorporation into NSCs, indicating an inhibition on proliferation of NSCs. Next, in western blotting analysis, the protein expression of neuron-specific marker tubulin-βⅢ (TuJ1) and Mash1 was increased, while the astrocyte-specific marker glial fibrillary acidic protein (GFAP) decreased. In immunofluorescence analysis, OA significantly elevated the percentage of TuJ1-positive cells and reduced GFAP-positive cells. Using DNA microarrays, 183 genes were found to be differentially regulated by OA.

Transcriptome analysis of the uninjured Muller glia (MG) and MG derived retinal progenitors at 4 day post injured zebrafish retina

Project description:The study was aimed at comparing the transcriptome of MG cells of the retina with the progenitors derived from them after an injury. This information will help in the identification of factors that are responsible for the retinal regeneration. Muller glia were isolated by fluorescent activated cell sorting (FACS) using uninjured retinas from transgenice zebrafish (gfap:gfp) where green florescent protein (GFP) is under the control of the glial fibrillary acidic protein (gfap) promoter. MG-derived retinal progenitors were isolated by FACS at 4 days post retinal injury from 1016 tuba1a:gfp transgenic fish where GFP is driven by the tuba1a promoter which is specifically activated in these progenitors. Total RNA was isolated from these cell populations and subjected to microarray analysis to compare their transcriptomes. Samples were prepared in duplicate.
