Project description:Yucatan minipigs, fasted overnight, were previously fed either a control or a Western diet (high-fat, high-sucrose). They received 23.7ml/kg of their body weight in liquid diet via intra-gastric tubing, with blood samples collected 120 minutes post-administration. For sampling, catheters were inserted into the femoral artery and vein. Blood was drawn from the hepatic, portal, renal, splenic, and inferior mesenteric veins, the lateral auricular vein, the aorta, and the coronary sinus using a 23G butterfly needle attached to a 1 mL syringe. The external jugular vein and the left and right ventricles were sampled using a 27G needle connected to a 1 mL syringe. All organ blood samples were taken within 5-10 minutes of the initial sampling. Tissue samples were collected post-terminal surgery.

Project description:Acute pulmonary thromboembolism (PTE) refers to the obstruction of the pulmonary artery or its branches by thrombus. Recent studies suggest that PTE-induced endothelium injury is the major physiological consequence of PTE, and PTE-induced endothelium injury can be used to stratify disease severity. Several plasma biomarkers have been identified in PTE patients, but genome-wide gene expression has not been attempted in PTE. In this study, using rabbit autologous thrombus PTE model, we applied gene expression array to identify genes and proteins changes in pulmonary arteries under PTE. Twenty healthy China big-ear rabbits, specific-pathogen-free (SPF) grade and weighing between 2.5 and 3 kg, were purchased from the Center of experimental animals of the Nantong University. These rabbits were randomly divided into control (n=10) and experimental group (n=10). One hour before the experiment, thrombi were generated by collecting 0.5 ml blood from the marginal ear vein and placing into a sterile petri dish for 45 minutes at room temperature. The autologous blood clot was subsequently cut into 5 mm sized aliquots and suspended in sterile saline. After anesthesia, a venous catheter was placed in the exposed and isolated femoral vein. In the experimental group, the blood clot in 10 mL of saline was injected into the femoral vein. An additional 5 mL of saline were used to flush the catheter. In the control group, 15 mL of saline were injected into the femoral vein. Afterwards, the animals were placed back in the cage.

The study was approved by the Thomas Jefferson University Institutional Animal Care and Use Committee and was conducted in accordance with the National Institutes of Health Guide for the Care and Use of Laboratory Animals. The animals were housed in Thomas Jefferson University's animal care facilities. Animals were anesthetized with isoflurane dissolved in O2 (5% induction; 1% maintenance) and one femoral artery and vein were cannulated (PE-50 tubing) via a small medial incision for measurement of arterial pressure and infusion of drugs, respectively. The cannulae were run subcutaneously to an exit incision between the scapulae. The leg wound was sutured and topical anesthetic (lidocaine) was applied to both skin incisions. Following surgery, after one hour of stable and normal resting blood pressure and heart rate, intravenous infusion was initiated of approximately 1 mL saline, as a control, or phenylephrine (200 µg/mL; 1 mL/hr), to induce hypertension. We followed standard methods in the use of phenylephrine (PE) to elevate blood pressure. PE does not cross the blood brain barrier and the elevated blood pressure it produces has been shown to cause molecular effects in the NTS principally via increased baroreceptor afferent drive by both pharmacological and sinoaortic denervation studies. Project description:We measured genome-wide transcript abundance patterns in 246 whole blood samples collected from 35 cynomolgus macaques before and after vaccination with a live attenuated tetravalent dengue vaccine (TDV) or PBS (placebo). Animal work was conducted at the Charmany Instructional Facility of the School of Veterinary Medicine, University of Wisconsin-Madison, and all animal procedures were approved by the University of Wisconsin-Madison's Animal Care and Use Committee. Thirty-five adult male, DENV-seronegative cynomolgus macaques from Vietnam were placed in quarantine for 30 days prior to the start of the study. Five groups of animals (n=6 animals per group) received TDV either subcutaneously (SC) in 0.5 mL inocula or intradermally (ID) in 0.1 mL inocula using a needle-free injector (PharmaJet' device) or needle and syringe (N&S). Animals received two doses (same route) of TDV either on the same day (Day 0) or sixty days apart. One group (n=5) received PBS by ID route and needle-free injector. On day 90 after primary vaccination, 3 animals from each group were challenged with 105 PFU wt DENV-2 (New Guinea C strain) or 105 PFU wt DENV-4 (Dominica/81 strain) by SC route using N&S. Whole blood samples were collected for transcriptional profiling from all animals on days -11, -2, 1, 3, 5, and 7 before and after immunization, and for placebo animals on days 88, 91, 93, 95, and 97 before and after wt DENV challenge. Blood samples were drawn into PAXgene Blood RNA tubes and stored at -80'C prior to RNA extraction and processing.

Keywords: Hypertension, time series In keeping with practice of previous studies we induced a defined period of hypertension and then measured effects on the NTS at time points following that standard period of blood pressure disturbance. The previous studies show that blood pressure elevations of 25-30mmHg above baseline induce c-fos expression in the NTS after 30-40 minutes. Based on this result, we estimated that transcriptional regulation in the NTS might be observed after 60 minutes. For this reason we used a standard 60 minute acute hypertension experimental treatment. We took the first samples immediately after the 60 minutes of PE or saline infusion at 15 minute intervals. Our experimental design is for time series data at 60, 75, 90, 105 and 120 minutes (see Figure 1). Gas anaesthesia has previously been reported to induce p-ERK1/2 phosphorylation in some central autonomic structures so the present study reports only differential data that compares to controls that experience the same surgery and fluid (saline) infusion. In order to maximize the statistical power of the experimental design and capture biological variability we did not pool mRNA across animals, but used one array per animal. Four control and four treated animals were taken for each time point and condition for a total of 40 animals. In our previous studies, we found that four replicates in each condition were necessary to accurately and reproducibly measure small, physiologically relevant fold changes as low as 1.2-fold.

Project description:The study aims to determine the molecular signature associated with varying applications of a novel skin vaccination array, compared to traditional needle and syringe immunization. The hypothesis is that the vaccination array induces a better immune response compared to the needle and syringe and this is due to a heightened inflammatory profile at the site of vaccination. We performed the study on wild-type mice that received varying forms of immunization, including intradermal needle and syringe with or without vaccine, Nanopatch with or without vaccine, Nanopatch with vaccine with QS-21 adjuvant, and Nanopatch with vaccine at higher application energy.

Project description:The locational predisposition of vascular pathologies illustrates the need for a better insight into vascular heterogeneity. To investigate the transcriptomic basis of angiodiversity, we isolated and analyzed transcriptomes from endothelial cells and vascular smooth muscle cells from nine different adult canine macrovessels: the aorta, coronary artery, vena cava, portal vein, femoral artery, femoral vein, saphenous vein, pulmonary vein, and pulmonary artery. We identified both reported and novel expression patterns defining specialized adult blood vessels. Our findings also show that adult vascular cells in culture express a remarkably high number of transcription factors crucial to organ development in the embryo. The persistent expression of these genes in culture indicates that these genes are not regulated by the flow or surrounding cell types but are rather fixed in the molecular memory. Therefore, our findings prompt the re-thinking of the extrapolation of results from single-origin endothelial cell systems.
