Specialized metabolome data from Arabidopsis accession seeds (ESI -)

ABSTRACT: Untargeted metabolomic analyses (ESI -) were carried out on dry seeds from 85 Arabidopsis thaliana accessions grown in a greenhouse with a minimum photoperiod of 13 h ensured by supplementary lighting (with Philips-E40 red spectrum - HPS 400W SONT PIA PLUS bulbs; 120 mE.m-2.s-1), and irrigated with Plan-Prod nutritive solution (Fertil).

Gene expression in dormant, germinating and thermoinhibited Arabidopsis seeds

Project description:To establish the basis for understanding molecular mechanism of seed germination response to temperature, we analyzed transcriptomes in freshly harvested dormant and dry stored after-ripened seeds. The after-ripened seeds started to show visible germination from 36h after the start of imbibition, and almost all the seeds germinated after 3 days. The freshly harvested seeds stayed dormant by imbibition at 26°C, and germination of the after-ripened seeds was almost completely inhibited at 34°C. Total RNA was prepared from 0 (dry), 6 and 24h imbibed seeds to find regulatory genes of seed dormancy and germination.

Specialized metabolome data of dry seeds from WT, aop3, scpl17 and bzo1 genotypes developed under high temperatures

Project description:Arabidopsis thaliana (thale cress) wild-type (WT) and transgenic lines were in the Columbia-0 (Col-0) background. The T-DNA insertion mutants of AOP3 (SALK_001655C), BZO1 (two independent lines, SALK_094196 and GABI_565B09) and SCPL17 (two lines independent, SALK_075481 and SALK_126264) and the WT were sowed directly in fertilized soil. Plants were grown in optimal conditions (control) in a growth chamber with long day photoperiod (16h light/21 celsius degrees - 8h dark/19 celsius degrees). Watering was made with water. At the onset of flowering (occurring between 5 and 6 weeks after seed sowing), high temperature (HT) was applied to half of the plants of the corresponding experiment by transferring them to another growth chamber (SANYO, Thermo Fisher Scientific) displaying high temperature conditions (16h of light/27 celsius degrees - 8h of dark/24 celsius degrees). The other half of the plants, constituting control plants, remained in the initial growth chamber with control temperature conditions. Dry seeds were harvested. Untargeted metabolomics (LC-MS/MS) were performed with those samples of dry seeds.

Barley seeds miRNome stability during long-term storage and aging

Project description:Seed aging is a complex biological process attracting the scientists’ attention for many years. High-throughput small RNA sequencing was applied to examine microRNAs contribution in barley seeds senescence. Unique samples of seeds that despite the same genetic makeup differed in viability after over 45 years of storage in a dry state were investigated. In total, 61 known and 81 novel miRNA were identified in dry seeds. The highest level of expression was found in four conserved miRNA families i.e. miR159, miR156, miR166 and miR168. However, the most astonishing result was the lack of significant differences in the level of almost all miRNAs in seed samples with significantly different viability. This result reveals that miRNAs in dry seeds are extremely stable. This is also the first identified RNA fraction that is not deteriorating along to the loss of seed viability. Moreover, the novel miRNA hvu-new41, with higher expression in seeds with the lowest viability was detected by RT-qPCR, has the potential to become an indicator of the decreasing viability of seeds during storage in a dry state. It might be responsible for the removal of (1-3.1-4)-beta-D-glucanase transcripts and lowering or completely blocking the synthesis of this key enzyme for seed germination.

Influence of osmopriming on rape seeds germination

Project description:Transcriptomic study of the impact of osmopriming on rape seeds (Brassica napus L.; cv 'Libomir') during priming process and after germination. The assays were replicated twice on two independent priming and germination experiments. Seeds were osmoprimed in PEG solution (-1.2 MPa osmotic potential) during 7 days, dried to initial moisture content and then germinated for 7 hours on water. The analysis during different phases of priming procedure (soaking and drying), after whole osmopriming process and germination were done. 10 samples, four condition experiment; non dried primed seeds (Pnd) vs. dry unprimed seeds (UPd) (PEG soaking), non dried primed seeds (Pnd) vs dry primed seeds (Pd) (drying after soaking), dry primed seeds (Pd) vs. dry unprimed seeds (UPd) (full osmopriming process), primed seeds imbibed on water (P7h) vs unprimed seeds imbibed on water (UP7h) (germination after osmopriming). Biological replicates: 2 replicates for comparison PEG soaking, drying after soaking, full osmopriming process and germination after osmopriming.
