Introducing Eyewear Lenses as Passive Samplers for Assessing Inhalation Exposure to Airborne Chemicals

Project description:Data-dependent acquisition results for air samples, collected using PDMS-coated and uncoated polycarbonate lenses. Separation was conducted on an Agilent 1290 Infinity II HPLC with InfinityLab Poroshell 120 EC-C18 (Agilent) column (100 mm x 2.1 mm x 1.9 um). Lenses were extracted by wiping with isopropyl alcohol wipes (for uncoated polycarbonate lenses) or acetonitrile wipes (for PDMS-coated lenses). Wipes were extracted by shaking in acetonitrile for 15 minutes at 150 RPM, then concentrated to near dryness and reconstituted in 0.5 ml.

Multi-Omics Profiling Reveals Distinct Lipidomic and Transcriptomic Alterations in Gestational Diabetes Mellitus, Preeclampsia, and Coexistence of Both: Implications for Maternal and Neonatal Outcomes

Project description:Gestational diabetes mellitus (GDM) and preeclampsia (PE) are major contributors to maternal and perinatal morbidity and mortality, characterized by dysregulated lipid metabolism, chronic inflammation, and immune dysfunction, disrupting maternal-fetal homeostasis. The coexistence of GDM and PE (PG) presents a uniquely complex clinical scenario with molecular mechanisms that remain poorly understood. Peripheral blood samples from 42 pregnant women were categorized into CTL (n=10), GDM (n=12), PE (n=10), and PG (n=10) groups. Lipidomic profiling via LC-MS and transcriptomic analysis through RNA sequencing were performed. Multi-omics integration correlated molecular signatures with clinical and neonatal outcomes, and a predictive model for PE was constructed using differentially expressed lipids. Additionally, UK Biobank data was analyzed to explore associations between pregnancy complications and maternal postpartum depression (PD).PE exhibited the most pronounced lipidomic alterations, correlating with adverse neonatal outcomes, including low birth weight, while GDM showed the most significant transcriptomic changes. PG demonstrated unique transcriptomic signatures associated with neuronal and immune dysfunction, suggesting a potential link to maternal PD. Seven lipid biomarkers were identified, significantly correlating with neonatal outcomes, and a lipid-based predictive model for PE showed robust performance across PE and PG cohorts. GDM and PE were significantly associated with PD, though PG’s association remains inconclusive due to limited data.This study highlights the utility of multi-omics in unraveling shared and disease-specific mechanisms underlying pregnancy complications. The identified biomarkers and PE predictive model offer promising avenues for advancing diagnosis, prognosis, and personalized maternal-fetal care.

Cardiac lipidomic profiling and mechanistic exploration by transcriptome in murine model of dilated cardiomyopathy

Project description:Dilated cardiomyopathy (DCM) and heart failure are associated with mitochondrial dysfunction and dysregulated lipid metabolism. Recent studies have implicated a pivotal role of Hippo pathway activation in the development of DCM with profound metabolic remodelling. However, alterations in the cardiac lipid profile and responsible molecular mechanisms, including upregulated galectin-3, remain unclear in the setting of DCM. Mice with DCM induced by Hippo pathway activation (Mst1 gene overexpression alone or with galectin-3 gene deletion) were studied by lipidomic profiling, transcriptomic profiling and immunoblotting for mRNA and protein expression. Profound alterations in cardiac lipid classes were observed, notably elevated sphingolipids, reduced triacylglycerol (TG) and ether lipids, alterations in phospholipid species, elevated lysophosphatidylcholine, and a profound reduction in mitochondria-specific cardiolipin and coenzyme Q10. Relative to adult mice with advanced DCM, reduced TG content was evident but sphingolipid accumulation was absent in young mice at early phase of DCM. Mechanistically, the lipidomic profile in DCM heart was in consistent with dysregulated expression of specific gene sets for biosynthesis of ceramides or TG, TG storage/hydrolysis, mitochondrial lipid metabolism, peroxisome biogenesis, and suppressed PPAR-alpha signaling. Furthermore, galectin-3 gene deletion resulted in changes in some lipid classes and numerous lipid species in settings of healthy and DCM. Multiple alterations in cardiac lipids were identified by lipidomics in a mouse model of DCM due to activation of the Hippo pathway. By employing transcriptome and galectin-3 gene deletion, we revealed underlying mechanisms for lipid abnormalities involving upregulated galectin-3, downregulated expression of specific gene sets of lipid metabolism, and attenuated PPAR-alpha signaling.

Transcriptomic and lipidomic analysis of the differential pathway contribution for the incorporation of erucic acid to triacylglycerol during pennycress seed maturation

Project description:Thlaspi arvense (Pennycress) is an emerging feedstock for biofuel production because of its high seed oil content enriched in erucic acid. A transcriptomic and a lipidomic study were performed to analyze the dynamics of gene expression, glycerolipid content and acyl-group distribution during seed maturation. Genes involved in fatty acid biosynthesis were expressed at the early stages of seed maturation. Genes encoding enzymes of the Kennedy pathway like TaDGAT1, TaLPAT or TaGPAT increased their expression with maturation, coinciding with the increase in TAG species containing 22:1. Positional analysis showed that the most abundant TAG species contained 18:2 at sn-2 position in all maturation stages, suggesting no specificity of the TaLPAT for VLCFAs. TaDGAT2 mRNA was more abundant at the initial maturation stages, coincident with the rapid incorporation of 22:1 to TAG, suggesting a coordination between DGAT enzymes for TAG biosynthesis. Genes encoding TaPDAT1, TaLPCAT or TaPDCT, involved in acyl-editing or PC-derived DAG/TAG biosynthesis showed also higher expression at the early maturation stages, coinciding with a higher proportion of TAG species containing C18 fatty acids. These results suggested a higher contribution of these two pathways at the early stages of seed maturation. LC-MS analysis of the content and acyl-group distribution of DAG and PC pools was compatible with the acyl content in TAG at the different maturation stages. Our data point to a model in which a strong temporal coordination between pathways and isoforms in each pathway, both at the expression and acyl-group incorporation, contributes to high erucic TAG accumulation in Pennycress.
