Project description:Quantification of the human S100A8/S100A9 tetrameric protein complex in stool, referred to as fecal calprotectin, is an extensively validated biomarker supporting the diagnosis and management of gastrointestinal diseases1,2. The quaternary protein structures (called configuration here) of S100A8 and S100A9 and their biological function in the human intestine is unknown. This study unravels a diagnostic value for fecal S100A9 detection in IBD and identifies pleiotropic inflammatory mechanisms of S100A8 and S100A9 homodimers in the intestine.

Investigation of the Dietary Ingestion of 2-Aminoanthracene in Sprague Dawley Rats using genomic tools.

Project description:The present research focuses on 2-aminoanthracene (2AA), recognized as a derivative of PAHs. The Centers for Disease Control and Prevention (CDC) characterizes PAHs as exhibiting properties that adversely affect the health of various living organisms. These compounds increase the risk of neoplasms in humans and animals and are implicated in the increase of genetic mutations. 2AA, an aromatic amine, has demonstrated elevated carcinogenic and mutagenic potential on animals7. In the long term, exposure to PAHs may cause damage to the liver and kidneys. Furthermore, 2AA has been associated with the onset of early diabetic symptoms9. Consumption of 2AA by Sprague-Dawley rats has shown inflammatory and genetic overregulation, increased glucose, and decreased insulin production. This study aims to examine the hepatic impact of 2AA ingestion by Sprague-Dawley Rats. Rats have anatomical structures of the organs similar to those of humans, so they are suitable for research in biomedical science.

Project description:Carnitine is a water soluble quaternary amine which is essential for normal function of all tissues. Besides improving performance characteristics of livestock animals carnitine has attracted scientific interest due to several health-related effects, like protection against neurodegeneration, mitochondrial decay, and oxidative stress, improvement of glucose tolerance and insulin sensitivity as well as reduction of hepatic steatosis. To gain insight into mechanisms through which carnitine exerts its beneficial metabolic effects, we fed piglets either a control or a carnitine supplemented diet, and analyzed the transcriptome in the liver, which plays a central role in whole body metabolism.

Phosphoproteomic analysis of methanoarchaeon Methanohalophilus portucalensis FDF1T reveals diverse functions in methanogenesis and osmoadaptation

Project description:Methane is a well-known factor in green house effect and also could be an alternative energy, which produced via methanogenesis by methanogenic archaea. Methanohalophilus portucalensis FDF1T is a halophilic methylotrophic methanogen and has been used as a model to study osmoadaptation in fluctuated abiotic environmental conditions. Here we report a genome-wide phosphoproteomic analysis of M. portucalensis FDF1T to uncover the possible phosphorylation-mediated regulations in methanogenesis and osmoadaptation. According to the biological function analysis revealed 20.6% of total 149 phosphoproteins were involved in the regulation of the single carbon metabolism for methanogenesis and osmoadaptation. Together with genome sequencing and phosphoproteomic analysis, we provided the first evidence to indicate protein phosphorylation networks in regulating the pyrrolysine (Pyl) mediated translation of methylotrophic methyltransferase. Furthermore, the functional assays of glycine sarcosine N-methyltransferase revealed that the enzymatic activity was modulated by threonine phosphorylation. Collectively, our results not only provide new insight into phosphorylation-mediated regulations in single carbon energy metabolism but also clearly demonstrate that phosphorylation plays a critical role in osmolyte betaine synthesis through a phospho-switch mechanism.

Genomic context-dependent histone H3K36 methylation by Drosophila methyltransferases

Project description:How different histone methyltransferases (HMT) cooperate to progressively catalyze different methylation states of the same residue as well the functional consequences of the different methylation states are not well known. Here, we address in the context of Drosophila H3K36 methylation by using single and combinatorial RNAi of HMTs along with genome-wide MNase-ChIPseq analyses. Our study reveals that K36me1/2/3 each mark distinct chromatin compartments. Each HMT predominantly contributes to only one modification state, however our approach also identifies context-dependent cooperative and compensative patterns. Further, correlating changes in K36me1/2/3 to reader binding reveals that K36me2/3 can redundantly contribute to faithful reader binding ensuring robustness.
