Proteomic analysis of phosphorylation in HCoV-229E-infected immortalized human lung epithelial cells (A549)

ABSTRACT: TMT18-labeled proteome-wide profiling of post-translational modifications (phosphorylation) in mock-infected and infected human lung epithelial cells (A549). Human lung epithelial (A549) cells were infected and mock-infected with HCoV-229E. Cells were harvested at 8, 16, and 24 hours post-infection. Extracted proteins were digested with LysC + Trypsin, then TMT18-labeled and enriched for phosphorylation. Samples were split into 6 fractions and run on the Orbitrap Exploris 480. Data was searched with MS-GF+ using PNNL's DMS Processing pipeline.

Host microRNA molecular signatures associated with human H1N1 and H3N2 influenza A viruses

Project description:In this study, we performed a miRNA global profiling in human lung epithelial cells (A549) infected by two different subtypes of human influenza A viruses (H1N1 and H3N2). A549 cells were either mock-infected or infected at a multiplicity of infection (MOI) of 1 with H1N1 or H3N2 viruses, and total RNAs were isolated at 24 hours post-infection (hpi). An MOI of 1 was performed to ensure that 100% of the cells were infected at 24 hpi, a strategy that we have previously validated and used for a transcriptional profiling study of infected cells (Josset et al. , 2010). The purified RNAs were subjected to reverse transcription using a pool of miRNA RT primers (Human pool A v2.1, Applied Biosystems) and subsequently amplified and quantified by RT-qPCR in a TaqMan array MicroRNA card (Applied biosystems).

Project description:PISA assays performed on mock-infected and human coronavirus 229E-infected immortalized human lung fibroblasts (MRC5) cells (BTOL0001590). Samples were harvested at 8, 16, and 24 hours post infection (HCoV-299E). Samples are derived from human whole cell lysates (BTO:0004304). Mock-infected and human coronavirus 229E-infected immortalized human lung fibroblasts (MRC5) cells were collected at 8, 16, and 24 hours post infection (HCoV-299E). Cells were lysed by freeze thaw cycles and each replicate was split in half and treated for 1 minute with proteinase K and then trypsin and the other half just treated with trypsin prior to processing for proteomic analysis. Data was searched with MS-GF+ using PNNL's DMS Processing pipeline.
