Project description:Nitrosomonas Europaea Nm50 culture - concentrated ~600 ml culture to 280 ml mother culture - let acclimate back to 26C for ~1h without substrate - add 1 mM NH4Cl (final concentration) to mother culture bottle: - T0 M1 - 15:30, T1 M1 - 16:30 (working, already nitrite visible), T2 - 18:00 (working, more nitrite clearly), T3 - 19:20 (still increasing, time to split) Split mother culture to 15 subcultures for treatment: LNH - linolenic 8 ul 98% (triplicates, 17 ml culture) LNL - linolenic 2 ul 98% (triplicates, 17 ml culture) LEH - linoleic 8 ul 98% (triplicates, 17 ml culture) LEL - linoleic 2 ul 98% (triplicates, 17 ml culture) C - control, nothing added (just 17 ml, culture) shake up really well after addition: T1 19:40 (500 ul samples for MS) T2 21:30 (500 ul samples for MS) T3 23:00 (500 ul samples for MS) T4 1:30 - 1:50 (500 ul samples for MS) Harvest at T4 by filtration 15 ml from each, freeze @ -80C LC-HRMS measurments according to 10.1007/s00216-025-05818-y

Expression profiling of various colony phenotypes of wild Saccharomyces cerevisiae strain

Project description:Transcriptome comparison of cells from 4 and 7 day-old microcolonies of wild Saccharomyces cerevisiae BR-F strain, 4 and 7 day-old microcolonies of feral BR-RF strain and 4- and 7 day-old microcolonies of domesticated BR-S strain. All colonies grown on solid complex media with glycerol as carbon source. The aim of the study was to identify genes required for fluffy (structured) colony formation as well as the genes specific for certain phenotypic variant. BR-F is wild strain isolated from natural habitat and forms structured colonies when grown on media with non-fermentable carbon source. BR-S strain arose by phenotypic switch from the original wild BR-F strain during the cultivation of BR-F strain under rich and favourable conditions (process of so-called domestication), forms smooth colonies. BR-RF strain is derived from the domesticated BR-S strain under adverse conditions and restores the formation of structured colonies and other properties of original wild BR-F strain.

Analysis of colorectal adenocarcinoma tissue samples by DESI mass spectrometry imaging

Project description:Ex vivo colorectal adenocarcinoma samples were analysed by desorption electrospray ionisation using an Orbitrap mass spectrometer. The samples were analysed in negative mode over the m/z range 600-1000. Some of the samples presented in this dataset were analysed as part of [1]. The study makes available imzML files of profile and centroid mode data, as well as low- and high- resolution optical image files of H&E-stained sections.
