ABSTRACT: The project aims to identify biomarkers of pulmonary dysbiosis (bacterial, fungal, and viral) associated with exacerbations in people with cystic fibrosis (23 patients).
The research plan is organized into four work packages: metagenomics (WP1), metabolomics (WP2), serum inflammatory biomarkers (WP3), and multi-block statistical data integration (WP4).
WP1 involves a longitudinal metagenomic analysis of 120 sputum samples collected from the 23 patients across multiple time points (M0 to M18).
WP2 focuses on plasma metabolomics using Orbitrap LC-HRMS, with standardized sample preparation, addition of isotopically labeled internal standards, and the use of quality control (QC) samples. In total, 136 plasma samples were analyzed, with sampling every three months over more than 18 months of longitudinal follow-up.
