Lipidomic and Metabolomic Changes in Commercial Brewing Strains of Saccharomyces cerevisiae to QSM 2-Phenylethanol

ABSTRACT:

Commercially relevant Saccharomyces cerevisiae strains were cultivated under nitrogen-limiting (SLAD) and nitrogen-sufficient (SHAD) conditions, some supplemented with 200 μM of the aromatic alcohol 2-phenylethanol. After incubation at 24 °C for 72 h, cells were harvested from each condition and analyzed using lipidomic and metabolomic techniques.

Transcript and Proteomic Analyses of Wild-Type and GPA2 Mutant Saccharomyces cerevisiae Strains

Project description:In response to limited nitrogen and abundant carbon sources, diploid Saccharomyces cerevisiae strains undergo a filamentous transition in cell growth as part of pseudohyphal differentiation. Use of the disaccharide maltose as the principal carbon source, in contrast to the preferred nutrient monosaccharide glucose, has been shown to induce a hyper-filamentous growth phenotype in a strain deficient for GPA2 which codes for a Galpha protein component that interacts with the glucose-sensing receptor Gpr1p to regulate filamentous growth. In this report, we compare the global transcript and proteomic profiles of wild-type and Gpa2p deficient diploid yeast strains grown on both rich and nitrogen starved maltose media. We find that deletion of GPA2 results in significantly different transcript and protein profiles when switching from rich to nitrogen starvation media. The results are discussed with a focus on the genes associated with carbon utilization, or regulation thereof, and a model for the contribution of carbon sensing/metabolism-based signal transduction to pseudohyphal differentiation is proposed. Regulation involved in difference in nitrogen requirements of Saccharomyces cerevisiae wine yeasts in alcoholic fermentation

Project description:The objective of this work is to identify, in enological condition, differences in transcriptomic regulation that may explain the differences in strains fermentation properties. For this, we compared the global expression patterns of 8 Saccharomyces cerevisiae strains with different nitrogen requirement (L2868, 4CAR1, Fermiflor, Zymasil, MTF1782, 7013, K1M, EC1118). The transcriptome was analyzed on cells fermenting in a nitrogen limited medium containing 100mg.l-1 assimilable nitrogen (SM100) and harvested at stationary phase (45g/L of CO2 produced and corresponding to 6% ethanol). At this stage the strains exhibited differences in fermentation rates. Moreover, the transcriptome was shown to be rather stable at this stage since growth was stopped for all cells and nutrients have been depleted (Rossignol et al. 2003).

Comparative genomic hybridization of Saccharomyces boulardii and Saccharomyces cerevisiae to determine genomic differences

Project description:A comparative genomic hybridisation experiment using Affymetrix YG-S98 arrays to study the genetic background of S. Boulardii compared to S. Cerevisiae strain BY4743. Background: Saccharomyces boulardii, a yeast that was isolated from fruit in Indochina has been used as a remedy for diarrhoea since 1950, and is now a commercially available treatment throughout Europe, Africa and South America. Though initially classified as a separate species of Saccharomyces, recent publications have shown that the genome of S. boulardii is so similar to Saccharomyces cerevisiae that the two should be classified as conspecific. This raises the question of the distinguishing molecular and phenotypic characteristics present in S. boulardii that make it perform more effectively as a probiotic organism compared to other strains of S. cerevisiae. This investigation reports some of these characteristics including enhanced ability for pseudohyphal switching upon nitrogen limitation and increased resistance to acidic pH. However, these differences did not correlate with increased adherence to epithelial cells or transit through mouse gut. Pertinent characteristics of the S. boulardii genome such as trisomy of chromosome IX, altered copy number of a number of individual genes and sporulation deficiency have been revealed by comparative genome hybridisation using oligonucleotide-based microarrays coupled with a rigorous statistical analysis. The contributions of the different genomic and phenotypic features of S. boulardii to its probiotic nature are discussed.
