Somatic mutations of the PIK3CA gene, which encodes the p110alpha catalytic subunit of phosphatidylinositol 3-kinase (PI3K), are frequent in various cancer types. The majority of mutations cluster at hotspots within exons 9 and 20, which encode the helical and kinase domains of p110alpha. PIK3CA mutations in bile duct and gallbladder carcinomas have not been reported yet. Toxicogenomic effects of nano- and bulk TiO2 particles in the soil nematode Carnorhabditis elegans

Project description:The toxicity and toxicogenomics of selected anatase and rutile nanoparticles (NP) and bulk titanium dioxide (TiO2) particles were evaluated in the soil nematode Caenorhabditis elegans. Results indicated that bulk or nano-TiO2 particles were slightly toxic to soil nematode C. elegans, as measured by reproduction EC50 values ranging from 4 to 32 mg/L. Whole-genome microarray results indicated that the regulation of glutathione-S-transferase gst-3, cytochrome P450 cypp33-c11, stress resistance regulator scl-1, oxidoreductase wah-1, and embryonic development pod-2 genes were significantly affected by nano-sized and bulk TiO2 particles. More specifically, it was determined that anatase particles exerted a greater effect on metabolic pathways, whereas rutile particles had a greater effect on developmental processes. The up-regulation of the pod-2 gene corroborated the phenotypic effect observed in the reproduction test. Our results demonstrated that C. elegans is a good genomic model for nano-TiO2 toxicity assessment.

Project description:Arsenic is one of the most relevant environmental pollutants and human health threats. Several arsenic species occur in soil pore waters. Recently it was discovered that these include inorganic and organic thioarsenates. Dimethylmonothioarsenate (DMMTA) belong to organic thioarsenates and in mammalian cells its toxicity was found to exceed even that of arsenite. We investigated DMMTA toxicity in Arabidopsis thaliana (Col-0) and we found strong transcriptome changes dominated by stress-responsive genes.

The melatonin delivery system based on nano-SiO2 optimizes photosynthetic carbon metabolism and yield formation of soybeans in saline-alkali land

Project description:Nanomaterials show a great potential in regulating plant growth and improving stress resistance due to their unique physical and chemical characteristics. In this study, nano-silicon dioxide (nano-SiO2) was prepared by the sol-gel method and used to construct a highly efficient melatonin delivery system. Soybean cultivars Hefeng 50 and Henong 95 were field-grown in saline-alkali soil and foliar-sprayed with 200 mg/L nano-SiO2, 300 μM melatonin (MT), or their combination at the V3 stage. In the MT-nano-SiO2system, the encapsulation efficiency and drug-loading capacity of melatonin by nano-SiO2were 35% and 53%, respectively. The system can significantly enhanced melatonin photostability. Scanning electron microscopy results showed that the MT-nano-SiO2 composite can be directly adsorbed on the leaf surface. The MT-nano-SiO2 treatment significantly reduced salt-alkali stress induced reactive oxygen species (ROS) accumulation by activated the enzymatic and non-enzymatic antioxidant system expressed as enhancing the activities of SOD, CAT, and APX and regulating glutathione (GSH/GSSG) levels. The results of physiology, RNA-seq and qRT-PCR indicated that MT-nano-SiO2 treatment can repair photosynthetic carbon metabolism damage induced by salt-alkali stress. Specifically, it increased leaf area, photosynthetic pigment content, photosystem II (PSⅡ) efficiency (Fv/Fm, Qy), net photosynthetic rate (Pn), stomatal conductance (Gs), and intercellular CO2concentration (Ci). It also promoted the activities of sucrose phosphate synthase (SPS), sucrose synthase (SuSy), but inhibited the activities of α-amylase, β-amylase to enhance soluble sugar, sucrose, and starch accumulation. Ultimately, MT-nano-SiO2 treatment significantly improved pod number per plant, seed number per plant, 100-seed weight, and yield of soybean plants.

Project description:To deeply investigate the details of the nano-SiO2 effects, we examined the gene expression profile alterations after nano-SiO2 treatment in BMMCs. The difference analysis between the groups showed that 285 genes were significantly expressed after treatment with nano-SiO2. Compared with the blank group, both nano-SiO2 exposure and DNP-HSA stimulation increased the expression of genes related to the MAPK signaling pathway in mast cells to varying degrees.

Revealing nascent RNA processing dynamics with nano-COP

Project description:During maturation, eukaryotic precursor RNAs undergo processing events including intron splicing, 3’-end cleavage, and polyadenylation. Here, we describe nanopore analysis of CO-transcriptional Processing (nano-COP), a method for probing the timing and patterns of RNA processing. An extension of native elongating transcript sequencing (NET-seq), which quantifies transcription genome-wide through short-read sequencing of nascent RNA 3’ ends, nano-COP uses long-read nascent RNA sequencing to observe global patterns of RNA processing. First, nascent RNA is stringently purified through a combination of 4-thiouridine metabolic labeling and cellular fractionation. In contrast to cDNA or short-read–based approaches relying on reverse transcription or amplification, the sample is sequenced directly through nanopores to reveal the native context of nascent RNA. nano-COP identifies both active transcription sites and splice isoforms of single RNA molecules during synthesis, providing insight into patterns of intron removal and the physical coupling between transcription and splicing. The nano-COP protocol yields data within 3 days.
